HSP110 aggravates ischemia-reperfusion injury after liver transplantation by promoting NF-κB pathway

Background:Ischemia-reperfusion injury(IRI)poses a significant challenge to liver transplantation(LT).The underlying mechanism primarily involves overactivation of the immune *** shock protein 110(HSP110)functions as a molecular chaperone that helps stabilize protein ***:An IRI model was established by performing LT on Sprague-Dawley rats,and HSP110 was silenced using ***-eosin staining,TUNEL,immunohistochemistry,ELISA and liver enzyme analysis were performed to assess IRI following *** blotting and quantitative reverse transcription-polymerase chain reaction were conducted to investigate the pertinent molecular ***:Our findings revealed a significant increase in the expression of HSP110 at both the mRNA and protein levels in the rat liver following LT(P<0.05).However,when rats were injected with siRNAHSP110,IRI subsequent to LT was notably reduced(P<0.05).Additionally,the levels of liver enzymes and inflammatory chemokines in rat serum were significantly reduced(P<0.05).Silencing HSP110 with siRNA resulted in a marked decrease in M1-type polarization of Kupffer cells in the liver and downregulated the NF-κB pathway in the liver(P<0.05).Conclusions:HSP110 in the liver promotes IRI after LT in rats by activating the NF-κB pathway and inducing M1-type polarization of Kupffer *** HSP110 to prevent IRI after LT may represent a promising new approach for the treatment of LT-associated IRI.

Sepsis progression is significantly associated with the disruption of gut ***,the modulatory mechanisms of gut microbiota operating during sepsis are still ***,we investigated how gut commensals impact sepsis development in a pre-clinical *** ligation and puncture(CLP)surgery was used to establish polymicrobial sepsis in *** depleted of gut microbiota by an antibiotic cocktail(ABX)exhibited a significantly higher level of mortality than *** determined by metabolomics analysis,ABX treatment has depleted many metabolites,and subsequent supplementation with L-rhamnose(rhamnose,Rha),a bacterial carbohydrate metabolite,exerted profound immunomodulatory properties with a significant enhancement in macrophage phagocytosis,which in turn improved organ damage and ***,rhamnose binds directly to and activates the solute carrier family 12(potassium-chloride symporter),member 4(SLC12A4)in macrophages and promotes phagocytosis by activating the small G-proteins,Ras-related C3 botulinum toxin substrate1(Rac1)and cell division control protein 42 homolog(Cdc42).Interestingly,rhamnose has enhanced the phagocytosis capacity of macrophages from sepsis *** conclusion,by identifying SLC12A4 as the host interacting protein,we disclosed that the gut commensal metabolite rhamnose is a functional molecular that could promote the phagocytosis capacity of macrophages and protect the host against sepsis.

Discharge mode and particle transport in radio frequency capacitively coupled Ar/O_(2)plasma discharges

Influence of organic matter input and temperature change on soil aggregate-associated respiration and microbial carbon use efficiency in alpine agricultural soils

Understanding the dynamics of soil respiration,microbial carbon use efficiency(CUE),and temperature sensitivity(Q_(10))in response to exogenous organic matter(EOM)input,soil aggregate size,and incubation temperature is crucial for predicting soil carbon cycling responses to environmental *** this study,these interactions were investigated by 180-day incubation of soil aggregates supplemented with EOM at various temperatures(5°C,15°C and 25°C).The results reveal an‘L-shaped’trend in soil respiration on the time scale across all treatments,characterized by initial rapid declines followed by *** input and higher temperatures significantly enhance respiration ***,the respiratory rates of soil aggregates of different sizes exhibit distinct patterns based on the presence or absence of *** conditions without the addition of EOM,larger aggregates show relatively lower respiration ***,in the presence of EOM,larger aggregates exhibit higher respiratory ***,Q_(10)decreases with increasing aggregate *** relationship between Q_(10)and the substrate quality index(SQI)supports the carbon quality temperature(CQT)hypothesis,highlighting SQI’s influence on Q_(10)values,particularly during later incubation *** CUE decreases with EOM input and rising ***,aggregate size plays a role in microbial CUE,with smaller aggregates exhibiting higher CUE due to enhanced nutrient *** conclusion,the intricate interplay of EOM input,aggregate size,and temperature significantly shapes soil respiration,microbial CUE,and Q_(10).These findings underscore the complexity of these interactions and their importance in modeling soil carbon dynamics under changing environmental conditions.

Low-frequency Stimulation at the Subiculum Prevents Extensive Secondary Epileptogenesis in Temporal Lobe Epilepsy

Secondary epileptogenesis is characterized by increased epileptic susceptibility and a tendency to generate epileptiform activities outside the primary *** is one of the major resultants of pharmacoresistance and failure of surgical outcomes in epilepsy,but still lacks effective ***,we aimed to test the effects of low-frequency stimulation(LFS)at the subiculum for secondary epileptogenesis in a mouse ***,secondary epileptogenesis was simulated at regions both contralateral and ipsilateral to the primary focus by applying successive kindling *** kindled at the right CA3 showed higher seizure susceptibilities at both the contralateral CA3 and the ipsilateral entorhinal cortex and had accelerated kindling processes compared with naive *** at the ipsilateral subiculum during the primary kindling progress at the right CA3 effectively prevented secondary epileptogenesis at both the contralateral CA3 and the ipsilateral entorhinal cortex,characterized by decreased seizure susceptibilities and a retarded kindling process at those secondary *** application along with the primary epileptogenesis was ***,the effects of LFS on secondary epileptogenesis were associated with its inhibitory effect at the secondary focus through interfering with the enhancement of synaptic connections between the primary and secondary *** results imply that LFS at the subiculum is an effective preventive strategy for extensive secondary epileptogenesis in temporal lobe epilepsy and present the subiculum as a target with potential translational importance.

Innovative 3D Microfluidic Intestinal Organoid Model for Assessing Cadmium Bioavailability in Food: Implications for Enhanced Exposure Risk Assessment

Given the severe toxicity and widespread presence of cadmium (Cd) in staple foods such as rice， accurate dietary exposure assessments are imperative for public health. In vitro bioavailability is commonly used to adjust dietary exposure levels of risk factors; however， traditional planar Transwell models have limitations， such as cell dedifferentiation and lack of key intestinal components， necessitating a more physiologically relevant in vitro platform. This study introduces an innovative three-dimensional (3D) intestinal organoid model using a microfluidic chip to evaluate Cd bioavailability in food. Caco-2 cells were cultured on the chip to mimic small intestinal villi's 3D structure， mucus production， and absorption functions. The model's physiological relevance was thoroughly characterized， demonstrating the formation of a confluent epithelial monolayer with well-developed tight junctions (ZO-1)， high microvilli density (F-actin)， and significant mucus secretion (Alcian blue staining)， closely resembling the physiological intestinal epithelium. Fluorescent particle tracking confirmed its ability to simulate intestinal transport and diffusion. The Cd bioavailability in rice measured by the 3D intestinal organoid model (9.07% ± 0.21%) was comparable to the mouse model (12.82% ± 3.42%) but significantly lower than the Caco-2 monolayer model (26.97% ± 1.11%). This 3D intestinal organoid model provides a novel and reliable strategy for in vitro assessment of heavy metal bioavailability in food， with important implications for food safety and risk assessment.
