AIM： To investigate the effect of NS398 on the metastasis-associated gene expression in LoVo colorectal cancer cells.METHODS： LoVo cells were treated with NS398 at the concentration of 100 IJmol/L for 24 and 48 h respectively. Total RNA was extracted with TRIzol reagents and reverse transcribed with Superscript Ⅱ and hybridized with cDNA mlcroarray （containing oncogenes, tumor suppressor genes, signal transduction molecules, adhesive molecules, growth factors, and ESTs） fabricated in our laboratory. After normalization, the ratio of gene expression of NS398 treated to untreated LoVo cells was either 2-fold up or 0.5-fold down was defined as the differentially expressed genes. Semi-quantitative RT-PCR was used to validate the microarray results.RESULTS： Among the 447 metastasis-associated genes, 9 genes were upregulated and 8 genes were downregulated in LoVo cells treated with NS398 for 24 h compared to untreated cells. While 31 genes were upregulated and 14 genes were downregulated in LoVo cells treated with NS398 for 48 h. IGFBP-5, PAI-2, JUN,REL, BRCA1, and BRCA2 might be the new targets of NS398 in treatment of colorectal cancer.CONCLUSION： NS398 might exert its anti-metastasis effect on colorectal cancer by affecting several metastasis-associated gene expression.

The sensitivity of the dark photon search through invisible decay final states in low-background experiments relies sig-nificantly on the neutron and muon veto efficiencies,which depend on the amount of material used and the design of the detector *** paper presents the optimized design of the hadronic calorimeter(HCAL)used in the DarkSHINE experiment,which is studied using a GEANT4-based simulation *** geometry is optimized by comparing a traditional design with uniform absorbers to one that uses different thicknesses at different locations on the detector,which enhances the efficiency of vetoing low-energy neutrons at the sub-GeV *** overall size and total amount of material used in the HCAL are optimized to be lower,owing to the load and budget requirements,whereas the overall performance is studied to satisfy the physical objectives.

The Xuyi Schmidt Telescope Photometric Survey of the Galactic Anti-center（XSTPS-GAC）is a photometric sky survey that covers nearly 6000 deg^2 towards the Galactic Anti-center（GAC） in the g,r,i bands. Half of its survey field is located on the Galactic Anti-center disk,which makes XSTPSGAC highly suitable to search for new open clusters in the GAC region. In this paper,we report new open cluster candidates discovered in this survey,as well as properties of these open cluster candidates,such as age,distance and reddening,derived by isochrone fitting in the color-magnitude diagram（CMD）.These open cluster candidates are stellar density peaks detected in the star density maps by applying the method from Koposov et al. Each candidate is inspected in terms of its true color image composed from three XSTPS-GAC band images. Then its CMD is checked,in order to identify whether the central region stars have a clear isochrone-like trend differing from background stars. The parameters derived from isochrone fitting for these candidates are mainly based on three band photometry of ***,when these new candidates are able to be seen clearly in 2 MASS data,their parameters are also derived based on the 2 MASS（J-H,J） CMD. There are a total of 320 known open clusters rediscovered and 24 new open cluster candidates discovered in this work. Furthermore,the parameters of these new candidates,as well as another 11 previously known open clusters,are properly determined for the first time.

Although microsurgical vasoepididymostomy(MVE)is an effective treatment for epididymal obstructive azoospermia,some patients may experience delayed patency or suboptimal semen parameters after ***,research into patency time,semen quality postpatency,and associated influencing factors remains *** study aimed to address these issues by evaluating 181 patients who underwent at least one-sided MVE employing asingle-armed longitudinal intussusception vasoepididymostomy technique,with a follow-up period of over 12 months for 150 *** overall patency rate was 75.3%,with 86.0%of patients achieving patency within 6 months following ***,factors such as age,history of epididymitis,duration of surgery,side of anastomosis,sperm motility in epididymal fluid,and the site of anastomosis showed no correlation with patency ***,our univariate and multivariate linear regression analysis indicated that only the site of anastomosis was positively correlated with and could independently predict postoperative total motile sperm ***,the site of anastomosis might serve as a predictor for optimal postoperative semen quality following the MVE procedure.

AIM: To discover novel biomarkers for early diagnosis, prognosis or treatment of human colorectal cancer. METHODS: i TRAQ 2D LC-MS/MS analysis was used to identify differentially expressed proteins(DEPs) in the human colonic epithelial carcinogenic process using laser capture microdissection-purified colonic epithelial cells from normal colon, adenoma, carcinoma in situ and invasive carcinoma tissues. RESULTS: A total of 326 DEPs were identified, and four DEPs(DMBT1, S100A9, Galectin-10, and S100A8) with progressive alteration in the carcinogenic process were further validated by immunohistochemistry. The DEPs were involved in multiple biological processes including cell cycle, cell adhesion, translation, m RNA processing, and protein synthesis. Some of the DEPs involved in cellular process such as "translation" and "m RNA splicing" were progressively up-regulated, while some DEPs involved in other processes such as "metabolism" and "cell response to stress" was progressively downregulated. Other proteins with up- or down-regulation at certain stages of carcinogenesis may play various roles at different stages of the colorectal carcinogenic process. CONCLUSION: These findings give insights into our understanding of the mechanisms of colorectal carcinogenesis and provide clues for further investigation of carcinogenesis and identification of biomarkers.
