A biosynthetic gene cluster for the bioactive fungal sesterterpenoids variecolin(1)and variecolactone(2)was identified in Aspergillus aculeatus ATCC *** production of 1 and 2 was achieved in Aspergillus oryzae by expressing the sesterterpene synthase VrcA and the cytochrome P450 ***,the replacement of VrcB with homologous P450s from other fungal terpenoid pathways yielded three new variecolin analogues(5-7).Analysis of the compounds’anticancer activity in vitro and in vivo revealed that although 5 and 1 had comparable activities,5 was associated with significantly reduced toxic side effects in cancer-bearing mice,indicating its potentially broader therapeutic *** study describes the first tests of variecolin and its analogues in animals and demonstrates the utility of synthetic biology for creating molecules with improved biological activities.

Congenital central hypoventilation syndrome （CCHS, OMIM #209880） is a rare autosomal dominant disorder of the autonomic nervous system （ANS） characterized by an abnormal autonomic ventilatory response to progressive hypercarbia and sustained hypoxemia. Patients typically present in the newborn period with hypoventilation or apnea asleep, awake, or both, without any associated cardiac, pulmonary, neuromuscular or brainstem lesions. Rarely, some patients may present at a later age and are diagnosed to have late onset central hypoventilation syndrome （loCHS）.1 Other features of ANS dysfunction such as feeding difficulty due to oesophageal dysmotility, severe constipation in the absence of Hirschsprung disease, poor regulation of basal body temperature, episodes of profuse sweating, pupillary and ocular abnormalities, decreased beat-to-beat variability of heart rate, attenuated response of heart rate to exercise, abnormal fluctuation of blood pressure, decreased perception to pain, and decreased perception to anxiety may be variably present but not essential for diagnosis Furthermore, this central hypoventilation can occur as an isolated feature or in association with a number of neurocristopathies, notably Hirschsprung disease （Haddad syndrome, OMIM #209880） and tumours of the sympathetic nervous system particularly neuroblastoma, ganglioneuro- blastoma, and ganglioneuroma, which were found in 20% and 5%--10% of CCHS patients, respectively.2 Studies of genes pertinent to the early embryologic development of the neural crest cells, specifically the endothelin and the RET-GDNF signaling pathways, have recently led to the identification of PHOX2B as the major disease causing gene for CCHS.2-6 PHOX2B was mapped to chromosome 4p12 and consists of 3 exons. It encodes a highly conserved paired-like homeobox transcription factor of 314 amino acids linked to the RET-GDNF signaling

Conventional hepatectomy is an effective way to treat hepatocellular ***,it is invasive and *** use of laparoscopy in hepatectomy,while technically demanding,reduces surgical invasiveness and stressfulness but still achieves complete resection with adequate *** with conventional hepatectomy,laparoscopic hepatectomy provides a better chance and situation for further surgery in the case of recurrence of hepatocellular *** aged patients can successfully endure repeated hepatectomy using laparoscopy,as shown in the present *** report presents a case of repeated laparoscopic hepatectomy treating hepatocellular carcinoma and its recurrence in an aged patient having cirrhosis,a disease causing extra difficulty for performing laparoscopic *** report also describes techniques of the operation and displays characteristic results of laparoscopic hepatectomy such as smaller wounds,less blood loss,less pain,less scars and adhesion,shorter postoperative hospital stay,and faster recovery.

Drug delivery to the posterior segment of the eye, especially the retina, faces the unique challenges of various anatomical and physiological barriers. Invasive intravitreal injection is currently the mainstream clinical approach to posterior eye diseases such as age-related macular degeneration, diabetic retinopathy, uveitis, and cytomegalovirus retinitis. Repeated injections could be complicated by endophthalmitis, intra- ocular pressure elevation, retinal vascular occlusion, and ret- inal detachment, let alone the psychological and economical burdens suffered by patients （Schwartz et al., 2014）. In view of this, several extended release drug delivery systems have been devised and the idea of encapsulated-cell technology （ECT） has gained popularity in recent decades since its first proposal in the 1960s.

Analyzing Industrial Structure Evolution of Old Industrial Cities Using Evolutionary Resilience Theory：A Case Study in Sheny.ng of China

The recession and revitalization of old industrial cities concerns urban industrial evolution and its characteristics. Based on the theory of evolutionary resilience, we developed an analytical framework for the industrial structure evolution of old industrial cities, and applied the framework to a case study in Sheny.ng. The following conclusions are drawn. First, since 1978, Sheny.ng's industrial growth capacity has shown fluctuation between ‘contraction-expansion'. As the secondary industry has a much stronger expansionary and contractionary capacity for growth, this results in lacking stability leading to industrial structure transformation. Second, since 1999, the orientation towards a high-end manufacturing industry in Sheny.ng has weakened, and the evolution of the new and old growth path is characterized by low-end orientation. Third, since 2007, Sheny.ng's industrial innovation output capacity has dropped sharply which has been significantly affected by scientific and technological personnel and enterprise-owed science and technology institutions and to a less extent by R&D expenditure. We applied the resilience theory to study the industrial evolution of an old industrial city, explored new study perspectives on industrial evolution and verified the applicability of the resilience theory. This paper provides a scientific reference for understanding the recent deceleration in economic growth in the Northeast old industrial base, and for exploring new paths toward revitalization.

Microbial symbioses have had profound impacts on the evolution of ***,changes in host biology may impact the evolutionary trajectory of symbionts *** cuenoti is present in almost all cockroach species and enables hosts to subsist on a nutrient-poor *** investigate if host biology has impacted Blattabacterium at the genomic level,we sequenced and analyzed 25 genomes from Australian soil-burrowing cockroaches(Blaberidae:Panesthiinae),which have undergone at least seven separate subterranean,subsocial transitions from above-ground,wood-feeding *** find at least three independent instances of genome erosion have occurred in Blattabacterium strains exclusive to Australian soil-burrowing *** shrinkages have involved the repeated inactivation of genes involved in amino acid biosynthesis and nitrogen recycling,the core role of Blattabacterium in the host-symbiont *** most drastic of these erosions have occurred in hosts thought to have transitioned underground the earliest relative to other lineages,further suggestive of a link between gene loss in Blattabacterium and the burrowing behavior of *** Blattabacterium is unable to fulfill its core function in certain host lineages,these findings suggest soil-burrowing cockroaches must acquire these nutrients from novel *** study represents one of the first cases,to our knowledge,of parallel host adaptations leading to concomitant parallelism in their mutualistic symbionts,further underscoring the intimate relationship between these two partners.

Primary and metastatic liver cancer is an increasingly common and difficult to control disease *** offers a non-invasive treatment alternative for these patients who often have few options and a poor ***,the anatomy and aggressiveness of liver cancer poses significant challenges such as accurate localization at simulation and treatment,management of motion and appropriate selection of dose *** article aims to review the options available and provide information for the practical implementation and/or improvement of liver cancer radiation programs within the context of stereotactic body radiotherapy and image-guided radiotherapy *** patient inclusion and exclusion criteria are presented given the significant toxicity found in certain sub-populations treated with ***,certain sub-populations,such as those with tumor thrombosis or those with larger lesions treated with transarterial chemoembolization,have been shown to have significant improvements in outcome with the addition of radiation and merit special *** a liver radiation programrequires three primary challenges to be addressed:(1) immobilization and motion management;(2) localization;and(3) dose regimen and constraint *** to deal with motion include simple internal target volume(ITV) expansions,non-gated ITV reduction strategies,breath hold methods,and surrogate marker methods to enable gating or *** of the tumor and organs-at-risk are addressed using contrast infusion techniques to take advantage of different normal liver and cancer vascular anatomy,imaging modalities,and margin ***,a dose response has been demonstrated and dose regimens appear to be converging.A more uniform approach to treatment in terms of technique,dose selection and patient selection will allow us to study liver radiation in larger and,hopefully,multicenter randomized studies.
