Arteriolosclerosis differs from venular collagenosis in relation to cerebrovascular parenchymal damages:an autopsy-based study

Recombinant Mouse Canstatin Inhibits Chicken Embryo Chorioallantoic Membrane Angiogenesis and Endothelial Cell Proliferation

Human canstatin, a 24 kD fragment of the α2 chain of type IV collagen, has been proved to be one of the most effective inhibitors of angiogenesis and tumor growth. To investigatein vivoantiangiogenesis activity andin vitroeffects on endothelial cell proliferation of recombinant mouse canstatin, the cDNA of mouse canstatin was introduced into an expression vector pQE40 to construct a prokaryotic expression vector pQE-mCan. The recombinant mouse canstatin efficiently expressed inE. coliM15 after IPTG induction was monitored by SDS-PAGE and by Western blotting with an anti-hexahistidine tag antibody. The expressed mouse canstatin, mainly as inclusion bodies, accounted for approximately 35% of the total bacterial proteins. The inclusion bodies were washed, lysed and purified by the nickel affinity chromatography to a purity of approximately 93%. The refolded mouse canstatin was tested on the chicken embryo chorioallantoic membranes (CAM), and a large number of newly formed blood vessels were significantly regressed. In addition, recombinant mouse canstatin potently inhibited endothelial cell proliferation with no inhibition on non-endothelial cells. Taken together, these findings demonstrate that the recombinant mouse canstatin effectively inhibited angiogenesis of the chicken embryo in a dose-dependent manner and specially suppressed in vitro the proliferation of human umbilical vein endothelial cells.

The action potential of pacemaker cells in the sino-atrial node forms the automatic rhythm of the heart. The automatic depolarization in phase 4 is me DaSlS of the automaticity in pacemaker cells. Many currents are included in phase 4, such as calcium current, TTX-sensitive sodium current, sustained inward current （Isi）, decay of delayed rectifier potassium current, etc. Funny current （If） has long been recognized as important in this phase and is activated at hyperpolarized potentials during cell diastole and in turn activates other currents to form automatic depolarization.

The current animal models of stroke primarily model a single intracerebral hemorrhage(ICH)attack,and there is a lack of a reliable model of recurrent *** this study,we established 16-month-old C57 B L/6 male mouse models of ICH by injecting collagenaseⅦ-S into the left ***-one days later,we injected collagenaseⅦ-S into the right striatum to simulate recurrent *** results showed that mice subjected to bilateral striatal hemorrhage had poorer neurological function at the early stage of hemorrhage,delayed recovery in locomotor function,motor coordination,and movement speed,and more obvious emotional and cognitive dysfunction than mice subjected to unilate ral striatal *** findings indicate that mouse models of bilateral striatal hemorrhage can well simulate clinically common recurrent *** models should be used as a novel tool for investigating the pathogenesis and treatment targets of recurrent ICH.

基于群组分析的组织信任在护士信任患者与工作投入间的中介效应

目的基于多群组分析,探讨不同级别医院间组织信任在护士信任患者与工作投入之间的中介效应。方法采取便利抽样法,于2022年6月-10月选取福建省三明市共35所三级、二级、一级医院的1 141名临床护士作为研究对象。采用一般资料问卷、工作投入量表、护士信任患者量表、组织信任量表进行调查。采用SPSS 26.0分析护士信任患者、组织信任、工作投入的相关性,采用AMOS 26.0构建结构方程模型,分析组织信任在护士信任患者与工作投入间的中介效应,采用多群组分析不同级别医院间模型的差异。结果本研究护士总体的工作投入、护士信任患者、组织信任总分分别为41.00(27.00,48.00)分、48.00(38.00,53.00)分、52.00(52.00,64.00)分。组织信任在护士信任患者与工作投入间存在部分中介作用(β=0.318,P<0.001),中介效应占总效应的56.28%。多群组分析结果显示,二级医院的护士信任患者对组织信任的路径影响(B=0.771,P<0.001)及护士信任患者对工作投入(B=0.289,P<0.001)的路径影响作用最高,三级医院的组织信任对护士工作投入的路径影响作用最高(B=0.454,P<0.001)。结论护士信任患者、组织信任及工作投入总分均处于中等偏上水平,护士对患者的信任可以通过组织信任间接影响工作投入。不同级别医院间护士信任患者、组织信任及工作投入的路径关系存在差异。建议不同级别医院的护理管理者应针对性地制订不同的措施,塑造积极的工作氛围,以提升护理团队的工作投入水平。

目的探讨青春期起病的迟发型甲基丙二酸血症患儿的临床特点、诊断治疗及随访。方法回顾性病例分析。选择2002年1月至2023年6月于北京大学第一医院确诊的青春期起病的55例甲基丙二酸血症患儿为研究对象,收集患儿症状、体征、检验结果、基因变异等临床资料,每3或6个月通过微信、电话或门诊进行随访(截至2023年12月),对患儿的临床表型、基因型及转归进行总结。结果55例患儿中男31例、女24例,于10~18岁发病,中位发病年龄为12岁,第二性征发育均正常,就诊时Tanner 2~5期。9例(16%)在感染后起病,5例(9%)在剧烈运动后起病,从发病到确诊的时间历经2个月至6年。45例(82%)主要表现为神经精神疾病,12例(22%)出现心血管损伤,7例(13%)出现肾脏损伤,12例(22%)合并眼病。54例(98%)患甲基丙二酸血症合并同型半胱氨酸血症,1例患单纯型甲基丙二酸血症(2%)。54例获得了基因诊断,发现20种MMACHC基因变异和2种MMUT基因变异。53例MMACHC基因变异患儿中,1例为PRDX1和MMACHC双基因变异,共105个等位基因变异,前5位高频度变异为39个(37%)c.482G>A、17个(16%)c.609G>A、11个(10%)c.658_660delAAG、10个(10%)c.80A>G、4个(4%)c.567dupT和4个(4%)c.394C>T。所有患儿经过钴胺素、左卡尼汀、甜菜碱和对症治疗后逐渐康复,54例(98%)恢复正常就学和工作。结论青春期起病的甲基丙二酸血症患儿多为合并型,发病隐匿,部分因疲劳或感染诱发疾病,临床识别困难,容易被延误诊断。生化代谢和基因检测是获得正确诊断的关键,通过钴胺素、左卡尼汀和甜菜碱治疗可以显著改善病情。

Overpressure origins and evolution in deep-buried strata:A case study of the Jurassic Formation,central Junggar Basin,western China

本研究以联吡啶为骨架合成了一种双齿膦配体BiPyPhos,通过核磁共振波谱、高分辨质谱和红外分析证实了双膦配体的结构。该配体的设计充分考虑了联吡啶骨架中N原子对骨架电子效应的调节作用,以及对铑配合物结构的调控能力。进一步的催化性能测试表明,该配体与铑原位形成的均相催化剂在催化端烯烃和内烯烃等底物时表现出优异的催化活性、化学选择性和区域选择性。例如,在1-辛烯氢甲酰化反应中,底物转化率高达100.0%,成醛选择性达到88.6%,其正异比高达43。该配体具有柔变的骨架和大的自然咬角,有利于形成空间位阻较小的直链烷基-铑活性物种,从而实现对区域选择性的调控。配体BiPyPhos的成功应用为羰基化反应催化剂的设计与合成提供了新的思路。

Hyperpolarization activated cation current (If) in cardiac myocytes from pulmonary vein sleeves in the canine with atrial fibrillation

Objective To investigate the characteristics of ectopic automaticity and cation current (If) of cardiac myocytes from pulmonary vein sleeves (PVs) in canines with atrial fibrillation. Methods The canines (8–10 years old) were subjected to long-term, rapid atrial pacing (RAP) for 10 weeks, which induced the atrial fibrillation model. Disassociation of PVs of canines yielded single cardiac myocytes from a Landengorff column. Action potential, If and hyperpolarisation activated cyclic nucleotide-gated (HCN) currents were measured with the patch-clamp technique. Results Compared with the control group, cardiac myocytes from the RAP canine PVs had spontaneous diastolic depolarization, shorter action potential duration, and larger If densities. In the group of RAP cells, the half maximal activation potential (V1/2) was found to be less negative (��105.5 ±5.2 mV) compared to control cells (��87.3 ±4.9 mV). Current densities of If were increased significantly by β-adrenergic receptor stimulation with isoproterenol and caused an acceleration of current activation. In contrast, If currents in the RAP were reduced by carvedilol, a selective beta-adrenergic receptor. Another important finding is that HCN4-based channels may make a significant contribution to If in PVs cells, but not HCN2. Meanwhile, HCN4 current significantly increases in canine PVs cardiac myocytes with RAP. Conclusions The spontaneous action potential and larger If currentwere observed in the PVs cardiac myocytes using RAP, whichmay contribute tomore ectopic activity events to trigger and maintain atrial fibrillation.
