Phototoxic effect of UVR on wild type, ebony and yellow mutants of Drosophila melanogaster: Life Span, fertility, courtship and biochemical aspects

Melanin plays an important role in protecting organisms from ultraviolet radiation (UVR). Therefore, it is possible that differently colored strains can show different sensitivities to UVR. In the present work, life span, fertility and courtship behavior of wild type (w), ebony (e) and yellow (y) strains of Drosophila melanogaster were studied to evaluate their sensitivity to ultraviolet (UV). Because a range of photo- toxic effects of UVR are mediated through generation of free radicals, levels of free radicals, lipid per- oxide (malondialdehyde, MDA) and superoxide dismutase (SOD) activity of three strains were examined to indicate their antioxidant defending ability and oxidative status. It was shown that w always had the highest lifespan and fertility not only in the control but also in UV-exposed groups. Moreover, lifespan and fertility of e were significantly higher (P2) effect on the SOD activities. After pooling data from the control and UV-exposed groups, we found that w had a significantly (P0.05). MDA levels were increased in the UV dose-dependent manner (P=0.0495). In con- clusion, our results suggested that UVR can decrease life span and fertility of flies and do harm to courtship, which may be due to oxidative damage to flies tissues (e.g. central nervous system) induced by free radicals. w had the highest tolerance to UVR, which may be ascribed to its advantage of survival under the natural condition and at high level of SOD activity. This paper is concerned with the theoretical foundation of support vector machines (SVMs). The purpose is to develop further an exact relationship between SVMs and the statistical learning theory (SLT). As a representative, the standard C-support vector classification (C-SVC) is considered here. More precisely, we show that the decision function obtained by C-SVC is just one of the decision functions obtained by solving the optimization problem derived directly from the structural risk minimization principle. In addition, an interesting meaning of the parameter C in C-SVC is given by showing that C corresponds to the size of the decision function candidate set in the structural risk minimization principle.

Single nucleotide polymorphism analysis on melanocortin receptor 1 (MC1R) of Chinese native pig

Melanocortin receptor 1 (MC1R) gene, one of the important candidate genes for coat color trait, was used to analyze the single nucleotide polymorphism (SNP) in Chinese native pig breeds by PCR-single strand conformation polymorphism (PCR-SSCP). The study had also taken 3 imported pig breeds as control. The results showed that the three mutations G284A, T309C and T364C found in Chinese native pigs were consistent to the mutation found in the European Large Black individuals. However, 68CC or C492T and G728A were only found in the imported individuals, which were obviously different from the Chinese native pigs. Accordingly, we presumed that the coat colors of Chinese native pigs belonged to dominant black color sys-tem, which was completely distinct to that of imported pig breeds. Thus it was implied that MC1R gene was not the principal factor affecting the coat color differences of Chinese native pig breeds, but could be used to trace the molecular evolution of pig breeds.

Mutations of the melanocortin-4 receptor (MC4R) gene are associated with the appetite, obesity; growth in pig, mice; human. But little is known about the function of chicken MC4R gene. In this study, F2 chicken resource population derived from broilers crossing to Silky was screened for the polymorphisms of the MC4R gene using PCR-single strand conformation polymorphism (PCR-SSCP); DNA sequencing methods. Four single nucleotide polymorphisms (SNPs) sites were found. The mutation (C→T) in the 5′ regulation region of chicken MC4R gene results in one more NF-E2; cap transcription factor binding sites in the mutation allele than in the wild allele. One missense mutation (G→A) occurs in the coding region (61nt), which changes the glycine to arginine. Moreover, in the coding region there are 2 synonymous mutations, one G→T mutation at 315nt; one C→T mutation at 336nt. Least square analysis of the SNPs; carcass traits showed that BB, DD; FF genotypes are significantly associated with body weight, carcass weight (or half carcass weight),; leg muscle weight (P>0.05 or P>0.01). But no significant association between the genotypes; abdominal fat weight is found. The results present the evidence that the chicken MC4R gene can be selected as the major candidate gene for the carcass traits such as body weight; growth.

We have identified DNA polymorphisms in the gene of insulin-like growth factor 2 by PCR-SSCP in a resource population, which was generated by Silky reciprocally crossing to Broilers. A C→G mutation was detected in the exon 2 (at position 71) by sequencing. This single nucleotide polymorphism (SNP) was found to be associated with production traits. Chicken with BB genotype showed more chest angle width but less 3 week body weight and glandular stom- ach weight than chicken with AA genotype (Pduction of heterozygotes to confirm whether IGF2 locus was imprinting. Progeny from heterozygote × homozygote reciprocal cross was assayed for ex- pression after the genotype was determined. The transcription of IGF2 was detected by RT-PCR-SSCP. IGF2 gene was expressed bialleleically in 1-day-old neonatal liver and 90-day-old liver, kidney, heart, and muscle of both heterozygote AB and BA chickens. Therefore, IGF2 was not an imprinting gene in chicken. The different genetic effects between the het- erozygote AB and BA remain to be elucidated.

Lmbr1 is the key candidate gene controlling vertebrate limb development, but its effects on animal growth and carcass traits have never been reported. In this experiment, lmbr1 was taken as the candi-date gene affecting chicken growth and carcass traits. T/C and G/A mutations located in exon 16 and one A/C mutation located in intron 5 of chicken lmbr1 were detected from Silky, White Plymouth Rock broilers and their F2 crossing chickens by PCR-SSCP and sequencing methods. The analysis of vari-ance (ANOVA) results suggests that T/C polymorphism of exon 16 had significant association with eviscerated yield rate (EYR), gizzard rate (GR), shank and claw rate (SCR) and shank girth (SG); A/C polymorphism of intron 5 was significantly associated with SCR, liver rate and head-neck weight (HNW), while both sites had no significant association with other growth and carcass traits. These results demonstrate that lmbr1 gene could be a genetic locus or linked to a major gene significantly affecting these growth and carcass traits in chicken.

The Yangtze River is the mother river of *** promote the aquatic ecosystem protection of the great river,the Project of Yangtze Fisheries Resources and Environment Investigation(2017-2021)supported by the Ministry of Agriculture and Rural Affairs,*** carried out by 24 institutes and universities that located in the Yangtze River basin surveys the status of(1)fish species composition and spatial distribution,(2)current fish abundance,(3)endangered fishes,(4)Yangtze finless porpoise,(5)aquatic eco-environments,(6)water-level fluctuation areas,(7)capture fisheries and recreational fisheries of the Yangtze River mainstream and 10 of its main tributaries,including Yalong River,Heng River,Min River(including Dadu River),Chishui River,Tuo River,Jialing River,Wu River,Han River,Dongting Lake and Poyang *** results showed that there were 443 fish species(belonging to 163 genus,37 families,and 18 orders)before 2017,but only 323 fish species(including 15 new recorded exotic species)were recorded in the project of *** them,Cyprinus carpio,Carassius auratus,Hypophthalmichthys molitrix,Pelteobagrus fulvidraco,Coilia brachygnathus,Silurus asotus,Saurogobio dabryi,Ctenopharyngodon idellus,Pelteobagrus nitidus,Hemiculter leucisculus,Siniperca chuatsi,Coreius heterodon,Culter alburnus,Parabramis pekinensis,and Aristichthys nobilis were the dominant species across the whole Yangtze River *** is estimated that there were 886 million individuals weighing 124.8 million kg,merely equivalent to 27.3%of the resources in 1950s,30.9%of the resources in 1960s,or 58.7%of the resources in *** the new list of protected fishes that recorded in the Yangtze River system,only 15 of 29 were collected in this project of *** gladius has been affirmed to be Extinct by International Union for Conservation of Nature(IUCN).The wild individuals of Tenualosa reevesii and Luciobrama macrocephalus have disappeared for many years and maybe have been extinct
