The relationship between controllability, optimal testing resource allocation, and incubation-latent period mismatch as revealed by COVID-19

The severe shortfall in testing supplies during the initial COVID-19 outbreak and ensuing struggle to manage the pandemic have affirmed the critical importance of optimal supplyconstrained resource allocation strategies for controlling novel disease epidemics.To address the challenge of constrained resource optimization for managing diseases with complications like pre-and asymptomatic transmission,we develop an integro partial differential equation compartmental disease model which incorporates realistic latent,incubation,and infectious period distributions along with limited testing supplies for identifying and quarantining infected individuals.Our model overcomes the limitations of typical ordinary differential equation compartmental models by decoupling symptom status from model compartments to allow a more realistic representation of symptom onset and presymptomatic transmission.To analyze the influence of these realistic features on disease controllability,we find optimal strategies for reducing total infection sizes that allocate limited testing resources between‘clinical’testing,which targets symptomatic individuals,and‘non-clinical’testing,which targets non-symptomatic individuals.We apply our model not only to the original,delta,and omicron COVID-19 variants,but also to generically parameterized disease systems with varying mismatches between latent and incubation period distributions,which permit varying degrees of presymptomatic transmission or symptom onset before infectiousness.We find that factors that decrease controllability generally call for reduced levels of non-clinical testing in optimal strategies,while the relationship between incubation-latent mismatch,controllability,and optimal strategies is complicated.In particular,though greater degrees of presymptomatic transmission reduce disease controllability,they may increase or decrease the role of nonclinical testing in optimal strategies depending on other disease factors like transmissibility and latent period length.Importantly,our model a

We apply a zircon redox index to a global compilation of detrital zircons to track the variation of oxidation state,expressed asΔfMQ,through Earth's history.Those from I-type rocks,which comprise mantle and crustal igneous protoliths,including tonalite–trondhjemite–granodiorites(TTGs),generally have a high oxidation state(ΔfMQ>0).In contrast,zircons from igneous rocks derived from supracrustal source rocks(S-type)are commonly reduced(ΔfMQfunction derived from the Gaussian-Kernel-Density-Estimation,we use the maximum likelihood estimation(MLE)to distinguish Stype from I-type zircons through Earth's history using zircon redox.Voluminous S-type magma production shows a ca.600 Ma cyclicity that is closely related to the supercontinent cycle.We link a cyclic drop in redox values after 2.6 Ga to periodic S-type magma generation associated with burial and melting of metasedimentary rocks during supercontinent assembly and amalgamation.TheΔfMQ of the detrital zircons rise at~3.5 Ga followed by a consistent averageΔfMQ>0 over the last 3 Ga.Given that the redox state of magmas is independent of crustal thickness and silica variation,and elevated values are likely more closely related to tectonic setting,we suggest that the consistent averageΔfMQ>0 from ca.3.5 Ga onwards relates to recycling of oceanic lithosphere back into the mantle in what eventually became established as subduction zones.The more reduced magmas associated with sedimentary sources,became established at 2.6 Ga,presumably in response to continental rocks rising above sealevel,and follow peaks of productivity associated with the supercontinent cycle.

Chicken is an important food animal worldwide and plays an important role in human life by providing meat and eggs.Despite recent significant advances in gut microbiome studies,a comprehensive study of chicken gut bacterial,archaeal,and viral genomes remains unavailable.In this study,we constructed a chicken multi-kingdom microbiome catalog(CMKMC),including 18,201 bacterial,225 archaeal,and 33,411 viral genomes,and annotated over 6,076,006 protein-coding genes by integrating 135 chicken gut metagenomes and publicly available metagenome-assembled genomes(MAGs)from ten countries.We found that 812 and 240 MAGs in our dataset were putative novel species and genera,respectively,far beyond what was previously reported.The newly unclassified MAGs were predominant in Phyla firmicutes_A(n=263),followed by firmicutes(n=126),Bacteroidota(n=121),and Proteobacteria(n=87).Most of the classified species-level viral oper-ational taxonomic units belong to Caudovirales.Approximately,63.24%of chicken gut viromes are predicted to infect two or more hosts,including complete circular viruses.Moreover,we found that diverse auxiliary metabolic genes and antibiotic resistance genes were carried by viruses.Together,our CMKMC provides the largest integrated MAGs and viral genomes from the chicken gut to date,functional insights into the chicken gastrointestinal tract microbiota,and paves the way for microbial interventions for better chicken health and productivity.

Use of animal models in preclinical transplant research is essential to the optimization of human allografts for clinical transplantation.Animal models of organ donation and preservation help to advance and improve technical elements of solid organ recovery and facilitate research of ischemia-reperfusion injury,organ preservation strategies,and future donor-based interventions.Important considerations include cost,public opinion regarding the conduct of animal research,translational value,and relevance of the animal model for clinical practice.We present an overview of two porcine models of organ donation:donation following brain death(DBD)and donation following circulatory death(DCD).The cardiovascular anatomy and physiology of pigs closely resembles those of humans,making this species the most appropriate for pre-clinical research.Pigs are also considered a potential source of organs for human heart and kidney xenotransplantation.It is imperative to minimize animal loss during procedures that are surgically complex.We present our experience with these models and describe in detail the use cases,procedural approach,challenges,alternatives,and limitations of each model.

Antibacteria Activity of Peganum harmala and Haloxylon salicornicum Leaves Extracts

The use of antibiotics in humans and animals has been marked as a significant step in health due to their effectiveness in controlling and treating bacterial infections. The misuse and overuse of antibiotics have been identified as risk factors for bacterial resistance since microorganisms adapt and develop mechanisms to defend against antibiotics. According to the Centers for Disease Protection and Control (CDC), around 23,000 individuals die every year in the United States due to antibiotic resistance complications. As a result, a demand for alternative treatments has been a goal for scientists as the microbes adapt to selective pressure. The aim of this study is to test the antibacterial activity of leaf extracts of Peganum harmala and Haloxylon salicornicum on both Gram-positive and Gram-negative bacteria on various mediums. The results of the study showed that both P. harmala and H. salicornicum inhibited the bacterial growth in two different media. The results were also compared with different common antibiotics used in both human’s and animal’s fields and showed a promising outcome as alternative antibiotics.

Stromal cell decidualization and embryo implantation: a vulnerable step leading to successful pregnancy

Endometrial stromal cell decidualization is a crucial step in endometrial remodeling during pregnancy. Decidualization is controlled by orchestrated ovarian hormones, followed by the activation of various downstream signaling pathways. Accumulating evidence has shown multiple functions of decidualized endometrial stromal cells during embryo implantation, including tissue remodeling, antioxidative stress, angiogenesis, and immune tolerance. The distinct secretomes of decidualized stromal cells also reveal their intensive interactions with epithelial, endothelial, and immune cells. However, aberrant decidualization leads to pregnancy failures, such as recurrent pregnancy loss and repeated implantation failure. This review aimed to provide an overview of the molecular mechanisms underlying the divergent functions of decidualized endometrial stromal cells and their potential clinical applications. Moreover, the use of single-cell RNA sequencing data further enhances our understanding of these biological processes. This review discusses decidualization-related signaling pathways that serve as potential therapeutic targets for treating implantation failure inin vitro fertilization and provides novel approaches to investigate the underlying causes of female infertility.

CDC42 controls intestinal epithelial(IEC)stem cell(IESC)division.How aberrant CDC42 initiates intestinal inflammation or neoplasia is unclear.We utilized models of inflam-matory bowel diseases(IBD),colorectal cancer,aging,and IESC injury to determine the loss of intestinal Cdc42 upon inflammation and neoplasia.Intestinal specimens were collected to determine the levels of CDC42 in IBD or colorectal cancer.Cdc42 floxed mice were crossed with Villin-Cre,Villin-CreERT2 and/or Lgr5-eGfP-IRES-CreERT2,or Bmi1-CreERT2 mice to generate Cdc42 deficient mice.Irradiation,colitis,aging,and intestinal organoid were used to evaluate CDC42 upon mucosal inflammation,IESC/progenitor regenerative capacity,and IEC repair.Our studies revealed that increased CDC42 in colorectal cancer correlated with lower survival;in contrast,lower levels of CDC42 were found in the inflamed IBD colon.Colonic Cdc42 depletion significantly reduced Lgr5+IEsCs,increased progenitors'hyperplasia,and induced mucosal inflammation,which led to crypt dysplasia.Colonic Cdc42 depletion markedly enhanced irra-diation-or chemical-induced colitis.Depletion or inhibition of Cdc42 reduced colonic Lgr5+IESC regeneration.In conclusion,depletion of Cdc42 reduces the IESC regeneration and IEC repair,leading to prolonged mucosal inflammation.Constitutive monogenic loss of Cdc42 in-duces mucosal inflammation,which could result in intestinal neoplasia in the context of aging.

World Class Elite Breaststrokers Downward Kick EMG Muscle Activity of the Gluteus Maximus, Quadriceps femoris, Hamstring, Gastrocnemius Medialis, Rectus Abdominal, and Erector Spinae

This study analyzed the difference between using a downward breaststroke kick and a horizontal breaststroke kick in a sample of world class elite swimmers.We compared average muscle activity of the gluteus maximus,quadriceps femoris(vastus medialis and rectus femoris),hamstring/long head of the biceps femoris,gastrocnemius medialis,rectus abdominal,and erector spinae when using the downward breaststroke kick technique.We find that when this sample of swimmers utilized the downward breaststroke kick,max speed and velocity per stroke increased,measured by 12,788 EMG samples,where the results are highly correlated to duration of the aerodynamic buoyant force in breaststroke kick technique.The increases in performance observed from measuring the world class elite swimmers is highly correlated to the duration of the kick aerodynamic buoyant force.Among this sample of elite swimmers,the longer a swimmer demonstrates a buoyant force breaststroke kick,the lower the time in a 100 breaststroke.

Prevalence of Work-Aggravated Asthma among Bakery Workers in Abidjan (Cote d’Ivoire)

Introduction: Work-aggravated asthma is pre-existing or concomitant asthma whose symptoms are aggravated by the work environment. The aim of this study was to determine the prevalence of this pathology and its associated factors among bakery workers in Abidjan. Materials and Methods: This descriptive and analytical cross-sectional study was conducted over a period of four (4) months from 18 December 2019 to 18 April 2020. Two questionnaires were used, one on employees and the other on the indoor environment of bakeries. In addition, a spirometry test was performed on all bakery workers. Statistical analysis was performed using stata 15.1 software. Results: A total of 599 bakery employees, including bakers (59.73%), sales assistants/ cashiers (23.52%), cleaners (6.34%) and administrative staff (10.18%), were investigated. The mean age was 30.8 ± 8 years, with a sex ratio (M/f) = 2.2. Asthma symptoms were found in 95 (15.86%) employees, of whom 74 (77.9%) had work-related asthma and 11 (14.9%) had asthma aggravated by work. The factors associated with work-aggravated asthma were personal or family history of allergy or atopy [ORa = 3.75;CI95%: 1.56 - 8.93;p = 0.003], exposure to dust [ORa = 5.01;CI95%: 1.43 - 7.50;p = 0.011] and humidity level (60% - 70%) [ORa = 1.80;CI 95%: 0.99 - 3.28;p = 0.05]. Conclusion: Work-aggravated asthma is a reality in bakeries in Abidjan, with an estimated prevalence of 14.9%. Two of the three factors associated with this condition suggest a link with indoor air pollution. Combating air pollution in these establishments must therefore be a priority for the relevant authorities, in order to provide employees with a working environment that protects their health.

An update on hormonal regulation of early embryo-fallopian tube interactions: insights for improvements of assisted reproductive technology outcomes

Human fallopian tubes (fTs) play important roles in the whole reproductive process. fTs are the site for the final maturation of gametes, fertilization, transport of preimplantation embryos, and early embryonic development before entering the uterus. All these functions are critical in the establishment of a successful pregnancy. When the embryo is in transit through the fT, the fT microenvironment changes through endocrine, paracrine, autocrine, and juxtacrine mechanisms which involve various hormones and bioactive substances mainly from the neighboring cells. Therefore, the interactions between the embryo and the fT may be the earliest embryo-maternal communications that are essential for the establishment of pregnancy. This review critically discusses the evidence of various hormonal and bioactive mediated control of the microenvironment in the fTs during the earliest embryo-maternal communications. Moreover, we highlight existing gaps in the knowledge and the importance of exploring more of the physiology of fTs which will hold the key to producing high-quality embryos for patients seekingin vitro fertilization (IVf) treatments.
