Resolving the geometric structure of trastuzumab by mobility capillary electrophoresis and native mass spectrometry

Available online Immunoglobulins G(IgGs)are y-shaped globular proteins,however,their high flexibility and heterogeneity pose great challenges to their structure and conformation *** structure of IgG closely correlates to its biofunctions,such as the antibody escape of human immunodeficiency virus(HIV)could attribute to the distance mismatch between the ends of two Fab arms(antigen-binding sites)and envelope glycoprotein spikes on virion ***,we report the first use of mobility capillary electrophoresis(MCE)and native mass spectrometry(nMS)to resolve the internal geometric structure and conformation of an IgG(trastuzumab)in solution *** proteolysis,the ellipsoid dimensions of IgG and its subunits were measured by MCE-nMS *** was then reconstructed,in which the sizes and relative positions of these three subunits in three-dimensional space were *** was found that the two Fab arms have an angle of~102.1°and a distance of~11.0 nm between the two antigen-binding sites under native condition,and the Fc arm was tilted~16.0°towards one of the Fab *** was not on the plane of Fab-Fab,but has an angle of no larger than 103.1°.Under acidic environment(pH 3.0),each subunit of the IgG would unfold into larger dimensions,and the angles between these subunits also *** great potential for tumor imaging and therapy,the structure of F(ab')_(2)fragments was also measured and validated by molecular dynamic *** was found that the electrostatic force among these three subunits and steric hindrance stemming from Fc help maintaining the angle between two Fab arms.

Risk factors,prognostic factors,and nomograms for distant metastasis in patients with diagnosed duodenal cancer:A population-based study

BACKGROUND Duodenal cancer is one of the most common subtypes of small intestinal cancer,and distant metastasis(DM)in this type of cancer still leads to poor *** nomograms have recently been used in tumor areas,no studies have focused on the diagnostic and prognostic evaluation of DM in patients with primary duodenal *** To develop and evaluate nomograms for predicting the risk of DM and person-alized prognosis in patients with duodenal *** Data on duodenal cancer patients diagnosed between 2010 and 2019 were extracted from the Surveillance,Epidemiology,and End Results *** and multivariate logistic regression analyses were used to identify independent risk factors for DM in patients with duodenal cancer,and univariate and multivariate Cox proportional hazards regression analyses were used to determine independent prognostic factors in duodenal cancer patients with *** novel nomograms were established,and the results were evaluated by receiver operating characteristic(ROC)curves,calibration curves,and decision curve analysis(DCA).RESULTS A total of 2603 patients with duodenal cancer were included,of whom 457 cases(17.56%)had DM at the time of *** analysis revealed independent risk factors for DM in duodenal cancer patients,including gender,grade,tumor size,T stage,and N stage(Pyses further identified independent prognostic factors for duodenal cancer patients with DM,including age,histological type,T stage,tumor grade,tumor size,bone metastasis,chemotherapy,and surgery(Py of the nomograms was validated in the training set,validation set,and expanded testing set using ROC curves,calibration curves,and DCA *** results of Kaplan-Meier survival curves(Py predicted the occurrence and prognosis of DM in patients with duodenal *** The two nomograms are expected as effective tools fo

Optical coherence tomography angiography metrics in different stages of diabetic macular edema

Background To investigate the optical coherence tomography angiography(OCTA)characteristics of diabetic macular edema(DME)at different *** This study was a cross-sectional *** diagnosed with DME were *** was classified into early,advanced,and severe *** vessel density(VD)in the superficial vascular plexus(SVP),deep vascular plexus(DVP)and foveal avascular zone(FAZ)parameters,including FAZ area,FAZ perimeter,acircularity index and foveal VD in a 300-μm-wide region around the FAZ(FD-300),were calculated by the AngioVue software.A multivariate generalized estimating equation was used to evaluate the associations between visual acuity and OCTA *** Ninety-two eyes from 74 patients with DME were included in this *** to early(P=0.006)and advanced DME(P=0.003),the acircularity index was higher in severe *** whole and parafoveal VD in the DVP decreased in eyes with severe DME compared to early DME(P=0.018,P=0.005,respectively)and advanced DME(P=0.035,P=0.012,respectively).In the multivariate generalized estimating equation,DME severity,FAZ area and foveal thickness were positively associated with worse visual acuity(P=0.001,P=0.007 and P=0.001,respectively).Conclusion Compared to early and advanced DME,severe DME showed increased irregularity in the FAZ and more extensive vessel damage in the *** severity level of DME,larger FAZ area,and increased foveal thickness could be risk factors for poor visual *** registration The protocol was published in the Chinese Clinical Trial Registry(ChiCTR2000033082).

Effect of fore/aft-loaded rotor on compressor stability under inlet circumferential distortion

A modified small perturbation stability prediction model for axial compressors with circumferential inlet distortions is established and applied to investigate the effect of fore/aft-loaded rotor on compressor stability under circumferentially distorted inlet *** inlet total pressure distribution downstream of the distortion screen is measured in experiments and employed for simulations which are implemented via time-space spectral *** stall inception prediction results via the stability model indicate that the compressor with aft-loaded rotor not only performs better in terms of stability under uniform inlet,but also maintains a larger stability margin under circumferentially distorted *** experiments for compressors with fore-loaded and aft-loaded rotor are respectively carried *** results validate the reliability of numerical simulations and the predicted conclusion that the aft-loaded rotor is beneficial for compressor ***,the ability of the developed theoretical model for compressor stability prediction under circumferential distortions is *** addition,dynamic pressure signals at rotor tip measured in experiments illustrate that the circumferential distortion has little effect on the compressor stall pattern.

Intravitreal conbercept injection with panretinal photocoagulation for high-risk proliferative diabetic retinopathy with vitreous hemorrhage

AIM:To assess the clinical efficacy and safety of combining panretinal photocoagulation(PRP)with intravitreal conbercept(IVC)injections for patients with high-risk proliferative diabetic retinopathy(HR-PDR)complicated by mild or moderate vitreous hemorrhage(VH),with or without diabetic macular edema(DME).METHODS:Patients diagnosed with VH with/without DME secondary to HR-PDR and received PRP combined with IVC injections were recruited in this retrospective *** establishing the patient’s diagnosis,an initial IVC was performed,followed by prompt administration of *** cases who significant bleeding persisted and impeded the laser operation,IVC was sustained before supplementing with *** the completion of PRP,patients were meticulously monitored for a minimum of six *** therapy and IVC injections were judiciously adjusted based on fundus fluorescein angiography(FFA)*** effect and the incidence of adverse events were ***:Out of 42 patients(74 eyes),29 were male and 13 were female,with a mean age of 59.17±12.74y(33-84y).The diabetic history was between 1wk and 26y,and the interval between the onset of visual symptoms and diagnosis of HR-PDR was *** affected eye received 2.59±1.87(1-10)IVC injections and underwent 5.5±1.02(4-8)sessions of *** these,68 eyes received PRP following 1 IVC injection,5 eyes after 2 IVC injections,and 1 eye after 3 IVC *** absorption of VH was observed in all 74 eyes 5-50wk after initial treatment,with resolution of DME in 51 eyes 3-48wk after initial treatment.A newly developed epiretinal membrane was noted in one *** acuity significantly improved in 25 *** complications such as glaucoma,retinal detachment,or endophthalmitis were ***:The study suggests that the combination of PRP with IVC injections is an effective and safe modality for treating diabetic VH in patients with HR-PDR.

Jasmonates(JAs)are essential hormones that regulate plant responses to biotic and abiotic stresses and are involved in various developmental processes,including flowering time control,floral organ development,and fertility of male and female gametes(Howe et al.,2018).However,our understanding of the mechanisms by which JAs integrate multiple signals to regulate diverse physiological processes remains *** the canonical JA signaling pathway,activated JAs are perceived by the F-box protein receptor CORONATINE INSENSITIVE 1(COl1),which induces the interaction between COl1 and the Jas domain of the repressor proteins JASMONATE-ZIM DOMAIN PROTEINs(JAZs),leading to the ubiquitination and degradation of JAZ *** proteolytic destruction of the JAZ repressors is a crucial step that releases the inhibition of JAZs on the JA response transcription factor MyC2,allowing MyC2 to activate the expression of downstream genes(Howe et al.,2018).Therefore,the stability and specific spatiotemporal expression pattern of the JAZ proteins are critical for the regulation of JA signaling output,as they determine the activation and repression of JA responses.
