Effect of Different Bacterial-Feeding Nematode Species on Soil Bacterial Numbers,Activity,and Community Composition

The effects of bacterial-feeding nematodes on bacterial number, activity, and community composition were studied through a microcosm experiment using sterilized soil inoculated with soil bacteria （soil suspension） and with bacteria and three species of bacterial-feeding nematodes （ Cephalobus persegnis, Protorhabditis filiformis, and Caenorhabditis elegans）. Catalyzed reporter deposition-fluorescence in situ hybridization, CO2 evolution, and denaturing gradient gel electrophoresis （DGGE） of PCR ampli- fied 16S rRNA gene fragments were used to investigate bacterial numbers, antivity, and community composition, respectively. Our results showed that bacterial numbers and activity significantly increased in the presence of bacterial-feeding nematodes, which indicated that bacterial-feeding nematodes had a significant positive effect on soil bacteria. The different nematode species had different effects on bacterial numbers and activity. C. persegnis and P. filiformis, isolated from native soil, increased the bacterial number and activity more than C. elegans. The DGGE analysis results showed that dominant bacterial species significantly differed among the treatments, which suggested that bacterial-feeding nematode species modified the bacterial community composition in soil. Further gene sequence analysis results showed that the dominant bacterial species in this study were gram-negative bacteria. Given the completely same conditions except nematode species, the varied selective feeding behavior of different nematode species was the most likely reason for the altered bacterial community composition. Overall, the alteration of bacterial numbers, activity and community composition resulting from the bacterial-feeding nematodes may ult!mately affect soil ecological functioning and processes.

目的综合运用生物信息学方法及机器学习算法筛选与非酒精性脂肪性肝炎相关的趋化因子核心基因。方法公共数据库GEO下载非酒精性脂肪性肝病芯片数据集GSE49541,采用R studio软件进行差异分析筛选差异基因,对差异基因进行GO功能注释和KEGG信号通路富集分析,将差异基因与趋化因子通路相关基因集取交集获取趋化因子相关差异基因,然后采用机器学习LASSO回归及SVM-RFE算法筛选核心基因,通过Genemania数据库构建核心基因互作网络图,构建核心基因列线图预测模型,并通过ROC曲线验证列线图效能。结果共筛选获取差异基因148个,GO及KEGG富集分析提示差异基因富集于脂质代谢、趋化因子、细胞外基质等。最后筛选获得核心基因CCL19、CD24、ROBO1、SLC12A2,构建核心基因互作网络图,基于核心基因建立NASH列线图预测模型,该模型ROC曲线的AUC=0.997,95%置信区间(confidence interval,CI)为0.988~1.000。结论CCL19、CD24、ROBO1、SLC12A2可能与非酒精性脂肪性肝炎发生与进展密切相关,有望成为诊断和精准治疗的潜在靶点。

Combining the inhibited aptazyme and molecular beacon （MB）, we developed a versatile sensing strategy for amplified detection of adenosine. In this strategy, the adenosine aptamer links to the 8-17 DNAzyme to form an aptazyme. A short sequence, denoted as inhibitor, is designed to form a duplex spanning the aptamer-DNAzyme junction, which blocks the catalytic function of the DNAzyme. Only in the presence of target adenosine, the aptamer binds to adenosine, thus the inhibitor dissociates from the aptamer portion of the aptazyme and can no longer form the stable duplex required to inhibit the catalytic activity of the aptazyme. The released DNAzyme domain will hybridize to the MB and catalyze the cleavage in the presence of Zn2＋, making the fluorophore separate from the quencher and resulting in fluorescence signal. The results showed that the detection method has a dynamic range from 10 nmol/L to 1nmol/L, with a detection limit of 10 nmol/L.

Background Frozen-thawed embryo transfer （FET） is the most common way to prevent serious late ovarian hyperstimulation syndrome and increase the cumulative pregnancy rate. We evaluated the effectiveness of an FET program for improving the embryo implantation and clinical pregnancy rates, and ultimate embryo utilization rate in infertility treatment. Methods Patients undergoing in vitro fertilisation （IVF） cycles from January 2006 to June 2008 were enrolled, including 179 patients who had undergone the first FET cycle after controlled ovarian hyperstimulation （COH） in which all embryos were frozen （group C1） and 1306 patients who had COH with fresh embryo transfer （ET）（group T1）. Logistic regression was used to model the embryo implantation and clinical pregnancy rates based on the mother＇s age, numbers of oocytes retrieved, embryos transferred and high-quality embryos transferred. The embryo implantation and clinical pregnancy rates were also compared between two groups after adjusting for age, the numbers of oocytes retrieved and the numbers of embryos transferred. Results Logistic regression analysis confirmed that embryo implantation and clinical pregnancy rates in group C1 were both significantly higher than those in group T1 after adjusting for confounding factors （43.6% vs 29.0%, 63.1% vs 47.0%, respectively; P 〈0.01）. The embryo implantation and clinical pregnancy rates were consistently higher in group C1 by comparing the age groups ≥35 or 〈35 years. The clinical pregnancy rates for the numbers of oocytes retrieved per cycle being ≥15 or 〈15 were higher in group C1, as was the embryo implantation rate. These differences were statistically significant for oocyte numbers 〉15 （P 〈0.05）. The embryo implantation and clinical pregnancy rates in group C1 were both significantly higher than in group T1 when two or three embryos were transferred （P 〈0.05）. Heat shock proteins （Hsps） have been linked to stresses and winter diapause in insects, but whether they are components of summer diapause is still unknown. In this study, complementary DNAs of lisp90 from Pieris melete, Pieris rapae and Pieris canidia named PmHsp90, PrHsp90 and PcHsp90, respectively, were cloned and sequenced. The deduced amino acid sequence consisted of 718 amino acid residues with a putative molecular mass of 82.6, 82.6 and 82.7 kDa, respectively. The amino acid sequences contained all of the five conserved signature motifs in the Hsp90 family and a bHLH protein folding activity region. The differential expression pattern of PmHsp90 in response to summer diapause and winter diapause, which are related to heat/cold stress, was investigated. Cold stress induced Hsp90 up-regulation in summer and winter diapause pupae, but not in non-diapause individuals. Heat shock up-regulated PmHsp90 gradually with an increase in temperature in summer diapause, and PmHsp90 was rapidly up-regulated in winter diapause. After 30 min heat shock at 39℃, substantial up-regulation of PmHsp90 transcript levels were observed both in summer and winter diapause. However, in non-diapause a relatively stable expression was found under different durations of 39℃ heat shock. Compared to the optimal treatment of 18℃ for diapause development, a high temperature acclimation of 31 ℃ induced PmHsp90 up-regulation in summer diapause, whereas a low temperature acclimation of 4℃ induced up-regulation in winter diapause. The current results indicate that Hsp90 may play an important role in response to heat/cold stress both in summer and winter diapause.

ICU患者口渴影响因素的Meta分析

目的总结ICU患者并发口渴的影响因素，为临床医护人员及早识别口渴风险人群，以便为进行及时干预提供循证医学的依据。方法计算机检索美国国立医学图书馆PubMed数据库、Cochrane图书馆数据库、科学网（Web of Science）、荷兰医学文摘EMbase数据库、中国生物医学文献服务系统（SinoMed）、中国知网（CNKI）、维普中文科技期刊数据库（VIP）和万方数据库，收集自建库至2023年1月有关ICU患者并发口渴的研究，由2名研究员对文献进行资料提取和质量评价，使用RevMan5.3软件进行Meta分析。结果共纳入8篇文献，涉及1561例ICU患者，提取33项危险因素。Meta分析结果显示，饮酒史[OR=3.31,95%CI(1.99,5.51),P<0.000 01]、吸烟史[OR=2.79,95%CI(2.12,3.65),P<0.0001]、利尿剂[OR=4.72,95%CI(2.16,10.34),P=0.0001]、呼吸频率[OR=3.59,95%CI(1.61,8.01),P=0.002]、是否首次使用机械通气[OR=1.12,95%CI(1.03,1.52),P=0.005]、面罩舒适度[OR=3.5,95%CI(1.58,7.74),P=0.002]是ICU患者并发口渴的危险因素。结论 ICU患者发生口渴与饮酒史、吸烟史、利尿剂、呼吸频率、是否首次使用机械通气、面罩舒适度多种因素相关，应尽早识别相关危险因素，采取预防措施降低口渴发生及其严重程度。

A novel series of first procaspase activating compound（PAC-1） analogues was designed, synthesized and evaluated for antitumor activity towards two cell lines[human promyelocytic leukemia cell line（HL60） and human embryonic lung fibroblast cell line（HLF）] by the MTT[3-（4,5）-dimethylthiahiazo（-z-yl）-3,5-di-phenytetrazo- liumromide] method in vitro. The structures of all the compounds were confirmed by IH NMR, MS and elemental analysis. Among the compounds synthesized^（E）-2-[（3-{[4-（tert-buty~）benzy~]（methy~）amin~}pr^py~）（methy~）amin~]- N＇-[4-（diethylamino）-2-hydroxybenzylidene]acetohydrazide（compound 6n） exhibits a good anti-proliferative activity to the majority of tumor cells tested, and selectively cleaves cancer cells. Thus, compound 6n was identified as promising lead compound for further structural modification.

[目的]通过分析西双版纳热带雨林不同生长型木本植物功能性状的差异,揭示热带雨林不同生长型木本植物功能多样性的变化规律以及影响因素。[方法]本文基于西双版纳国家级自然保护区勐仑、补蚌和尚勇等片区3块1 hm2动态监测样地的调查数据和木本植物功能性状数据,利用单因素方差分析、主成分分析、Pearson相关分析、冗余分析以及多元线性回归分析等方法,分析西双版纳热带雨林不同生长型木本植物功能多样性的差异,阐明热带雨林木本植物群落的功能特征及主要影响因子。[结果](1)乔木的叶面积、叶碳含量、叶干物质含量的群落加权平均值均显著高于灌木和藤本,而藤本叶氮含量和比叶面积的群落加权平均值显著高于乔木和灌木。(2)乔木功能丰富度和功能离散度高于灌木和藤本,而灌木功能均匀度和功能分异度均高于乔木和藤本。(3)乔木和藤本功能丰富度以及藤本功能离散度随物种丰富度、多度和系统发育多样性的增加而显著增大;乔木功能均匀度和功能分异度随土壤肥力的增大而显著增大,而灌木功能离散度随物种丰富度的增加而增大。(4)植物多度是影响乔木和灌木功能性状群落加权平均值的主要因素,而系统发育多样性、物种丰富度和土壤肥力是影响藤本功能性状群落加权平均值的主要因素。[结论]表明西双版纳热带雨林不同生长型木本植物群落在功能性状和功能多样性之间存在显著差异性,且植物多度和土壤肥力在热带雨林植物群落功能多样性维持机制中起到重要作用。

Beta-cell dysfunction is the primary contributor to the early postpartum diabetes among Chinese women with history of gestational diabetes mellitus

Background Women with a history of gestational diabetes mellitus （GDM） are at higher risk of future development of diabetes. This study investigated the risk factors associated with early postpartum abnormal glucose regulation （AGR） among Chinese women with a history of GDM. Methods A total of 186 women with a history of GDM were screened for early postpartum AGR at 6-8 weeks after delivery. Those with AGR were given lifestyle intervention therapy and reevaluated in 6-12 months. The demographic, anthropometric, prenatal and delivery data were recorded. The plasma high-sensitivity C-reactive protein （HsCRP） and lipid concentration were measured, and insulin secretion were analyzed. Insulinogenic index △ins30＇/△BG30＇, the homeostasis model assessment index （HOMA）-B, and HOMA-IR were calculated. Multiple regression analysis was performed to identify the risk factors. Results Of the GDM women 28.0% （52/186） had AGR at 6-8 weeks after delivery; 45.2% （17/40） of these AGR women reminded abnormal after 6-12 month lifestyle intervention. Compared to the women who reverted to normal, women with consistent AGR showed significantly lower fasting insulin concentration, lower △ins30＇/△BG30＇ as well as lower HOMA-B. No significant differences in age, body mass index （BMI）, waist circumference, blood pressure, lipid level HsCRP and HOMA-IR were observed between the two groups. Pre-pregnancy BMI ≥25 kg/m^2, fasting glucose level ≥5.6 mmol/L and/or 75 g oral glucose tolerance test （OGTT） 2 hours glucose level ≥11.1 mmol/L during pregnancy were predictors for the AGR at 6-8 weeks after delivery. △ins30＇/△BG30≤1.05 was a significant risk contributor to the consistent early postpartum AGR. Conclusion There is a high incidence of early postpartum AGR among Chinese woman with prior GDM. Beta-cell dysfunction, rather than insulin resistance or inflammation, is the predominant contributor to the early onset and consistent AGR after delivery.
