Lateral characteristics improvements of DBR laser diode with tapered Bragg grating

Broad area semiconductor laser(BAL)has poor lateral beam quality due to lateral mode competition,which limits its application as a high-power optical *** this work,the distributed Bragg reflector laser diode with tapered grating(TDBR-LD)is *** changing the lateral width,the tapered grating increases the loss of high-order lateral modes,thus improving the lateral characteristics of the laser *** measuring results show that the TDBR-LD can achieve a single-lobe output under 0.9 *** contrast to the straight distributed Bragg reflector laser diode(SDBR-LD),the lateral far field divergence of TDBR-LD is measured to be 5.23°at 1 A,representing a 17%*** linewidth of TDBR-LD is 0.4 nm at 0.2 A,which is reduced by nearly 43%in comparison with that of ***,both of the devices have a maximum output power value of approximate 470 mW.

Exploring binding mode for styrylquinoline HIV-1 integrase inhibitors using comparative molecular field analysis and docking studies

AIM: To understand pharmacophore properties of styrylquinoline derivatives and to design inhibitors of HIV-1 integrase. METHODS: Comparative molecular field analysis (CoMFA) was performed to analyze three-dimen- sional quantitative structure-activity relationship (3D-QSAR) of styrylquinoline derivatives. Thirty-eight compounds were randomly divided into a training set of 28 compounds and a test set of 10 compounds. The stability of 3D- QSAR models was proved by the analysis of cross-validated and non-cross-validated methods. Moreover, the binding mode of these compounds and integrase was constructed by AutoDock program. RESULTS: The CoMFA model of the training compounds was reasonably predicted with cross-validated coefficient (q2) and conventional (r2) values (up to 0.696 and 0.754). Then the model was validated by the test set. The resulting CoMFA maps visualized structural requirements for the biological activity of these inhibitors. Docking results showed that a carboxyl group at C-7 and a hydroxyl group at C-8 in the quinoline subunit, bound closely to the divalent metal cofactor (Mg2+) around the integrase catalytic site. Moreover, there is a linear correlation between the binding energy of the inhibitors with integrase and their inhibitory effect. CONCLUSIONS: The present study indicated that the CoMFA model together with docking results could give us helpful hints for drug design as well as interpre- tation of the binding affinity between these inhibitors and integrase.

Spatial and spectral filtering of tapered lasers by using tapered distributed Bragg reflector grating

A 1040 nm tapered laser with tapered distributed Bragg reflector(DBR) grating is designed and fabricated. By designing the grating with tapered layout, the tapered DBR grating exhibits the scattering effect on side backward-traveling waves, thus achieving additional suppression of parasitic oscillation. Under the suppression of parasitic oscillation, the spatial and spectral characteristics of the tapered laser are improved. The experimental results show that a near-Gaussian far-field distribution and a kink-free P–I characteristics are achieved, and a single peak emission with a wavelength of1046.84 nm and a linewidth of 56 pm is obtained.

High-throughput real-time assay based on molecular beacons for HIV-1 integrase 3'-processing reaction

Aim:To develop a high-throughput real-time assay based on molecular beaconsto monitor the integrase 3’-processing reaction in vitro and apply it to ***:The recombinant human immunodeficiency virus (HIV)-1integrase (IN) is incubated with a 38 mer oligonucleotide substrate,a sequenceidentical to the U5 end of HIV-1 long terminal repeats (LTR).Based on the fluores-cence properties of molecular beacons,the substrate is designed to form a stem-loop structure labeled with a fluorophore at the 5’ end and a quencher at the 3’ *** cleaves the terminal 3’-dinucleotide containing the quencher,resulting in anincrease in fluorescence which can be monitored on a *** this assay,tests were performed to investigate the effects of substrates,enzyme and the metal ion concentrations on the IN activity and optimal param-eters were ***,2 IN inhibitors were employed to test the perfor-mance of this assay in antiviral compound ***:The fluorescentintensity of the reaction mixture varies linearly with time and is proportional to thevelocity of the 3’-processing *** were performed and the results showedthat the optimal rate was obtained for a reaction mixture containing 50 mg/L recom-binant HIV-1 IN,400 nmol/L substrate,and 10 mmol/L Mn2+.The IN 3’-processingreaction under the optimal conditions showed a more than 18-fold increase in thefluorescence intensity compared to the enzyme-free *** IC50values ofthe IN inhibitors obtained in our assay were similar to the values obtained from aradiolabeled substrate ***:Our results demonstrated that this is afast,reliable,and sensitive method to monitor HIV IN 3’-processing reaction andthat it can be used for inhibitor screening.

Myocardial ischemia/reperfusion(/R)injury is a classic type of cardiovascular disease characterized by injury to cardiomyocytes leading to various forms of cell *** is believed that irreversible myocardial damage resulted from I/R occurs due to oxidative stress evoked during the reperfusion *** we demonstrate that ischemia triggers a specific redox reaction of polyunsaturated fatty acids(PUFA)-phospholipids in myocardial cells,which acts as a priming signaling that initiates the outbreak of robust oxidative damage in the reperfusion *** animal and in vitro models,the crucial lipid species in I/R injury were identifed to be oxidized PUFAs enriched *** multi-omics,arachidonic acid 15-lipoxygenase-1(ALOx15)was identified as the primary mediator of ischemia-provoked phospholipid peroxidation,which was further confirmed using chemogenetic ***,our results reveal that ALOx15 induction in the ischemia phase acts as a"burning point"to ignite phospholipid oxidization into ferroptotic *** finding characterizes a novel molecular mechanism for myocardial ischemia injury and offers a potential therapeutic target for early intervention of V/R injury.
