Since December 2019,just a month before the Chinese Spring Festival,multiple cases of pneumonia of unknown etiology appeared in Wuhan,Hubei Province,***,a novel coronavirus was identified in a bronchoalveolar lavage fluid sample from the Wuhan Seafood Market using metagenomic next-generation sequencing technology.1 On February 11,2020,the virus was named severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)by the International Committee on Taxonomy of Viruses(ICTV).SARS-CoV-2 is the seventh member of the coronavirus family that can infect humans after the emergence of severe acute respiratory syndrome coronavirus(SARS-CoV)and Middle East respiratory syndrome coronavirus(MERS-CoV).

裂缝类路面损坏是城市道路中常见的病害,能否在众多路面信息图像中高效获取路面裂缝信息,是当前领域研究的关键点.为得到更高精度的图像识别分析,结合深度学习领域,基于Res2Unet-CBAM网络模型进行路面裂缝图像分割工作,分析了公共数据集的实验结果,并对比了UGNet,Res2Net等多种语义分割模型,在此基础上设计了Res2-Unet多尺度路面裂缝分割网络模型.通过对常见通道注意力模块进行分析,将CBAM通道注意力模块引入Res2-Unet分割模型,构建了一种新的Res2Unet-CBAM网络模型用于路面裂缝图像的分割任务.将Res2Unet-CBAM模型与其他深度学习模型的实验结果进行对比,结果表明此模型具有更好的图像分割效果.

Single-cell RNA sequencing(scRNA-seq)is generally used for profiling transcriptome of individual *** droplet-based 10X Genomics Chromium(10X)approach and the plate-based Smart-seq2 full-length method are two frequently used scRNA-seq platforms,yet there are only a few thorough and systematic comparisons of their advantages and ***,by directly comparing the scRNA-seq data generated by these two platforms from the same samples of CD45cells,we systematically evaluated their features using a wide spectrum of ***-seq2 detected more genes in a cell,especially low abundance transcripts as well as alternatively spliced transcripts,but captured higher proportion of mitochondrial *** composite of Smart-seq2 data also resembled bulk RNA-seq data *** 10X-based data,we observed higher noise for mRNAs with low expression *** 10%30%of all detected transcripts by both platforms were from non-coding genes,with long non-coding RNAs(lncRNAs)accounting for a higher proportion in 10X.10X-based data displayed more severe dropout problem,especially for genes with lower expression ***,10X-data can detect rare cell types given its ability to cover a large number of *** addition,each platform detected distinct groups of differentially expressed genes between cell clusters,indicating the different characteristics of these *** study promotes better understanding of these two platforms and offers the basis for an informed choice of these widely used technologies.

The novel influenza A （H1N1） 2009 virus has emerged to cause the first pandemic of the twenty-first century. Disease outbreaks caused by the influenza A （H 1N 1） virus have prompted concerns about the potential for a pandemic and have driven the development of vaccines against this subtype of influenza A. In this study, we developed a monovalent influenza A （H 1N 1） split vaccine and evaluated its effects in BALB/c mice. Mice were immunized subcutaneously with 2 doses of the vaccine containing hemagglutinin （HA） alone or HA plus an aluminum hydroxide （AI（OH）3） adjuvant. Immunization with varying doses of HA （3.75, 7.5, 15, 30, 45 or 60μg） was performed to induce the production of neutralizing antibodies. The vaccine elicited strong hemagglutination inhibition （HI） and microneutralization, and addition of the adjuvant augmented the antibody response. A preliminary safety evaluation showed that the vaccine was not toxic at large doses （0.5 ml containing 60 μg HA＋600 μg AI（OH）3 or 60 μg HA）. Moreover, the vaccine was found to be safe at a dose of 120 μg HA＋ 1200 μg AI（OH）3 or 120 μg HA in 1.0 ml in rats. In conclusion; the present study provides support for the clinical evaluation of influenza A （H1N1） vaccination as a public health intervention to mitigate a possible pandemic. Additionally, our findings support the further evaluation of the vaccine used in this study in primates or humans.

随着淡水资源紧缺和海水淡化处理成本的下降,海水淡化厂数量快速增加,大量浓盐水进入海区会导致受纳海域盐度升高。高浓度盐水给海洋生态环境带来许多负面影响,有关浓盐水的海洋生态安全问题备受人们关注。文章就青岛市董家口港海水淡化浓盐水排放对海洋环境造成的潜在影响进行监测分析,结果显示浓盐水排放仅会对排放口周边极小范围产生影响(200 m以内),且未对琅琊湾周围海域海洋生物造成较大环境风险,表明董家口海水淡化厂浓盐水排放对环境影响较小。

Non-flow aqueous zinc-bromine batteries without auxiliary components(e.g.,pumps,pipes,storage tanks)and ion-selective membranes represent a cost-effective and promising technology for large-scale energy ***,they generally suffer from serious diffusion and shuttle of polybromide(Br^(-),Br^(3-))due to the weak physical adsorption between soluble polybromide and host carbon materials,which results in low energy efficiency and poor cycling ***,we develop a novel self-capture organic bromine material(1,10-bis[3-(trimethylammonio)propyl]-4,4'-bipyridinium bromine,NVBr4)to successfully realize reversible solid complexation of bromide components for stable non-flow zinc-bromine battery *** quaternary ammonium groups(NV^(4+)ions)can effectively capture the soluble polybromide species based on strong chemical interaction and realize reversible solid complexation confined within the porous electrodes,which transforms the conventional“liquid-liquid”conversion of soluble bromide components into“liquid-solid”model and effectively suppresses the shuttle ***,the developed non-flow zinc-bromide battery provides an outstanding voltage platform at 1.7 V with a notable specific capacity of 325 mAh g^(-1)NVBr4(1 A g^(-1)),excellent rate capability(200 mAh g^(-1)NVBr4 at 20 A g^(-1)),outstanding energy density of 469.6 Wh kg^(-1)and super-stable cycle life(20,000 cycles with 100%Coulombic efficiency),which outperforms most of reported zinc-halogen *** mechanism analysis and DFT calculations demonstrate that the chemical interaction of quaternary ammonium groups and bromide species is the main reason for suppressing the shuttle *** developed strategy can be extended to other halogen batteries to obtain stable charge storage.

Identification of immunoreactive proteins of Brucella melitensis by immunoproteomics

Infection with Brucella causes brucellosis, a chronic disease in humans, which induces abortion and sterility in livestock. Among the different Brucella species, Brucella melitensis is considered the most virulent and is the predominant species associated with outbreaks in China. To date, no safe human vaccine is available against Brucella infection. The currently used live vaccines against Brucella in livestock induce antibodies that interfere with the diagnosis of field infection in vaccinated ani- mals, which is harmful to eradication programs. However, there is as yet no complete profile of immunogenic proteins of B. melitensis. Towards the development of a safer, equally efficacious, and field infection-distinguishable vaccine, we used immunoproteomics to identify novel candidate immunogenic proteins from B. melitensis M5. Eighty-eight immunoreactive protein spots from B. melitensis M5 were identified by Western blotting and were assigned to sixty-one proteins by mass spectrometry, including many new immunoreactive proteins such as elongation factor G, FOFI ATP synthase subunit beta, and OMPI. These provide many candidate immunoreactive proteins for vaccine development.

Influenza virus (IAV)infection is a major cause of severe respiratory illness that affects almost every country in the *** infections result in respiratory illness and even acute lung injury and death,but the underlying mechanisms responsible for IAV pathogenesis have not yet been fully *** this study,the basic fibroblast growth factor 2 (FGF2)level was markedly increased in H1N1 virus-infected humans and ***2,which is predominately derived from epithelial cells,recruits and activates neutrophils via the FGFR2-PI3K-AKT-NFKB signaling ***2 depletion or knockout exacerbated influenzaassociated disease by impairing neutrophil recruitment and *** importantly,administration of the recombinant FGF2 protein significantly aUeviated the severity of IAV-induced lung injury and promoted the survival of IAV-infected *** on the results from experiments in which neutrophils were depleted and adoptively transferred,FGF2 protected mice against IAV , infection by recruiting ***,FGF2 plays a critical role in preventing IAV-induced lung injury,and FGF2 is a promising potential therapeutic target during IAV infection.
