Comparative proteomic analysis of plasma exosomes reveals the functional contribution of N-acetyl-alpha-glucosaminidase to Parkinson’s disease

Parkinson’s disease is the second most common progressive neurodegenerative disorder,and few reliable biomarkers are available to track disease *** proteins,DNA,mRNA,and lipids carried by exosomes reflect intracellular changes,and thus can serve as biomarkers for a variety of *** this study,we investigated alterations in the protein content of plasma exosomes derived from patients with Parkinson’s disease and the potential therapeutic roles of these proteins in Parkinson’s *** a tandem mass tag-based quantitative proteomics approach,we characterized the proteomes of plasma exosomes derived from individual patients,identified exosomal protein signatures specific to patients with Parkinson’s disease,and identified N-acetyl-alpha-glucosaminidase as a differentially expressed protein.N-acetyl-alpha-glucosaminidase expression levels in exosomes from the plasma of patients and healthy controls were validated by enzyme-linked immunosorbent assay and western *** results demonstrated that the exosomal N-acetyl-alpha-glucosaminidase concentration was not only lower in Parkinson’s disease,but also decreased with increasing Hoehn-Yahr stage,suggesting that N-acetyl-alpha-glucosaminidase could be used to rapidly evaluate Parkinson’s disease ***,western blot and immunohistochemistry analysis showed that N-acetyl-alpha-glucosaminidase levels were markedly reduced both in cells treated with 1-methyl-4-phenylpyridinium and cells overexpressingα-synuclein compared with control ***,N-acetyl-alpha-glucosaminidase overexpression significantly increased cell viability and inhibitedα-synuclein expression in 1-methyl-4-phenylpyridinium-treated *** together,our findings demonstrate for the first time that exosomal N-acetyl-alpha-glucosaminidase may serve as a biomarker for Parkinson’s disease diagnosis,and that N-acetyl-alpha-glucosaminidase may reduceα-synuclein expression and 1-methyl-4-phenylpyridinium-induced n

In this paper a canonical neural network with adaptively changing synaptic weights and activation function parameters is presented to solve general nonlinear programming problems. The basic part of the model is a sub-network used to find a solution of quadratic programming problems with simple upper and lower bounds. By sequentially activating the sub-network under the control of an external computer or a special analog or digital processor that adjusts the weights and parameters, one then solves general nonlinear programming problems. Convergence proof and numerical results are given.

Breaking the structural symmetry of active sites in zeolite imidazolate framework for enhanced CO2 photoreduction

Adjusting the structure and composition of active sites is a highly effective method for activating linear ***, precise control of interfacial chemistry within molecular-scale environments presents a significant challenge in heterogeneous catalysis. In our study, we present symmetry-broken Mo-O-Co triple-atom active sites within the hybrid zeolite imidazole framework HZIF-CoMo. These sites disrupt the electronic balance and create a weak built-in electric field, thus promoting photocatalytic CO2reduction. The CO yield over HZIF-CoMo is improved to 3.1 mmol g-1h-1, which is over four times higher than the parent cobalt-imidazole framework MAF-6-Co. Both experimental and theoretical studies indicate that electronic modulation through the symmetry-breaking Mo-O-Co moiety acts as an organizing knob to accelerate charge separation and result in more carriers participating in the catalytic reaction. This work provides important insights into optimizing site-related electronic structures for targeted photocatalytic reactions.

AIM:To evaluate the efficacy of centralized culture and possible influencing ***:From January 2010 to July 2012,66452 patients with suspected Helicobacter pylori(***) infection from 26 hospitals in Zhejiang and jiangsu Provinces in China underwent gastrointestinal *** mucosal biopsies were taken from the antrum for *** biopsies were transported under natural environmental temperature to the central laboratory in Hangzhou city and divided into three groups based on their transport time:5,24 and 48 *** culture results were reported after 72 h and the positive culture rates were analyzed by a χ2 *** additional 5736 biopsies from ***-positive patients(5646 rapid urease test-positive and 90 14C-urease breath test-positive) were also cultured for quality control in the central laboratory ***:The positive culture rate was 31.66%(21036/66452) for the patient samples and 71.72%(4114/5736) for the ***-positive quality control *** the 5 h transport group,the positiveculture rate was 30.99%(3865/12471),and 32.84%(14960/45553) in the 24 h transport *** contrast,the positive culture rate declined significantly in the 48 h transport group(26.25%; P < 0.001).During transportation,the average natural temperature increased from 4.67 to 29.14℃,while the positive culture rate declined from 36.67%(1462/3987) to 24.12%(1799/7459).When the temperature exceeded 24℃,the positive culture rate decreased significantly,especially in the 48 h transport group(23.17%).CONCLUSION:Transportation of specimens within 24 h and below 24℃ is reasonable and acceptable for centralized culture of multicenter *** samples.

Thermally activated delayed fluorescence enhanced by macrocyclization

Thermally activated delayed fluorescence (TADF) depends on reverse intersystem crossing (RISC) from the lowest triplet to the singlet excited state. Here, a macrocyclization strategy is developed to achieve TADF emission by promoting RISC. 5-(9Hcarbazol-9-yl) isophthalonitrile, a fluorescent molecule, is coupled with 1,4-dimethoxybenzene by methylene bridges to form a macrocycle with TADF emission. Experimental results and theoretical calculation indicate that the macrocyclization strategy contributes to RISC promotion and suppression of nonradiative decay, resulting in effective TADF emission transformed from prompt fluorescence. Three TADF luminophores are cyclized with the same strategy, and an enhanced TADF emission of up to3.8 folds is observed. The marriage of macrocycle chemistry and luminescent materials will provide new guidelines for designing efficient TADF materials.

Objective: To prepare water-soluble quantum dots (QDs) bioconjugates for binding specifically to carcinoma cells. Method: Quantum dots have been conjugated with a certain carcinoma cells-targeting peptide using a bifunctional cross-linking reagent. And the MDA- MB- 435S breast carcinoma cells were then incubated with QDs-peptide for different time. Results: The solution of QDs-peptide conjugates emitted bright fluorescence and could remain transparent without precipitates for about half a year. These bioconjugates could specifically recognize the MDA- MB- 435S breast carcinoma cells, but not recognize the HL-60 promyelocytic leukemia cells. Additionally, confocal optical slices imaging showed that the QDs had been internalized by the receptor-mediated endocytosis of the MDA- MB- 435S carcinoma cells if the bioconjugates were incubated with the cells for about 48h at 37℃. Conclusions: In summary, we have developed a class of peptide-QDs bioconjugates for carcinoma cells targeting and imaging. The preparation of the bioconjugates was convenient, and the cross-linking reagent could react with the peptide and QDs easily. These nanoprobes were water-soluble, and bright enough for biological imaging. The peptide linked to the QDs remained high biological activities.

Role of the globus pallidus in motor and non-motor symptoms of Parkinson's disease

The globus pallidus plays a pivotal role in the basal ganglia circuit. Parkinson's disease is characterized by degeneration of dopamine-producing cells in the substantia nigra, which leads to dopamine deficiency in the brain that subsequently manifests as various motor and non-motor symptoms. This review aims to summarize the involvement of the globus pallidus in both motor and non-motor manifestations of Parkinson's disease. The firing activities of parvalbumin neurons in the medial globus pallidus, including both the firing rate and pattern, exhibit strong correlations with the bradykinesia and rigidity associated with Parkinson's disease. Increased beta oscillations, which are highly correlated with bradykinesia and rigidity, are regulated by the lateral globus pallidus. Furthermore,bradykinesia and rigidity are strongly linked to the loss of dopaminergic projections within the cortical-basal ganglia-thalamocortical loop. Resting tremors are attributed to the transmission of pathological signals from the basal ganglia through the motor cortex to the cerebellum-ventral intermediate nucleus circuit. The cortico–striato–pallidal loop is responsible for mediating pallidi-associated sleep disorders. Medication and deep brain stimulation are the primary therapeutic strategies addressing the globus pallidus in Parkinson's disease. Medication is the primary treatment for motor symptoms in the early stages of Parkinson's disease, while deep brain stimulation has been clinically proven to be effective in alleviating symptoms in patients with advanced Parkinson's disease,particularly for the movement disorders caused by levodopa. Deep brain stimulation targeting the globus pallidus internus can improve motor function in patients with tremordominant and non-tremor-dominant Parkinson's disease, while deep brain stimulation targeting the globus pallidus externus can alter the temporal pattern of neural activity throughout the basal ganglia–thalamus network. ＜正＞We report two cases of ectopic pancreas in the gastric antrum that were removed using minimally invasive approaches.A 65-year-old woman with bloating,dysphasia and heartburn,and a 49-year-old woman with developed sporadic epigastric pain and vomiting were referred for upper gastroendoscopic examination.A submucosal lesion in the antrum was revealed in both of them.A le- sion 2 cm in diameter was located in the anterior wall at 5 cm from the pylorus in the first patient,a 1.5 cm lesion in the posterior wall with a distance of 3 cm from the pylorus in the other patient.A la- paroseopic wedge resection of the gastric antrum was performed for the first patient.A distal Bill- roth-Ⅰ gastrectomy was Tarried out for the second *** of them had uneventful recoveries on 3-6 months *** experiences suggest that laparoscopic wedge resection and distal Bill- roth-Ⅰ gastrectomy are safe and effective approaches for treatment of ectopic pancreas in gastric antrum.

Staged dendrite suppression for high safe and stable lithium-sulfur batteries

The unavoidable dendrite growth and shuttle effect have long been stranglehold challenges limiting the safety and practicality of lithium-sulfur ***,we propose a dual-action strategy to address the lithium dendrite issue in stages by constructing a multifunctional surface-negatively-charged nanodiamond layer with high ductility and robust puncture resistance on polypropylene （PP） *** uniformly loaded compact negative layer can not only significantly enhance electron transmission efficiency and promote uniform lithium deposition,but also reduce the formation of dendrite during early deposition *** importantly,under the strong puncture stress encountered during the deterioration of lithium dendrite growth under limiting current,the high ductility and robust puncture resistance（145.88 MPa） of as-obtained nanodiamond layer can effectively prevent short circuits caused by unavoidable lithium *** Li||Li symmetrical cells assembled with nanodiamond layer modified PP demonstrated a stable cycle of over 1000 h at 2 mA cm-2with a polarization voltage of only 29.3 ***,the negative charged layer serves as a physical barrier blocking lithium polysulfide ions,effectively mitigating capacity *** improved cells achieved a capacity decay of only 0.042%per cycle after 700 cycles at 3 C,demonstrating effective suppression of dendrite growth and capacity attenuation,showing promising prospect.
