Neuropeptide y receptor y8b(npy8br)regulates feeding and digestion in Japanese medaka(Oryzias latipes)larvae:evidence from gene knockout

Neuropeptide y receptor y8(NPy8R)is a fish-specific receptor with two subtypes,NPy8AR and *** in expression levels during physiological processes or in vivo regulation after ventricular injection suggest that NPy8BR plays an important role in feeding regulation;this has been found in only a few fish,at *** order to better understand the physiological function of npy8br,especially in digestion,we used clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)technology to generate npy8br-/-japanese medaka(Oryzias latipes).We found that the deletion of npy8br in medaka larvae affected their feeding and digestion ability,ultimately affecting their ***,npy8br deficiency in medaka larvae resulted in decreased feed intake and decreased expression levels of orexigenic genes(npy and agrp).npy8br-/-medaka larvae fed for 10 d(10th day of feeding)still had incompletely digested brine shrimp(Artemia nauplii)in the digestive tract 8 h after feeding,the messenger RNA(mRNA)expression levels of digestion-related genes(amy,lpl,ctra,and ctrb)were significantly decreased,and the activity of amylase,trypsin,and lipase also significantly *** deletion of npy8br in medaka larvae inhibited the growth and significantly decreased the expression of growth-related genes(gh and igf1).Hematoxylin and eosin(H&E)sections of intestinal tissue showed that npy8br-/-medaka larvae had damaged intestine,thinned intestinal wall,and shortened intestinal *** far,this is the first npy8br gene knockout model established in fish and the first demonstration that npy8br plays an important role in digestion.

Effects of the urban landscape on heatwave-mortality associations in Hong Kong: comparison of different heatwave definitions

Despite increased attention given to potential modifiers of temperature-mortality associations, evidence for variations between different urban landscape characteristics remains limited. It is in this context that in this paper effect modifications of multiple urban landscape characteristics are explored under different heatwave definitions for different age groups and gender in Hong Kong, China. Daily meteorological data and heatwave-related mortality counts from 2008 to 2017 were collected from the Hong Kong Census and Statistics Department, China. A case-only design was adopted, combined with logistic regression models to examine the modification effects of five urban landscape characteristics under six heatwave definitions. Stratified analyses were conducted to investigate age- and gender-specific effect modifications. It is found that individuals living in greener areas experienced lower levels of mortality during or immediately after heatwaves. In contrast, a higher building density and nighttime land surface temperature (LST) were associated with a higher heatwave-related mortality risk. Pronounced effect modifications of these urban landscape characteristics were observed under hotter and longer heatwaves, and in older adults (age ≥ 65 years) and males. The findings provide a scientific basis for policymakers and practitioners when considering measures for coping with hotter, longer, and more frequent heatwaves in the context of global climate change.

Exploring the impacts of major events on the systemic risk of the international energy market

This study examines the systemic risk caused by major events in the international energy market(IEM)and proposes a management strategy to mitigate it. Using the tail-event driven network(TENET)method, this study constructed a tail-risk spillover network(TRSN) of IEM and simulated the dynamic spillover tail-risk process through the cascading failure mechanism. The study found that renewable energy markets contributed more to systemic risk during the Paris Agreement and the COVID-19pandemic, while fossil energy markets played a larger role during the Russia-Ukraine conflict. This study identifies systemically important markets(SM) and critical tail-risk spillover paths as potential sources of systemic risk. The research confirms that cutting off the IEM risk spillover path can greatly reduce systemic risk and the influence of SM. This study offers insights into the management of systemic risk in IEM and provides policy recommendations to reduce the impact of shock events.

The Triassic granitoids in Central Tianshan play a key role in determining the petrogenesis and tectonic evolution on the southern margin of the Central Asian orogenic *** this study,we present SHRIMP zircon U-Pb ages,Hf isotopic and geochemical data on the Xingxingxia biotite granite,amazonite granite and granitic pegmatite in Central Tianshan,NW *** U-Pb dating yielded formation ages of 242 Ma for the biotite granite and 240 Ma for the amazonite *** granitoid rocks have high K_(2)O with low MgO and CaO *** are enriched in Nb,Ta,Hf and y,while being depleted in Ba and Sr,showing flat HREE patterns and negative Eu *** have typical A-type granite geochemical signatures with high Ga/A_(1)(8–13)and TFeO/(TFeO+MgO)ratios,showing an A_(2) affinity for biotite granite and an A_(1) affinity for amazonite granite and granitic *** ε_(Hf)(t)values of the granitoids are 0.45–2.66,with Hf model ages of 0.99–1.17 *** suggests that these A-type granites originated from partial melting of the lower *** propose that Xingxingxia Triassic A-type granites formed under lithospheric extension from post-orogenic to anorogenic intraplate settings and NE-trending regional strike-slip fault-controlled magma emplacement in the upper crust.

Detecting changes in surface air temperature in mid-and low-altitude mountainous regions is essential for a comprehensive understanding of warming trend with *** use daily surface air temperature data from 64 meteorological stations in Wuyi Mountains and its adjacent regions to analyze the spatio-temporal patterns of temperature *** results show that Wuyi Mountains have experienced significant warming from 1961 to *** warming trend of the mean temperature is 0.20℃/decade,the maximum temperature is 0.17℃/decade,and the minimum temperature is 0.26℃/*** 1961-1990,more than 63%of the stations showed a decreasing trend in annual mean temperature,mainly because the maximum temperature decreased during this ***,in 1971-2000,1981-2010 and 1991-2018,the maximum,minimum and mean temperatures *** fastest increasing trend of mean temperature occurred in the southeastern coastal plains,the quickest increasing trend of maximum temperature occurred in the northwestern mountainous region,and the increase of minimum temperature occurred faster in the southeastern coastal and northwestern mountainous regions than that in the central ***,this study suggests that elevation does not affect warming in the Wuyi *** results are beneficial for understanding climate change in humid subtropical middle and low mountains.

Combined UAMP and MF Message Passing Algorithm for Multi-Target Wideband DOA Estimation with Dirichlet Process Prior

When estimating the direction of arrival (DOA) of wideband signals from multiple sources, the performance of sparse Bayesian methods is influenced by the frequency bands occupied by signals in different directions. This is particularly true when multiple signal frequency bands overlap. Message passing algorithms (MPA) with Dirichlet process (DP) prior can be employed in a sparse Bayesian learning (SBL) framework with high precision. However, existing methods suffer from either high complexity or low precision. To address this, we propose a low-complexity DOA estimation algorithm based on a factor graph. This approach introduces two strong constraints via a stretching transformation of the factor graph. The first constraint separates the observation from the DP prior, enabling the application of the unitary approximate message passing (UAMP) algorithm for simplified inference and mitigation of divergence issues. The second constraint compensates for the deviation in estimation angle caused by the grid mismatch problem. Compared to state-of-the-art algorithms, our proposed method offers higher estimation accuracy and lower complexity.

Biomimetic soft robotic wrist with 3-DOF motion and stiffness tunability based on ring-reinforced pneumatic actuators and a particle jamming joint

The human wrist, a complex articulation of skeletal muscles and two-carpal rows, substantially contributes to improvements in maneuverability by agilely performing three-degree-of-freedom(3-DOF) orienting tasks and regulating stiffness according to variations in interaction forces. However, few soft robotic wrists simultaneously demonstrate dexterous 3-DOF motion and variable stiffness;in addition, they do not fully consider a soft-rigid hybrid structure of integrated muscles and two carpal *** this study, we developed a soft-rigid hybrid structure to design a biomimetic soft robotic wrist(BSRW) that is capable of rotating in the x and y directions, twisting around the z-axis, and possessing stiffness-tunable capacity. To actuate the BSRW, a lightweight soft-ring-reinforced bellows-type pneumatic actuator(SRBPA) with large axial, linear deformation(η_(lcmax)=70.6%,η_(lemax)=54.3%) and small radial expansion(η_(demax)=3.7%) is designed to mimic the motion of skeletal muscles. To represent the function of two-carpal rows, a compact particle-jamming joint(PJJ) that combines particles with a membrane-covered ballsocket mechanism is developed to achieve various 3-DOF motions and high axial load-carrying capacity(>60 N). By varying the jamming pressure, the stiffness of the PJJ can be adjusted. Finally, a centrally positioned PJJ and six independently actuated SRBPAs, which are in an inclined and antagonistic arrangement, are sandwiched between two rigid plates to form a flexible,stable, and compact BSRW. Such a structure enables the BSRW to have a dexterous 3-DOF motion, high load-carrying ability,and stiffness tunability. Experimental analysis verify 3-DOF motion of BSRW, producing force of 29.6 N and 36 N and torque of2.2 Nm in corresponding rotations. Moreover, the range of rotational angle and stiffness-tuning properties of BSRW are studied by applying jamming pressure to the PJJ.
