A Biomimetic Surface for Infection-resistance through Assembly of Metal-phenolic Networks

Despite the fact that numerous infection-resistant surfaces have been developed to prevent bacterial colonization and biofilm formation, developing a stable, highly antibacterial and easily produced surface remains a technical challenge. As a crucial structural component of biofilm, extracellular DNA（eDNA） can facilitate initial bacterial adhesion, subsequent development, and final maturation. Inspired by the mechanistic pathways of natural enzymes（deoxyribonuclease）, here we report a novel antibacterial surface by employing cerium（Ce（Ⅳ）） ion to mimic theDNA-cleavage ability of natural enzymes. In this process, the coordination chemistry of plant polyphenols and metal ions was exploited to create an in situ metal-phenolic film on substrate surfaces. Tannic acid（TA） works as an essential scaffold and Ce（Ⅳ） ion acts as both a cross-linker and a destructor of eDNA. The Ce（Ⅳ）-TA modified surface exhibited highly enhanced bacteria repellency and biofilm inhibition when compared with those of pristine or Fe（Ⅲ）-TA modified samples. Moreover, the easily produced coatings showed high stability under physiological conditions and had nontoxicity to cells for prolonged periods of time. This as-prepared DNA-cleavage surface presents versatile and promising performances to combat biomaterial-associated infections.

Deregulated telomere length is a causative factor in many physiological and pathological processes,including aging and *** studies focusing on telomeres have revealed important roles for cooperation between the Shelterin protein complex and telomerase in maintaining telomere ***,it remains largely unknown whether and how aging-related stresses,such as deregulated protein homeostasis,impact telomere ***,we explored the possible roles of aminoacyl tRNA synthetases(AARSs),key enzymes catalyzing the first reactions in protein synthesis,in regulating telomere length and *** selected seryl tRNA synthetase(SerRS)since our previous studies discovered expanded functions of SerRS in the nucleus in addition to its canonical cytoplasmic role in protein *** this study,we revealed that overexpression of SerRS promoted cellular senescence and inhibited the growth of cervical tumor xenografts in mice by triggering the senescence of tumor *** the nucleus,SerRS directly bound to telomeric DNA repeats and tethered more POT1 proteins to telomeres through a direct interaction between the UNE-S domain of SerRS and the OB1 domain of *** further demonstrated that SerRS-induced enrichment of POT1 prevented the recruitment of telomerase to telomeres,resulting in progressive telomere *** data suggested a possible molecular link between protein synthesis and telomere length control,the deregulation of which may be associated with aging and cancer.

Pathological changes at early stage of multiple organ injury in a rat model of severe acute pancreatitis

BACKGROUND: Severe acute pancreatitis (SAP) is a commonly seen acute abdominal syndrome characterized by sudden onset, rapid progression and high mortality rate. The damage in peripheral organs may be more severe than that in the pancreas, and can even lead to multiple organ dysfunction. It is critical to recognize early pathological changes in multiple organs. This study aimed to assess the early pathological features of damaged organs in a rat model of SAP. METHODS: Thirty clean grade healthy male Sprague-Dawley rats weighing 250-300 g were randomly divided into a model control group (n=15) and a sham-operated group (n=15). The SAP rat model was induced by sodium taurocholate. Samples of blood and from multiple organs were collected 3 hours after operation. We assessed the levels of IL-6, TNF-alpha, PLA2, NO, ET-1, MDA, amylases and endotoxin in blood and observed the early pathological changes in multiple damaged organs. RESULTS: Levels of IL-6, TNF-alpha, PLA2, NO, ET-1 and MDA in serum and of amylase and endotoxin in plasma of the model control group rats were significantly higher than those of the sham-operated group (P<0.01). Different degrees of pathological change were observed in multiple damaged organs. CONCLUSION: Multiple organ injury may occur at the early stage of SAP in rats.

MULTI-TILLERING DWARF1, a new allele of BRITTLE CULM 12, affects plant height and tiller in rice

Plant height and tillering are crucial factors determining rice plant architecture and influencing rice grain production. In this study, rnulti-tillering dwarf1 （mtdl）, a stable multi-tiller and dwarf mutant, was screened from the ethylmethane sulfonate-treated japonica rice variety Wuyunging7. Compared with the wild type, mtdl mutant exhibited pleiotropic phenotypes, including dwarf- ism, more tillers, brittle culms and delayed heading date. By employing map-based cloning strategy, the gene MTD1 was finally mapped to an approximately 66-kb region on the short arm of chromosome 9. Sequencing results showed that the gene LOCOsO9g02650 （BC12） in mtdl mutant had a single nucleotide substitution （G to A）, which gen- erated a premature translation stop. Over-expressing MTD1/BC12 coding sequ（nce rescued all the phenotypes of mtdl mutants including plant height and tillers, which confirms that BC12 is the mutated gene in mtdl mutant. Quantitative reverse tran,-eription-PCR analysis showed that MTDI/BCI2 could negatively regulate the expression of MONOCULM 1, IDEAL PLANT ARCHITECTURE1 and Tillering and Dwarf 1, and control rice tillering. Remark- ably, a-amylase activity analysis and gibberellic acid （GA） treatment showed that the dwarf phenotype of mtdl mutant was dependent on GA biosynthesis pathway. These results facilitated to further uncover the molecular mechanism of the growth and development in rice.

Five new polyhydroxylated furostanol saponins were isolated from the roots and rhizomes of Tupistra chinensis,and their structures were determined as tupistrosides J–N(1–5),together with four known furostanol saponins(6–9),on the basis of physico-chemical properties and spectral *** them,compounds 3 and 5 showed cytotoxicity against human cancer cell lines SW620 with IC50 values of 72.5±2.4 and 77.3±2.5μmol·L^–1,*** 4 showed cytotoxicity against human cancer cell line HepG2 with IC50 value of 88.6±2.1μmol·L^–1.

The biological treatment of wastewater with high concentrations of ammonia nitrogen has become a hot research issue,but there are limited reports on the mechanism of ammonia nitrogen utilization by *** this paper,a transcriptomic approach was used to investigate the differences in gene expression at 500.0 mg/L(Amo 500)and 100.0 mg/L(Amo 100)ammonium concentrations to reveal the mechanism of ammonia nitrogen removal from water by Pseudomonas stutzeri *** transcriptome data showed 1015(459 up-regulated and 556 down-regulated)differentially expressed genes with functional gene annotation related to nitrogen source metabolism,glycolysis,tricarboxylic acid cycle,extracellular polysaccharide synthesis,energy conversion and transmembrane transport,revealing the metabolic process of ammonium nitrogen conversion to biological ni-trogen in *** F2 through *** verify the effect of ammonium transporter protein(AmtB)of cell membrane on assimilation,a *** F2-ΔamtB mutant strain was obtained by constructing a knockout plasmid(pK18mobsacB-ΔamtB),and it was found that the growth characteristics and ammonium removal rate of the mutant strain were significantly reduced at high ammonium *** carbon source components and dissolved oxygen conditions were optimized after analyzing the transcriptome data,and the ammonium removal rate was increased from 41.23%to 94.92%with 500.0 mg/L ammonium *** study of *** F2 transcript level reveals the mechanism of ammonia nitrogen influence on microbial assimilation process and improvement strategy,which provides a new strategy for the treatment of ammonia nitrogen wastewater.
