Separation of growth-stimulating peptides for Bifidobacterium from soybean conglycinin

AIM： To isolate and identify the soybean conglycinin peptides that selectively stimulates the growth of bifidobacteria in vitro, and to investigate the effect of soybean conglycinin peptides on intestinal ecosystem in vivo. METHODS： Soybean conglycinin was purified from soybean seeds by gel filtration （Sepharose-CL-6B）. These proteins were submitted to hydrolysis by pepsin. Several growth-stimulating peptides for bifidobacteria were isolated chromatographically from pepsin hydrolysis of soybean conglycinin and identified by means of matrixassisted laser desorption ionization time of flight mass spectrometry （MALDI-TOF-MS）. Parallel to in vitro study, in vivo experiments with soybean conglycinin peptides were performed in mice. Ninety male KM mice were randomly assigned into five groups of 16 mice each, and each group was administered for 21d intragastrically with physiological saline （control）, conglycinin, pepsin-treated conglycinin （PTC）, the most active fraction which isolated from pepsin-treated conglycinin （P2-PTC） and HCl-full hydrolysis of conglycinin （HCl-FHC）, respectively. Intestinal microflora were evaluated by standard microbiologic methods and biochemical assays of cecal content samples after treatment. RESULTS： The results showed that the peptides which were isolated from soybean conglycinin could stimulate the growth of bifidobacteria in vitro, and the molecular mass of purified peptides with MALDI-TOF-MS ranged from 693.32 to 1829.55. Compared with control group, in vivo experiments showed that P2-PTC group decreased cecal pH （7.08±0.08 vs7.21±0.09, P〈0.05） and enterococcicounts （5.38±0.26 log10CFU/g vs 5.78±0.19 log10CFU/g, P〈0.05）, significantly increased sigA level （172.08±35.40 ng/g vs 118.27±33.93 ng/g, P〈0.01） and β-galactosidase activity （1.28±0.23 U/g vs 1.82±0.58 U/g, P〈0.05） CONCLUsiON： The results have shown that conglycinin is good source for enzyme-mediated production of peptides which stimulate the growth of bifidobacteria. 90只清洁级昆明系小鼠,随机分为5组,预饲基础日粮一周后,各组分别灌胃:生理盐水(对照组,CK)、伴大豆球蛋白液(Ⅰ)、伴大豆球蛋白胃蛋白酶水解液(Ⅱ)、伴大豆球蛋白胃蛋白酶水解液提纯B组分(Ⅲ)、伴大豆球蛋白盐酸水解液(Ⅳ),除对照组外,各处理组灌胃液体总含氮量相等,实验期21d。结果显示:与对照组相比,第3周Ⅱ组和Ⅲ组每只小鼠日均采食量较对照组显著下降,分别降低8.75%(PsigA水平提高45.49%(P<0.01),大肠食糜中6磷酸果糖酮糖酶(F6PPK)活性极显著提高(P<0.01),pH值下降了1.80%(P<0.05),肠球菌数显著下降(P<0.05)。实验结果表明水解肽作为腔内信号分子可能与肠道内相关因素有着多层次调控关系,共同促进宿主的健康。

【目的】在分子水平上探讨伴大豆球蛋白水解肽对肠道双歧杆菌生长的影响。【方法】选取21日龄健康清洁级雄性SD大鼠,随机分为4个处理,每个处理3只,基础日粮预饲1周后,分别灌胃生理盐水(对照组)、伴大豆球蛋白液(Conglycinin组)、伴大豆球蛋白酶水解肽P2组分(P2-PTC组)、伴大豆球蛋白盐酸彻底水解液(HCl-FHC组),各0.5ml,1次/天。除对照组外,各处理组灌胃液体总含氮量相等(折合蛋白含量为0.5mg·ml-1),试验共21d。应用PCR-DGGE技术分析灌胃伴大豆球蛋白酶解肽对大鼠粪样细菌区系变化的影响,并采用连续稀释PCR的方法半定量研究伴大豆球蛋白酶解肽对大鼠肠道双歧杆菌数量的影响。【结果】灌胃伴大豆球蛋白酶解肽的大鼠在试验后第1周,其粪样微生物区系变化迅速,而后缓慢,最终形成稳定的体系,提示酶解肽在一定程度上影响了肠道细菌的组成,与对照组和蛋白盐酸彻底水解液组比较,肠道双歧杆菌数量明显增加。【结论】本试验在分子水平证明了伴大豆球蛋白水解肽对肠道双歧杆菌的生长有促进作用。

经碱提酸沉法制备大豆蛋白,胃蛋白酶水解,茚三酮法测定水解液水解度,系统观察了灌喂1mL的5mg/mL的大豆蛋白酶解物对大鼠免疫功能的影响。结果表明:大豆蛋白水解液含氮量为(2.15±0104)mg/mL,水解度为14.49%,平均肽链长度为6.9。灌喂21d后,发现胸腺、脾脏指数较对照组分别上升了24%(PsigA水平较对照组上升了34.3%,差异极显著(P<0101);灌喂大豆蛋白酶解物能极显著提高血液IL22和Leptin水平。试验结果表明灌喂大豆蛋白酶解物能增强大鼠免疫功能,主要是酶解肽的生物活性的作用结果,并非营养调节作用的结果,酶解物对大鼠消化代谢影响不大。

1 Introduction Early detection and diagnosis of stable coronary artery disease （SCAD） is essential for proactive secondary prevention of myocardial infarction （MI）, control of disease progress, and reduction of mortality. Clinical decision-making in modem medicine is increasingly dependent on cardiovascular imaging techniques. 2012 ACCF/AHA/ACP/AATS/ PCNA/SCAI/STS guideline for the diagnosis and management of patients with stable ischemic heart disease has been issued by American Heart Association （AHA）. European Society of Cardiology （ESC） has issued 2013 ESC guidelines on the management of stable coronary artery disease.

Physiological and Biochemical Characteristics of Lactation in Half-lactating Yak

Yaks of two lactating types, the half-Iactating yak and the total lactating yak, were investigated in their milk compositions, milk coagulation properties, and contents of three kinds of hormones, glucose, and protein in plasma. The half-lactating Maiwa yak and Jiulong yak contained significantly higher contents of protein, fat, and activities of alkaline phosphatase and Y-glutamyl transpeptidase in milk than that of the corresponding total-lactating yak breeds, with reduced milk yield and similar lactose level and relative percentages of main milk protein components. The half-lactatin yak resembled yaks in a late stage of lactation in their biochemical composition of milk; however, significant differences were also observed, which indicated that the former was in a special stage of lactation quite different from dairy cows. Milk of the total-lactating or half-lactating yaks could be coagulated normally by adding chymosin, with a similar coagulation time. No significant difference was observed between prolactin and progesterone concentrations in plasma of the half-lactating yaks and total-lactating yaks; however, half-lactating yaks had significantly lower level of oestradiol-17 β in plasma than total-lactating yaks.

The Large sky Area Multi-Object Fiber Spectroscopic Telescope（LAMOST） general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio（SNR） ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters： late A,FGK-type stars with high quality spectra（1 061 918 entries）, A-type stars（100 073 entries）, and M-type stars（121 522 entries）. This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. A description of the FITS structure of spectral files and parameter catalogs is also provided.

This paper describes the data release of the LAMOST pilot survey, which includes data reduction, calibration, spectral analysis, data products and data access. The accuracy of the released data and the information about the FITS headers of spectra are also introduced. The released data set includes 319 000 spectra and a catalog of these objects.

INTRODUCTION Sepsis is defined as an infection associated with systemic manifestations of inflammation, which has become the most common complication in the perioperative period caused by severe burn/trauma and major surgical operation.[1,2] The incidence of sepsis in adults is estimated to be 149–240/100 000 per year, and that of severe sepsis and septic shock is 56–91/100 000 per year. In the last decade, the short-term mortality of sepsis has declined to around 20% in developed countries partly due to the international Surviving Sepsis Campaign （SSC）, while the mortality remains very high （50%–80%） in 1–5 years after discharge from hospitals.[3–7] Thus, sepsis and its subsequent severe sepsis and septic shock are currently major issues in the field of medical and health care.
