The protocadherin alpha cluster is required for axon extension and myelination in the developing central nervous system

In adult mammals, axon regeneration after central nervous system injury is very poor, resulting in persistent functional loss. Enhancing the ability of axonal outgrowth may be a potential treatment strategy because mature neurons of the adult central nervous system may retain the intrinsic ability to regrow axons after injury. The protocadherin （Pcdh） clusters are thought to function in neuronal morphogenesis and in the assembly of neural circuitry in the brain. We cultured primary hippocampal neurons from E17.5 Pcdhα deletion （del-α） mouse embryos. After culture for 1 day, axon length was obviously shorter in del-α neurons compared with wild-type neurons. RNA sequencing of hippocampal E17.5 RNA showed that expression levels of BDNF, Fmod, Nrp2, OGN, and Sema3d, which are associated with axon extension, were significantly down-regulated in the absence of the Pcdhα gene cluster. Using transmission electron microscopy, the ratio of myelinated nerve fibers in the axons of del-α hippocampal neurons was significantly decreased; myelin sheaths of P21 Pcdhα-del mice showed lamellar disorder, discrete appearance, and vacuoles. These results indicate that the Pcdhα cluster can promote the growth and myelination of axons in the neurodevelopmental stage.

A practical synthesis of symmetrical vicinal diamines with broad chemistry scope is described. The key step is the ultrasonicwave promoted tandem-reaction, two-step reaction sequence combining Mannich double condensations with benzo- triazole, glyoxal and primary or secondary amines and reduction of the bis（benzotriazole）-adducts with sodium borohydride proceeded smoothly in tetrahydrofuran in high yield at room temperature.

Variable 2-Microlocal Besov–Triebel–Lizorkin-Type Spaces

This article is devoted to the study of variable 2-microlocal Besov-type and Triebel- Lizorkin-type spaces. These variable function spaces are defined via a Fourier-analytical approach. The authors then characterize these spaces by means of Q-transforms, Peetre maximal functions, smooth atoms, ball means of differences and approximations by analytic functions. As applications, some re- lated Sobolev-type embeddings and trace theorems of these spaces are Mso established. Moreover, some obtained results, such as characterizations via approximations by analytic functions, are new even for the classical variable Besov and Triebel-Lizorkin spaces.

A Truncated Arabidopsis NUCLEOSOME ASSEMBLY PROTEIN 1, AtNAP1;3T, Alters Plant Growth Responses to Abscisic Acid and Salt in the Atnap 1;3-2 Mutant

Chromatin remodeling is thought to have crucial roles in plant adaptive response to environmental stimulus. Here, we report that, in Arabidopsis, the evolutionarily conserved histone chaperone, NUCLEOSOME ASSEMBLY PROTEIN 1 （NAP1）, is involved in plant response to abscisic acid （ABA）, a phytohormone important in stress adaptation. We show that simultaneous loss-of-function of AtNAP1;1, AtNAP1;20 and AtNAP1;3 （the triple mutant m123-1） caused a slight hypersensitive response to ABA in seedling growth. Strikingly, the other triple mutant m123-2 containing a different mutant allele of AtNAP1;3, the Atnap 1;3-2 allele, showed a hyposensitive response to ABA and a decreased tolerance to salt stress. This ABA- hyposensitive and salt response phenotype specifically associated with the Atnapl;3-2 mutant allele. We show that this mutant allele produced a truncated protein, AtNAP1;3T, which lacks 34 amino acids at the C-terminus compared to the wild-type protein AtNAP1;3. We further show that the heterozygous plants containing the Atnapl;3-2 mutant allele as well as transgenic plants overexpressing AtNAP1;3Texhibit ABA-hyposensitive phenotype. It thus indicates that AtNAP1;3T functions as a dominant negative factor in ABA response. The expression of some ABA-responsive genes, including genes encoding protein kinases and transcription regulators, was found perturbed in the mutant and in theAtNAP1;3Ttransgenic plants. Taken together, our study uncovered AtNAP1 proteins as positive regulators and AtNAP1;3Tas a negative regulator in ABA signaling pathways, providing a novel link of chromatin remodeling to hormonal and stress responses.

Expression of the retinoic acid-metabolizing enzymes RALDH2 and CYP26b1 during mouse postnatal testis development

Aim： To study the expression pattern of the retinoic acid metabolizing enzymes RALDH2 and CYP26bl during mouse postnatal testis development at both mRNA and protein levels. Methods： Real-time polymerase chain reaction and Western blot analysis were performed to determine the relative quantity of RALDH2 and CYP26bl at both mRNA and protein levels at postnatal day 1, 5, 10, 20, and in adult mice （70 days testes）. Testicular localization of RALDH2 and CYP26b 1 during mouse postnatal development was examined using immunohistochemistry assay. Results： Aldhla2 transcripts and its protein RALDH2 began to increase at postnatal day 10, and remained at a high level through postnatal day 20 to adulthood. Cyp2661 transcripts and CYP26bl protein did not change significantly during mouse postnatal testis development. RALDH2 was undetectable in the postnatal 1, 5 and 10 day testes using immunohis- tochemistry assay. At postnatal day 20 it was detected in pachytene spermatocytes. Robust expression of RALDH2 was restricted in round spermatids in the adult mouse testis. In the developing and adult testis, CYP26bl protein was confined to the peritubular myoepithelial cells. Conclusion： Our results indicate that following birth, the level of retinoic acid in the seminiferous tubules might begin to increase at postnatal day 10, and maintain a high level through postnatal day 20 to adulthood.

Formation mechanism of interface reaction layer between microporous magnesia refractories and molten steel and its effect on steel cleanliness

The ceramic filter in continuous casting tundish can effectively improve the cleanliness of high-performance steel by regulating tundish flow field to promote the removal of inclusions and adsorbing or blocking fine inclusions in the molten steel into the *** interaction between microporous magnesia refractories used as tundish filter and molten interstitial-free(IF)steel at 1873 K was investigated to reveal the formation mechanism of their interface layer and its effect on steel cleanliness by laboratory research and thermodynamic *** results show that the magnesium–aluminum spinel layer at the interface between the molten IF steel and the microporous magnesia refractories is formed mainly by the reaction of MgO in the refractory with the[Al]and[O]in the molten steel,significantly reducing the total O content,the size and amount of inclusions of the molten *** addition,the interparticle phases of microporous magnesia refractories at high temperature can adsorb Al_(2)O_(3) and TiO_(2) inclusions in the molten steel into interparticle channels of the refractories to form high melting point spinel,impeding the further penetration of the molten *** a result,the consecutive interface layer of high melting point spinel between microporous magnesia refractories and molten steel can improve the cleanliness of the molten steel by adsorbing inclusions in the molten steel and avoid the direct dissolution of refractories of the tundish ceramic filter immersed in the molten steel,increasing their service life.
