The lysine methyltransferase SMYD2 facilitates neointimal hyperplasia by regulating the HDAC3-SRF axis

Coronary restenosis is an important cause of poor long-term prognosis in patients with coronary heart disease.Here,we show that lysine methyltransferase SMYD2 expression in the nucleus is significantly elevated in serum-and PDGF-BB-induced vascular smooth muscle cells(VSMCs),and in tissues of carotid artery injury-induced neointimal hyperplasia.Smyd2 overexpression in VSMCs(Smyd2-vTg)facilitates,but treatment with its specific inhibitor LLY-507 or SMYD2 knockdown significantly inhibits VSMC phenotypic switching and carotid artery injury-induced neointima formation in mice.Transcriptome sequencing revealed that SMYD2 knockdown represses the expression of serum response factor(SRF)target genes and that SRF overexpression largely reverses the inhibitory effect of SMYD2 knockdown on VSMC proliferation.HDAC3 directly interacts with and deacetylates SRF,which enhances SRF transcriptional activity in VSMCs.Moreover,SMYD2 promotes HDAC3 expression via tri-methylation of H3K36 at its promoter.RGFP966,a specific inhibitor of HDAC3,not only counteracts the pro-proliferation effect of SMYD2 overexpression on VSMCs,but also inhibits carotid artery injury-induced neointima formation in mice.HDAC3 partially abolishes the inhibitory effect of SMYD2 knockdown on VSMC proliferation in a deacetylase activity-dependent manner.Our results reveal that the SMYD2-HDAC3-SRF axis constitutes a novel and critical epigenetic mechanism that regulates VSMC phenotypic switching and neointimal hyperplasia.

Modulated noble metal/2D MOF heterostructures for improved hydrogen storage of MgH2

The conjugation of external species with twodimensional（2D） materials has broad application prospects.In this study,we have explored the potential of noble metal/2D MOF heterostructures in hydrogen storage.Specifically,the MgH2-Ni-MOF@Pd system has shown remarkable hydrogen desorption/sorption performances,starting to liberate hydrogen at 181 0C,which is 230 0C lower than that of pristine MgH2.Under the catalytic effect of Ni-MOF@Pd,the dehydrogenation apparent activation energy of MgH2 is noticeably decreased from（133.5 ± 17.5） to（34.58 ± 1.87） kJ·mol-1,and the hydrogenation apparent activation energy is reduced from（70.41 ± 7.43） to（25.78 ± 4.64） kJ·mol-1,which is lowered by 63.4%.The fully-dehydrogenated MgH2-NiMOF@Pd composite rapidly uptakes hydrogen,with 2.62wt% at 100℃ and 6.06 wt% at 150℃ within 300 s,respectively.The mechanism analysis of MgH2 catalyzed by Ni-MOF@Pd has revealed that the transformation of Mg2Ni and Mg2NiH4could act as a "hydrogen pump",providing numerous channels for fast diffusion and transport of hydrogen atoms.Moreover,in the dehydrogenation process,the element Pd reacts with MgH2to form the MgPd alloy phase,which makes MgH2take precedence to decompose through the Mg-Pd alloy rather than self-decomposition,further reducing thermal stability and improving de/hydrogenation kinetics.The synergistic effect of Mg-Pd,Mg2Ni,and the special ultra-thin 2D sheet structure of the additive is the main reason for the good hydrogen storage property of MgH2-Ni-MOF@Pd.Our findings provide inspiration for designing efficient multifunctional additives with unique morphologies to optimize the hydrogen desorption/sorption behaviors of hydrogen storage materials.

BACKGROUND Acute-on-chronic liver disease(AoCLD)accounts for the majority of patients hospitalized in the Department of Hepatology or Infectious Diseases.AIM To explore the characterization of AoCLD to provide theoretical guidance for the accurate diagnosis and prognosis of AoCLD.METHODS Patients with AoCLD from the Chinese Acute-on-Chronic Liver Failure(ACLF)study cohort were included in this study.The clinical characteristics and outcomes,and the 90-d survival rate associated with each clinical type of AoCLD were analyzed,using the Kaplan-Meier method and the log-rank test.RESULTS A total of 3375 patients with AoCLD were enrolled,including 1679(49.7%)patients with liver cirrhosis acute decompensation(LC-AD),850(25.2%)patients with ACLF,577(17.1%)patients with chronic hepatitis acute exacer-bation(CHAE),and 269(8.0%)patients with liver cirrhosis active phase(LC-A).The most common cause of chronic liver disease(CLD)was HBV infection(71.4%).The most common precipitants of AoCLD was bacterial infection(22.8%).The 90-d mortality rates of each clinical subtype of AoCLD were 43.4%(232/535)for type-C ACLF,36.0%(36/100)for type-B ACLF,27.0%(58/215)for type-A ACLF,9.0%(151/1679)for LC-AD,3.0%(8/269)for LC-A,and 1.2%(7/577)for CHAE.CONCLUSION HBV infection is the main cause of CLD,and bacterial infection is the main precipitant of AoCLD.The most common clinical type of AoCLD is LC-AD.Early diagnosis and timely intervention are needed to reduce the mortality of patients with LC-AD or ACLF.

＜正＞Objective:The present study was designed to test the hypothesis that Ligustrazin can inhibit hydrogen peroxide（H2O2）-induced injury in human umbilical vein endothelial cells（HUVECs）,and Ligustrazin has a preventive effect on atherosclerosis at the genetic level.Methods:The oxidative injury model was established with H2O2.HUVECs were treated with different concentrations of Ligustrazin in the presence of H2O2.The protective effects of Ligustrazin against H2O2-injury were investigated in the present study.Results:HUVECs incubated with 200?mol/L H2O2 had significantly decreased in cell viability,which was accompanied with apparent cells apoptosis and the up-regulative expression level of Bid,Caspase-3,8,9 mRNA.In addition,the results of quantitative real-time polymerase chain reaction （qRT-PCR）indicated that pretreatment with Ligustrazine down-regulated the expression of Bid and Caspase-3,8,9 mRNA,which had a key role in H2O2-induced cell apoptosis.Conclusion:The study shows that Ligustrazine can protect endothelial cells against oxidative injury induced by H2O2,suggesting that Ligustrazine is a potential compound for the prevention and treatment of atherosclerosis.

【目的】研究含热带种质玉米自交系胚乳发育分子机制及相关调控网络。【方法】通过高通量RNA-seq测序技术,对热带血缘玉米自交系ZOL-1在授粉后5、10、15、20、25、30 d 6个时期的胚乳转录组进行比较,得到的差异表达基因进行KEGG与GO功能注释,并对FPKM平均值大于1的基因进行时间序列分析。【结果】玉米自交系ZOL-1胚乳发育在授粉后25~30 d时籽粒长、宽、厚增长速率最快,增长率分别达到15.9%、16.1%和10.2%。经过转录组测序和同参考基因组比对,过滤后reads占比至少为83.99%,测序质量和组装效果均较好。按照连续比较系统方法,在10 d_VS_5 d、15 d_VS_10 d、20 d_VS_15 d、25 d_VS_20 d和30 d_VS_25 d分别检测到7129、5252、4254、775和312个差异表达基因。GO功能注释表明,差异表达基因主要分布于细胞分化过程、代谢过程、催化活性和细胞器等条目中。KEGG功能富集分析显示,差异表达基因主要富集在能量代谢、碳水化合物代谢、碳代谢、氨基酸代谢、信号转导等与胚乳发育相关的通路。通过对FPKM平均值大于1的基因进行时间序列分析,19554个基因共划分为10个Clusters,并在Cluster 7鉴定出8个淀粉合成酶相关基因,它们的表达量在授粉后15 d时开始显著提升,此后保持较高表达水平。【结论】玉米自交系胚乳发育可能主要受下调基因表达调控,在授粉后15 d前后胚乳的发育模式发生变化,转录组有重新编程的迹象,同时挖掘到玉米淀粉生物合成相关基因8个,有利于研究胚乳发育的分子调控机理。

Bi_(2)Te_(3)基化合物是目前室温附近性能最好的热电材料,但其存在着大量复杂的缺陷结构,缺陷工程是调控材料热电性能的核心手段,因此理解和有效地调控缺陷形态和浓度是获得高性能Bi_(2)Te_(3)基热电材料的关键.本文系统地研究了四元n型Bi_(2-x)Sb_(x)Te_(3-y)Se_(y)基化合物的缺陷演化过程及其对热电输运性能的影响规律.Sb和Se的固溶引入的带电伴生结构缺陷使得材料的载流子浓度发生了巨大变化,在Bi_(2-x)Sb_(x)Te_(2.994)Cl_(0.006)样品中,Sb的固溶降低了反位缺陷Sb_(Te)_(2)形成能,诱导产生了反位缺陷Sb_(Te)_(2),使得少数载流子空穴浓度从2.09×10^(16)cm^(-3)增加至3.99×10^(17)cm^(-3),严重劣化了电性能.在Bi_(1.8)Sb_(0.2)Te_(2.994-y)Se_(y)Cl_(0.006)样品中,Se的固溶使得Se(Te)_(2)+SbBi的缺陷形成能更低,抑制了反位缺陷Sb_(Te)_(2)的产生,Bi_(1.8)Sb_(0.2)Te_(2.694)Se_(0.30)Cl_(0.006)样品的少数载流子空穴浓度降至1.49×10^(16)cm^(-3),消除了其对材料热电性能的劣化效果,显著地提升了材料的功率因子,室温下达到4.49 mW/(m·K^(2)).结合Sb和Se固溶增强合金化散射降低材料的热导率,Bi_(1.8)Sb_(0.2)Te_(2.844)Se_(0.15)Cl_(0.006)样品在室温下获得最大ZT值为0.98.该研究为调控具有复杂成分的Bi_(2)Te_(3)基材料的点缺陷、载流子浓度和热电性能提供了重要的指导.

目的建立双金属复合管环焊缝耐蚀性能评价方法,为双金属复合管在高酸性气田现场应用提供依据。方法针对双金属复合管环焊缝在高酸性环境中的主要腐蚀类型,参考标准Q/SY 06018-2016中冶金复合管直焊缝和堆焊层腐蚀评价内容,确定以晶间腐蚀、抗SCC/SSC性能、点腐蚀、模拟工况腐蚀为评价指标,并通过整管段试验验证评价结果的双金属复合管环焊缝耐蚀性能评价方法,同时以国产L360/825冶金复合管环焊缝为例对建立的评价方法进行验证。结果通过使用建立的方法对L360/825双金属复合管环焊缝耐蚀性能进行评价,验证了评价方法有效可靠,评价结果显示L360/825双金属复合管环焊缝具有良好的耐蚀性能。结论建立的评价方法可以直接应用于高酸性气田用双金属复合管的环焊缝耐蚀性能评价,为双金属复合管的现场应用提供依据。

Objective:This cross-sectional study aimed to determine the epidemiology of olfactory and gustatory dysfunctions related to COVID-19 in China.Methods:This study was conducted by 45 tertiary Grade-A hospitals in China.Online and offline questionnaire data were obtained from patients infected with COVID-19 between December 28,2022,and February 21,2023.The collected information included basic demographics,medical history,smoking and drinking history,vaccination history,changes in olfactory and gustatory functions before and after infection,and other postinfection symptoms,as well as the duration and improvement status of olfactory and gustatory disorders.Results:Complete questionnaires were obtained from 35,566 subjects.The overall incidence of olfactory and taste dysfunction was 67.75%.Being female or being a cigarette smoker increased the likelihood of developing olfactory and taste dysfunction.Having received four doses of the vaccine or having good oral health or being a alcohol drinker decreased the risk of such dysfunction.Before infection,the average olfactory and taste VAS scores were 8.41 and 8.51,respectively;after infection,they decreased to 3.69 and 4.29 and recovered to 5.83 and 6.55 by the time of the survey.The median duration of dysosmia and dysgeusia was 15 and 12 days,respectively,with 0.5%of patients having symptoms lasting for more than 28 days.The overall self-reported improvement rate was 59.16%.Recovery was higher in males,never smokers,those who received two or three vaccine doses,and those that had never experienced dental health issues,or chronic accompanying symptoms.Conclusions:The incidence of dysosmia and dysgeusia following infection with the SARS-CoV-2 virus is high in China.Incidence and prognosis are influenced by several factors,including sex,SARS-CoV-2 vaccination,history of head-facial trauma,nasal and oral health status,smoking and drinking history,and the persistence of accompanying symptoms.

YAG (Y_(3)Al_(5)O_(12)) transparent ceramics have attractive application prospects for transparent armor protection modules because of their excellent light transmittance and anti-ballistic capability. Understanding the fracture behavior and damage mechanism of YAG is necessary for armor design. To explore the damage characteristics of YAG under compression and tension, shock compression and shockless spalling experiments with soft recovery technique are conducted. The spall strength of YAG is obtained and the recovered samples are observed by CT and SEM. It is shown that the macroscopic damage characteristic of YAG under compression is vertical split cracks with oblique fine cracks distributed in the entire sample, while that under tension is horizontal transgranular cracks concentrated near the main spall surface. The cracks generated by macroscopic compression, tension and shear stress extend in similar tensile form at the microscale. The proportion of transgranular fractures on spall surfaces is higher than that of cracks induced by macroscopic compression. Meanwhile, higher loading rate and longer loading duration increase the transgranular fracture percentage.

Plant photosynthesis assimilates CO_(2)from the atmosphere,and CO_(2)diffusion efficiency is mainly constrained by stomatal and mesophyll resistance.The stomatal and mesophyll conductance of plants are sensitive to abiotic stress factors,which affect the CO_(2)concentrations at carboxylation sites to control photosynthetic rates.Early studies conducted relevant reviews on the responses of stomatal conductance to the environment and the limitations of mesophyll conductance by internal structure and biochemical factors.However,reviews on the abiotic stress factors that systematically regulate plant CO_(2)diffusion are rare.Therefore,in this review,the rapid and long-term responses of stomatal and mesophyll conductance to abiotic stress factors(such as light intensity,drought,CO_(2)concentration and temperature)and their physiological mechanisms are summarized.Finally,future research trends are also investigated.
