The key pathogenesis of chronic myeloid leukemia(CML)is the formation of BCR-ABL fusion gene,encoding a 210 kDa Bcr-Abl tyrosine kinase,which is crucial for the occurrence and development of ***(IM)is the first targeted anticancer drug approved by FDA for the treatment of CML;however,some patients,especially those in accelerated phase and blastic phase,develop primary or secondary drug resistance to ***,the most challenging resistance is caused by T315I mutation of Bcr-Abl,which represents approximately 15%–20%of all acquired mutations and renders cell resistant to a variety of tyrosine kinase inhibitors.1,2 Thus,there is an urgent need to develop novel strategies to overcome Bcr-Abl T315I-meidated IM resistance.

Lysosomes are the“cell stomach”that digests various species including macromolecules and pathogens for self-repair and self-defense upon fusion with endosomes,autophagosomes,and ***,artificially manipulating intracellular vesicles could potentially promote lysosome-mediated intracellular digestion for disease prevention and ***,a supramolecular strategy to efficiently tether and fuse acidic vesicles(i.e.,lysosomes,late endosomes,autolysosomes,and phagosomes)has been developed for the first time to enhance the digestion of cellular wastes and foreign matter,including pathogens.A linear polyethylene glycol(PEG)derivative dually tagged with morpholine(MOR)and adamantane(ADA)on the opposite side,namely,MOR-PEG-ADA,was designed to target intracellular acidic vesicles via MOR and to decorate their surfaces with ***,the addition of cucurbit[7]uril(CB[7])grafted hyaluronic acid(HA)induced supramolecular tethering and fusion of acidic vesicles via strong host-guest interactions between CB[7]of CB[7]-HA and ADA residing on the surface of acidic *** a proof-ofconcept,the overall cellular metabolism,including endogenous autophagy and the exogenous endocytosis and phagocytosis,was effectively upregulated,demonstrating that supramolecular regulation of the dynamics of intracellular acidic vesicles may promote the lysosome’s digestion to significantly improve cellular self-repair and self-defense.

By hydrothermal synthesis, a manganese（H） tetracarboxylate [Mn（2,2′-bipyridi- ne）（btec）0.5（HEO）]En （H4btec = 1,2,4,5-benzenetetracarboxylic acid） was obtained and characterized by single-crystal X-ray diffraction, IR, elemental analysis and thermogravimetric analysis. It crystallizes in triclinic, space group P1, with a = 7.721（4）, b = 8.905（5）, c = 10.712（6）A, a = 80.899（8）, β = 70.371（8）, y = 78.201（8）°, Z = 2, V= 675.8（7）A3, Mr= 354.20, De= 1.741 g/cm^3,μ = 1.007 mm^-1, F（000） = 360, S = 0.956, （△/σ）max = 0.000, the final R = 0.0490 and wR = 0.1262. This compound presents an infinite 1D polymer featuring a double-chain structure. The Mn（Ⅱ） ion is five-coordinated. The four carboxylate groups of btec4- are all deprotonated and coordinated to four Mn atoms in a monodentate fashion, forming a 1D double chain structure with a cavity. Such chains are interconnected by π-π stacking interactions into 2D layers which are further interlinked into a supramolecular structure by hydrogen bonds.

Aims: Colorectal cancer is one of the most common malignant tumor of digestive tract. 5-Fluorouracil（5-Fu） is the first line therapy for patients with colorectal cancer（CRC）. However, acquired resistance limits the application of 5-Fu in clinical. NF-кB pathway activation, deubiquitinase mediated chemoresistance and redox homeostasis damage are responsible for 5-FU resist. Yet the specific mechanism of drug resistance is still unclear. To improve CRC patients survival, it’s urgently needed to identify novel drug targets and effective therapeutic agent.b-AP15 is an alternative molecular inhibitor targeting 19 S deubiquitinase USP14 and UCHL5, showing efficacy in several other cancer types. This assay first reports the cytotoxicity effect and its mechanism of b-AP15 in 5-FU sensitive and resistant ***: Cell lines including two pairs of CRC and its 5-FU resist cell lines（RKO/RKO 5-FU-R,HCT15/HCT 15 5-FU-R） were used in the current study. Cell viability was measured by MTS assay. Flow cytometry with Annexin V-fluoroisothiocyanate（FITC）/PI double staining were applied to test apoptosis level of b-AP15 treated CRC and 5-FU resist CRC cell lines. Cell proliferation was evaluated by plate clone formation assay and EdU incorporation assay. The mitochondrial membrane potential of b-AP15 treated or untreated cells were evaluated by mitochondrial membrane potential *** and message RNA（mRNA） level were detected by Western Blot and RT-qPCR *** were used to detect cell migration and invasiveness. DCF-DA kit was used to measure total reactive oxygen species（ROS）. ELISA was applied to evaluate cell ATP content of b-AP15 treated CRC cells with D-fluorescein potassium salt. Nuclear translocation was observed by immunofluorescence. Mouse xenograft models of Luciferase-RKO and Luciferase-RKO 5-FU-R cells were used to determine in vivo effects of b-AP15 in CRC ***: Compared to non-resistant cell, the expression of mRNA and protein level of USP

The perfluorinated cages with all the exo polarized Cδ+-Fδ-bonds have been proposed to be efficient containers of excess *** manipulate the localizations and conversions of excess electrons and spin states,we presented the theoretical investigations on the double-cage-like single molecular solvated dielectron systems,e2@C20F18（NH）2C20F18（e2@BB’）at the B3LYP/6-31G（d）+4s4p theory ***’s found that the lone pair of excess electrons is confined inside one cage（left or right）to form two different singlet electromers（1L and 1R）,while the two excess electrons are,respectively,trapped inside different cages to form triplet structure（3,ground state）.Interestingly,the application of small external electric field of 21（-21）×10-4 au results in single electron transfer of 3→1L（1R）and such transfer is also a spin state conversion of T→S.

四旋翼飞行器的姿态控制是无人机稳定飞行的关键技术之一。针对复杂多变作业条件下飞行器参数会经常发生变化的问题,在典型控制方法的基础上提出一种自适应的智能控制方法。首先,根据牛顿欧拉定律推导出无人机在地理坐标系下的动力学模型,并对其中参数进行测量计算;然后基于三角形隶属度函数建立模糊控制器,作为外环自主切换的两种控制方式之一,并设置平滑切换过程;最后结合外环对姿态角的控制方法以及内环对角速度快速调整的PD控制方法,实现了无人机串级PID控制方法。仿真和实验结果表明,该系统能够有效控制四旋翼飞行器的飞行姿态。相比较其它算法,其具有更好的鲁棒性和姿态调节的快速性。提升了无人机在飞行过程中抵抗环境扰动和系统动态响应的能力,为四旋翼飞行器控制研究提供了重要的理论与实践基础。

目的探讨血清降钙素原（PCT）和超敏C-反应蛋白（hs-CRP）对肾盂肾炎的临床价值。方法 2012年1~12月肾盂肾炎患者134例,检测其入院后的PCT和hs-CRP,并记录尿培养结果。同期健康体检组134例作为健康对照组。结果尿培养结果阳性率为32.1%,PCT和hs-CRP敏感度分别为94.0%、97.0%,特异度分别为82.8%、64.2%;PCT与hs-CRP联合诊断敏感度为98.5%,特异度为89.6%。结论 PCT和hs-CRP联合检测在肾盂肾炎的诊断上具有高敏感度和高特异度,对指导临床及时、合理用药有辅助作用。
