Gene Expressions Underlying Mishandled Calcium Clearance and Elevated Generation of Reactive Oxygen Species in the Coronary Artery Smooth Muscle Cells of Chronic Heart Failure Rats

Background： The calcium clearance and reactive oxygen species （ROS） generations in the coronary artery smooth muscle cells in chronic heart failure （HF） have not been fully investigated. Therefore, we attempted to understand the gene expressions underlying the mishandling of calcium clearance and the accumulations of ROS. Methods： We initially established an animal model of chronic HF by making the left anterior descending coronary artery ligation （CAL） in rats, and then isolated the coronary artery vascular smooth muscle cells from the ischemic and the nonischemic parts of the coronary artery vessels in 12 weeks after CAL operation. The intracellular calcium concentration and ROS level were measured using flow cytometry, and the gene expressions of sarco/endoplasmic reticulum Ca2＋-ATPase （SERCA2a）, encoding sarcoplasmic reticulum Ca2＋-ATPase 2a, encoding sodium-calcium exchanger （NCX）, andp47phox encoding a subunit of the nicotinamide adenine dinucleotide phosphate （NADPH） oxidase were examined using real-time quantitative reverse transcription polymerase chain reaction and Western blotting, respectively. Results： We found that the calcium accumulation and ROS generation in the coronary artery smooth muscle cells isolated from either the ischemic or the nonischemic part of the CAL coronary artery vessel were significantly increased irrespective of blood supply （all P 〈 0.01 ）. Moreover, these were accompanied by the increased expressions of NCX and p47phox, the decreased expression of S ERCA2a, and the increased amount of phosphorylated forms of p47phox in NADPH oxidase （all P 〈 0.05）. Conclusions： Our results demonstrated that the disordered calcium clearance and the increased ROS generation occurred in the coronary artery smooth muscle cells in rats with chronic HF produced by ligation of the left anterior descending coronary artery （CAL）, and which was found to be disassociated from blood supply, and the increased generation of ROS in the ceils was found to make

Slickwater fracturing fluids are widely used in the development of unconventional oil and gas resources due to the advantages of low cost,low formation damage and high drag reduction ***,their performance is severely affected at high *** reducing agent is the key to determine the drag reducing performance of *** this work,in order to further improve the temperature resistance of slickwater,a temperature-resistant polymeric drag reducing agent(PDRA)was synthesized and used as the basis for preparing the temperature-resistant *** slickwater system was prepared with the compositions of 0.2 wt%PDRA,0.05 wt%drainage aid nonylphenol polyoxyethylene ether phosphate(NPEP)and 0.5 wt%anti-expansion agent polyepichlorohydrindimethylamine(PDM).The drag reduction ability,rheology properties,temperature and shear resistance ability,and core damage property of slickwater were systematically studied and *** contrast to on-site drag reducing agent(DRA)and HPAM,the temperature-resistant slickwater demonstrates enhanced drag reduction efficacy at 90℃,exhibiting superior temperature and shear resistance ***,the drag reduction retention rate for the slickwater achieved an impressive 90.52%after a 30-min shearing ***,the core damage is only 5.53%.We expect that this study can broaden the application of slickwater in high-temperature reservoirs and provide a theoretical basis for field applications.

Enhancing Unsupervised Domain Adaptation for Person Re-Identification with the Minimal Transfer Cost Framework

In Unsupervised Domain Adaptation(UDA)for person re-identification(re-ID),the primary challenge is reducing the distribution discrepancy between the source and target *** can be achieved by implicitly or explicitly constructing an appropriate intermediate domain to enhance recognition capability on the target *** construction is difficult due to the absence of intermediate state supervision,making smooth knowledge transfer from the source to the target domain a *** explicitly construct the most suitable intermediate domain for the model to gradually adapt to the feature distribution changes from the source to the target domain,we propose the Minimal Transfer Cost Framework(MTCF).MTCF considers all scenarios of the intermediate domain during the transfer process,ensuring smoother and more efficient domain *** framework mainly includes threemodules:Intermediate Domain Generator(IDG),Cross-domain Feature Constraint Module(CFCM),and Residual Channel Space Module(RCSM).First,the IDG Module is introduced to generate all possible intermediate domains,ensuring a smooth transition of knowledge fromthe source to the target *** reduce the cross-domain feature distribution discrepancy,we propose the CFCM Module,which quantifies the difficulty of knowledge transfer and ensures the diversity of intermediate domain features and their semantic relevance,achieving alignment between the source and target domains by incorporating mutual information and maximum mean *** also design the RCSM,which utilizes attention mechanism to enhance the model’s focus on personnel features in low-resolution images,improving the accuracy and efficiency of person *** proposed method outperforms existing technologies in all common UDA re-ID tasks and improves the Mean Average Precision(mAP)by 2.3%in the Market to Duke task compared to the state-of-the-art(SOTA)methods.

Exploring multiple phases and first-order phase transitions in Kármán Vortex Street

Kármán Vortex Street, a fascinating phenomenon of fluid dynamics, has intrigued the scientific community for a long time. Many researchers have dedicated their efforts to unraveling the essence of this intriguing flow pattern. Here, we apply the lattice Boltzmann method with curved boundary conditions to simulate flows around a circular cylinder and study the emergence of Kármán Vortex Street using the eigen microstate approach, which can identify phase transition and its order-parameter. At low Reynolds number, there is only one dominant eigen microstate W_(1) of laminar flow. At Re_(c)^(1)= 53.6, there is a phase transition with the emergence of an eigen microstate pair W^(2,3) of pressure and velocity fields. Further at Re_(c)^(2)= 56, there is another phase transition with the emergence of two eigen microstate pairs W^(4,5) and W^(6,7). Using the renormalization group theory of eigen microstate,both phase transitions are determined to be first-order. The two-dimensional energy spectrum of eigen microstate for W^(1), W^(2,3) after Re_(c)^(1), W^(4-7) after Re_(c)^(2) exhibit-5/3 power-law behavior of Kolnogorov's K41 theory. These results reveal the complexity and provide an analysis of the Kármán Vortex Street from the perspective of phase transitions.

A new scheme of climate-vegetation regionalization in the Hengduan Mountains Region

The Hengduan Mountains Region(HMR) is essential for the future ecological protection, clean energy production,Sichuan-Xizang and yunnan-Xizang railways, and other major infrastructure projects in China. The distributions of climate and vegetation exhibit significant regional differentiation and vertical zonality due to the rugged longitudinal ranges and gorges and the complex disaster-prone environments in HMR. Therefore, it is urgent to develop the climate-vegetation regionalization in HMR to effectively satisfy the national requirements such as agricultural production and ecological protection, mountain disaster risk prevention, and major project construction. We here develop a new scheme of climate-vegetation regionalization with the latest demarcation outcome of HMR, the ground observation from 122 meteorological stations in HMR and its surrounding areas during 1990–2019, and the high-precision remote sensing data of land cover types. The new scheme first constructs the regionalization index system, fully considering the extraordinarily complicated geomorphic pattern of mountains and valleys, the scarcity of meteorological observations, and the remarkable differentiation of climate and vegetation in HMR. The system consists of three primary regionalization indices(i.e., days with daily average temperature steady above 10°C, aridity index, and main vegetation types, dividing the temperature zones, moisture regions, and vegetation subregions, respectively) and three auxiliary indices of the accumulated temperature above 10°C, and the temperatures in January and July. Then, the HMR is divided into five temperature zones, 20 moisture regions, and 55 vegetation subregions. Comparative evaluation of SPE methods for biotoxicity assessment of water and wastewater:Linkage between chemical extracting efficiency and biotoxicity outcome

Biotoxicity assessment results of environmental waters largely depend on the sample extraction protocols that enrich pollutants to meet the effect-trigger thresholds of ***,more chemical mixture does not necessarily translate to higher combined ***,there is a need to establish the link between chemical extracting efficiency and biotoxicity outcome to standardize extraction methods for biotoxicity assessment of environmental *** study compares the performance of five different extraction phases in solid phase extraction(SPE),namely HLB,HLB+Coconut,C18 cartridge,C18 disk and Strata-X,and evaluated their chemical extracting efficiencies and biotoxicity *** quantitatively assessed cytotoxicity,acute toxicity,genotoxicity,estrogenic activity,and neurotoxicity of the extracts using in vitro bioassays and characterized the chemical extracting efficiencies of the SPE methods through chemical recoveries of 23 model compounds with different polarities and total organic *** Pareto ranking,we identified HLB+Coconut as the optimal SPE method,which exhibited the highest level of water sample biotoxicity and recovered the most chemicals in water *** found that the biotoxicity outcomes of the extracted water samples significantly and positively correlated with the chemical extracting efficiencies of the SPE ***,we observed synchronous changing patterns in biotoxicity outcome and chemical extracting efficiencies in response to increasing sample volumes per cartridge(SVPC)during *** findings underscore that higher chemical extracting efficiency of SPE corresponds to higher biotoxicity outcome of environmental water samples,providing a scientific basis for standardization of SPE methods for adequate assessment of biotoxicities of environmental waters.

Understanding gene expression variations between species is pivotal for deciphering the evolutionary diversity in phenotypes. Rhesus macaques(Macaca mulatta, MMU)and crab-eating macaques(M. fascicularis, MFA) serve as crucial nonhuman primate biomedical models with different phenotypes. To date, however, large-scale comparative transcriptome research between these two species has not yet been fully explored. Here, we conducted systematic comparisons utilizing newly sequenced RNA-seq data from84 samples(41 MFA samples and 43 MMU samples)encompassing 14 common tissues. Our findings revealed a small fraction of genes(3.7%) with differential expression between the two species, as well as 36.5% of genes with tissue-specific expression in both macaques. Comparison of gene expression between macaques and humans indicated that 22.6% of orthologous genes displayed differential expression in at least two tissues. Moreover,19.41% of genes that overlapped with macaque-specific structural variants showed differential expression between humans and macaques. Of these, the FAM220A gene exhibited elevated expression in humans compared to macaques due to lineage-specific duplication. In summary,this study presents a large-scale transcriptomic comparison between MMU and MFA and between macaques and humans. The discovery of gene expression variations not only enhances the biomedical utility of macaque models but also contributes to the wider field of primate genomics.
