The Effects of Several Types of Induced Abiotic Stress on Cephalaria joppensisGermination under Controlled Conditions

The recently domesticated species, Cephalaria joppensis (CJ), is emerging as a new alternative forage crop in Israel. It has high biomass potential and nutritional values that are comparable to forage wheat. However, CJ emerges slowly under cold conditions, which hinders its development as a major winter crop. Additional tolerance for abiotic stress would improve its performance as a forage crop. We examined the effects of several abiotic factors (i.e., cold, salinity, drought and pH) on CJ germination under controlled conditions. The effect of temperature was studied by incubating seeds at different temperatures between 7°C and 35°C. The effects of salinity, osmotic potential and pH were tested by incubating seeds at different NaCl, PEG and pH levels, respectively. Temperature, salinity and osmotic potential significantly affected germination;whereas pH did not. Temperature did not affect the final proportion of germinated seeds, but did affect other germination-rate variables, indicating that germination rate might be the limiting factor under field conditions. Salinity also affected germination-rate variables, but not the proportion of seeds that germinated. Notably, CJ was found to be relatively resistant to high salt concentrations, with a 273 mM NaCl threshold for germination, indicating its potential as a relatively salt-tolerant forage crop. Both the proportion of germinated seeds and the germination rate were highly sensitive to the osmotic-potential treatments, indicating that drought resistance will remain the biggest challenge for CJ. This study provides baseline data for a rapid and efficient system for further screening for abiotic-stress tolerance among wild and cultivated lines of CJ.

Performance of Comparative Cervical Tuberculin Test and Serological Methods with Culturing of Nasal Swab in Diagnosis of Bovine Tuberculosis in Cross Breed Cattle Baghdad Iraq: A Comparative Evaluation

This study was designed to assess the diagnostic value of Rapid Antigen bovine TB antibody test kit (RAT) and any association with cervical comparative tuberculin test (CCTT), (iELISA) and nasal swabs culturing, among based detection of M. bovis infection. A herd of 21 animals aged 1 - 8 years cross bread cattle of college of veterinary medicine. 19 (90.47%) animals had good body condition scores, two bulls included, and 2 (9.52%) cows were fair. Serum samples were collected, analyzed for anti-bovine TB antibody using RAT and iELISA. Also the herd was screened by CCTT. The tests were carried out twice, more than ten month interval, and twelve nasal swabs were taken within second survey. The first survey results revealed prevalence rate: 4 (19.04%) animals considered positive results (one positive and 3 suspected results) for CCTT, while the prevalence rate according to RAT was 10 (47.61%). The difference between the two prevalence rate was significant (McNemar chi-statistic = 4.50, p-value = 0.03) Kappa = 0.215 95% confidence interval: from -0.128 to 0.558;the strength of agreement is considered to be “fair”. The study interprets: sensitivity 30%;specificity 99%. The second survey results revealed prevalence rate according to CCTT was 4 (36.36%), while prevalence rate according to RAT was 5 (45.45%). The difference between the two prevalence was not significant (McNemar chi-statistic = 0.33, p-value = 0.56). Kappa = 0.441 95% confidence interval: from -0.087 to 0.968;the strength of agreement is considered to be “moderate”;sensitivity: 60%;specificity: 83%. All serum samples and nasal swabs gave negative results for iELISA and culturing respectively. The study concluded that RAT was highly specific, easy, labor and time saving, suggesting its use as screening test in bovine tuberculosis, and CCTT could be used to confirm positive animals screened by RAT, while there was no association between RAT, CCTT with body condition scores, iELISA and nasal swabs culture resul

Molecular and Phenotypical Characterization of Mannheimia haemolyticaIsolated from Goats in Baghdad Province

Mannheimia haemolytica (M. haemolytica) is a gram negative bacterium which can infect humans and animals. It’s commensal as a normal flora of the nasopharynx and tonsils in cattle, sheep and goats, pneumonic pasteurellosis is one of the most economically important infectious disease in goats worldwide prevalence. This study aimed to investigate the incidence of M. haemolytica by bacteriological and molecular characterization in goats. One hundred nasopharyngeal swabs were collected from apparently healthy field goats, seven lung tissue specimens and five nasal mucus swabs from slaughtered goats in Baghdad. All samples were cultured on Blood and MacConky agars. Biochemical tests and EPI20E kit were used for identification of the suspected colonies. 5 (4.46%) isolates of M. haemolytica were identified phenotypicaly and confirmed diagnosis by polymerase chain reaction (PCR) technique using two primers 16s rRNA and 12s rRNA genes .The results of this study concluded that identification of M. haemolytica by PCR was in accordance with those of phenotypic tests and it providing the basis for effective preventative strategies through epidemiological studies performance.

In VitroEvaluation of Some Traits in Stevia rebaudiana(Bertoni) under Drought Stress and Their Relationship on Stevioside Content

The experiment was conducted to evaluate the differences of plant growth as well as sweetener material of two types of Stevia rebaudiana under conditions of absence and presence of drought stress. In vitro, the experiment was conducted as drought traits using four levels of polyethylene glycol (0.0, 10,000, 20,000, 30,000 ppm). It was observed that survival % and No. of shoots should be considered as the important factors associated with stevioside content in stevia plant. According to path coefficient analysis and positive direct effect of studied traits, survival % recorded the highest value (0.5386), whereas No. of shoots recorded the highest value (– 0.8827) as negative direct effect in related to stevioside content. The results showed that all traits were affected, especially under 30,000 ppm level of polyethylene glycol. Generally, the most of studies traits exhibited a recorded clearly difference between the two types and used drought levels on stevia plants. Therefore, selection based on survival % would be more effective to improving stevioside content of stevia plants in drought stress conditions.

Sensitivity and Specificity of Various Serological Tests for Detection of Brucella spp. Infection in Male Goats and Sheep

Brucellosis is a zoonosis found throughout the world with major implications both in the field of public health and farming economy. It is most often caused by Brucella abortus and Brucella melitensis. This study aimed for evaluation of the sensitivity and specificity of immunochromatographic assay (ICA) and comparing the results with those obtained from Rose Bengal Plate Test (RBPT) and in direct Enzyme Linked Immunsorbent Assay (ELISA) for serodiagnosis of sheep and goat brucellosis. Out of 41 sera samples (21 ram and 20 male goats) which were examined, antibodies against Brucella were detected in ram 8 (38.09%), 6 (28.57%) and 16 (76.19%) in ram samples, whereas they were detected in 3 (15%), 1 (5%) and 12 (60%) serum sample of goats males by RBPT, (ICA) and iELISA respectively. The kappa value between iELISA and ICA was -0.08 and that between RBPT and ELISA was -0.01 in ram and the kappa value between iELISA and ICA was -0.06 and that between RBPT and ELISA was -0.14 in male goats. The sensitivity for RBPT and ICA were 37.5% and 25% while specificity was 60% and 60% in ram;whereas, in male goats, sensitivity for RBPT was 8.33% and ICA was 8.33% while specificity was 75% and 100% respectively. It can be concluded that the developed ICA is immunodiagnostic assay, and it is rapid, non-expensive, economical and suitable for large-scale screening in developing countries and rural areas.
