Endoplasmic reticulum stress and autophagy in cerebral ischemia/reperfusion injury:PERK as a potential target for intervention

Several studies have shown that activation of unfolded protein response and endoplasmic reticulum(ER)stress plays a crucial role in severe cerebral ischemia/reperfusion *** occurs within hours after cerebral ischemia,but the relationship between ER stress and autophagy remains *** this study,we established experimental models using oxygen-glucose deprivation/reoxygenation in PC12 cells and primary neurons to simulate cerebral ischemia/reperfusion *** found that prolongation of oxygen-glucose deprivation activated the ER stress pathway protein kinase-like endoplasmic reticulum kinase(PERK)/eukaryotic translation initiation factor 2 subunit alpha(e IF2α)-activating transcription factor 4(ATF4)-C/EBP homologous protein(CHOP),increased neuronal apoptosis,and induced ***,inhibition of ER stress using inhibitors or by si RNA knockdown of the PERK gene significantly attenuated excessive autophagy and neuronal apoptosis,indicating an interaction between autophagy and ER stress and suggesting PERK as an essential target for regulating *** autophagy with chloroquine exacerbated ER stress-induced apoptosis,indicating that normal levels of autophagy play a protective role in neuronal injury following cerebral ischemia/reperfusion *** from this study indicate that cerebral ischemia/reperfusion injury can trigger neuronal ER stress and promote autophagy,and suggest that PERK is a possible target for inhibiting excessive autophagy in cerebral ischemia/reperfusion injury.

对黄河流域中熟杂交棉花区域试验参试品种进行分析和评价,可以为品种合理布局和品种性状改良提供科学依据。本研究利用GYT双标图对2022—2023年黄河流域30个参试品种的产量、品质、农艺性状、抗病性进行综合评价,深入分析皮棉产量与铃重、单株铃数、霜前花率、衣分、第一果枝节位、果枝数、生育期、株高、子指、纤维长度、断裂比强度、马克隆值、伸长率、整齐度、枯萎病指数、黄萎病指数等性状的组合水平,联合方差分析结果表明, 2年试验中所有性状的基因型效应和环境效应均达到了极显著水平,大部分基因型与环境互作效应达到了显著或极显著水平,同时大部分互作效应平方和占比总变异平方和比例大于基因型效应。筛选出中棉所9B07等产量-性状组合优良的品种,比对照品种中棉所9711适应性更广、丰产性更好,具有较高的应用推广价值。GYT双标图比GT双标图具有解释的变异比例更高、拟合度更好、分析结果可信度更高等优点,可以更加直观展示参试品种特点,为我国作物品种多性状综合评价提供参考。

Risk factors for chronic ankle instability after first episode of lateral ankle sprain:A retrospective analysis of 362 cases

Background:Chronic ankle instability(CAI) is a common sequela following an acute lateral ankle sprain(LAS).To treat an acture LAS more effectively and efficiently,it is important to identify patients at substantial risk for developing *** study identifies magnetic resonance imaging(MRI) manifestations for predicting CAI development after a first episode of LAS and explores appropriate clinical indications for ordering MRI scans for these ***:All patients with a first-episode LAS who received plain radiograph and MRI scanning within the first 2 weeks after LAS from December 1,2017 to December 1,2019 were *** were collected using the Cumberland Ankle Instability Tool at final *** and other related clinical variables,including age,sex,body mass index,and treatment were also *** and multivariable analyses were performed successively to identify risk factors for CAI after first-episode ***:A total 131 out of 362 patients with a mean follow-up of 3.0± 0.6 years(mean ± SD;2.0—4.1 years) developed CAI after first-episode *** to multivariable regression,development of CAI after first-episode LAS was associated with 5 prognostic factors:age(odds ratio(OR)=0.96,95% confidence interval(95%CI):0.93-1.00,p=0.032);body mass index(OR=1.09,95%CI:1.02-1.17,p=0.009);posterior talofibular ligament injury(OR=2.17,95%CI:1.05-4.48,p=0.035);large bone marrow lesion of the talus(OR=2.69,95%CI:1.30-5.58,p=0.008),and Grade 2 effusion of the tibiotalar joint(OR=2.61,95%CI:1.39-4.89,p=0.003).When patients had at least 1 positive clinical finding in the 10-m walk test,anterior drawer test,or inversion tilt test,they had a 90.2% sensitivity and 77.4% specificity in terms of detecting at least 1 prognostic factor by ***:MRI scanning is valuable in predicting CAI after first-episode LAS for those patients with at least 1 positive clinical finding in the10-m walk test,anterior drawer test,and inversion tilt

Anti-breast cancer effect of quercetin nanoparticles based on MOFs for chemo-phototherapy and heat shock proteins inhibition

Phototherapy, including photodynamic therapy(PDT) and photothermal therapy (PTT), is a promisingmethod of tumor treatment, which has the advantages ofstrong specificity and small side effects. However, PDT willaggravate the hypoxic environment in tumor cells, and PTTwill lead to heat-resistant reaction, both of which will make theorganism quickly produce heat shock proteins (HSPs) toprotect itself, thus seriously hindering the therapeuticefficiency of phototherapy. As a natural HSPs inhibitor,quercetin (QR) provides a feasible way to solve this ***, a kind of metal organic framework (MOF), Fe-TCPP(FT), was used as a delivery material and photosensitizer tobuild a chemo-phototherapy platform, and QR was used as achemotherapy drug and HSPs inhibitor. On this basis, the nanoparticles (NPs) were modified with PEG-CPP30 (a cellpenetratingpeptide that specifically recognizes human breast cancer MCF-7 cells), which showed excellent targetingcapacity and biocompatibility. The combination of chemotherapy and phototherapy significantly improved the anti-cancereffect. At the same time, NPs also showed prominent anti-HSPs effect. The effective connection of the two played acomplementary role in the process of tumor treatment, which provided a new strategy for the accurate and efficienttreatment of tumors in the future.

Electrocatalytic water splitting offers a promising way for hydrogen production with near-zero ***,such as molybdenum carbides(Mo_(2)C),are promising materials for hydrogen evolution reaction(HER)but still suffer from poor intrinsic water activation ***,we developed a plasmon-induced local electric field(PILEF)strategy to solve this ***(Ag)nanoparticles decorated Mo_(2)C nanosheets(Ag/Mo_(2)C)were successfully prepared by electrostatic *** visible light excited the PILEF on Ag/Mo_(2)C remarkably reducing the activation energy by 92.7 kJ·mol^(-1)from 147.3 kJ·mol^(-1)of Mo_(2)C to 54.6 kJ·mol^(-1).As a result,the plasmonic Ag/Mo_(2)C significantly enhances~2.3-fold of the current density from 2.8 mA·cm^(-2)of Mo_(2)C to 6.5 mA·cm^(-2)at-3 V *** and reduces the overpotential by 104 mV from 403 mV of dark state to 299 mV of light state at the current density of 10 mA·cm^(-2),achieving better performance than reported *** research demonstrates that PILEF enhances HER activities,offering a potential strategy for boosting the intrinsic activities of catalysts.

Cells are the fundamental units of biological systems and exhibit unique development trajectories and molecular features. Our exploration of how the genomes orchestrate the formation and maintenance of each cell, and control the cellular phenotypes of various organismsis, is both captivating and intricate. Since the inception of the first single-cell RNA technology, technologies related to single-cell sequencing have experienced rapid advancements in recent years. These technologies have expanded horizontally to include single-cell genome, epigenome,proteome, and metabolome, while vertically, they have progressed to integrate multiple omics data and incorporate additional information such as spatial sc RNA-seq and CRISPR screening. Single-cell omics represent a groundbreaking advancement in the biomedical field,offering profound insights into the understanding of complex diseases, including cancers. Here, we comprehensively summarize recent advances in single-cell omics technologies, with a specific focus on the methodology section. This overview aims to guide researchers in selecting appropriate methods for single-cell sequencing and related data analysis.

Breast cancer bone metastasis poses significant challenge for therapeutic *** the metastatic environment,osteoclasts and tumor cells interact synergistically to promote cancer *** this study,the proprotein convertase furin is targeted due to its critical roles in both tumor cell invasion and osteoclast ***,the furin inhibitor is specifically delivered by bone targeting superparamagnetic iron oxide(SPIO)*** in vitro and in vivo data demonstrate that this system can effectively inhibit both osteoclastic bone resorption and breast cancer invastion,leading to alleviated ***,the bone targeting&furin inhibition nanoparticle system is a promising therapeutic and diagnostic strategy for breast cancer bone metastasis.
