Comprehensive Flow Cytometry Analysis of PEI-Based Transfections for Virus-Like Particle Production

The generation of stable clones for biomolecule production is a common but lengthy and labor-intensive *** complex molecules,such as viruses or virus-like particles(VLPs),the timeline becomes even more ***,in the early stages of development,transient production methods serve as a reasonable alternative to stable clone *** this work,an investigation of a polyethylenimine-(PEI-)based transfection method for the transient production of Chikungunya(Chik)VLPs,a vaccine candidate molecule,was *** effort focuses on tracking cell population responses during transfection,understanding how process changes affect these responses,and monitoring patterns in cell performance over the culture *** labeling and VLP staining were employed to comprehensively track cells via flow cytometry and to draw correlations between plasmid DNA(pDNA)uptake and the resulting VLP *** method detected high transfection efficiency(≥97%)in all samples tested and demonstrated the capability to track kinetics of plasmid-cell *** varied transfection cell concentrations,the pDNA binding kinetics are altered and saturation binding is observed in the lowest cell concentration sample tested in less than 3 hours of ***,in all samples,the flow cytometry analysis of relative pDNA amount versus VLP expression staining showed that cells which contained fewer pDNA complexes resulted in the highest levels of VLP ***,to determine the potential breadth of our observations,we compared daily expression patterns of ChikVLP with a reporter,monomeric GFP *** similarities detected suggest the interpretations presented here to likely be more broadly informative and applicable to PEI-based transient production of additional biological products as well.

Institutional experience of PTH evaluation on fine-needle washing after aspiration biopsy to locate hyperfunctioning parathyroid tissue

Assaying parathyroid hormone(PTH) in the washing liquid after fine-needle aspiration biopsy(FNAB) seems to be a valid approach to locate parathyroid ***-FNAB was evaluated in 47 patients with a clinical picture of primary hyper-parathyroidism(PHP) and ultrasonography(US) suggestive of parathyroid *** patients were subdivided into two groups on the basis of the absence or presence of US thyroid *** result of PTH-FNAB was compared with those of cytology,scintigraphy and,in 24 patients,surgical ***-FNAB samples with a value higher than that recorded in the serum and higher than our institutional cut-off were deemed to be probable samples of parathyroid *** proved diagnostic for benign thyroid lesions,non-diagnostic for thyroid lesions,hyperplastic parathyroid tissue,undetermined or malignant thyroid lesions and other lesions in 45%,30%,17%,4%,and 4% of cases,*** 47% of cases,PTH-FNAB indicated that the sample had been taken in parathyroid *** patients without US alterations,the diagnostic accuracy of PTH-FNAB was greater than that of *** surgery,comparison between the results of PTH-FNAB and scintigraphy,in terms of positive predictive value(PPV),revealed the superiority of PTH-FNAB;PPV was 94% for FNAB and 71% for scintigraphy,while sensitivity was 83% and 69%,***-FNAB evaluation after FNAB appears to be more diagnostic than cytology and *** all the procedures used,PTH-FNAB appears to be the method of choice when the target is US suggestive and ***-FNAB appears to be a useful method of guiding surgical intervention.

DNA Copy Number Aberrations in Breast Cancer by Array Comparative Genomic Hybridization

Array comparative genomic hybridization (CGH) has been popularly used for analyzing DNA copy number variations in diseases like cancer. In this study, we investigated 82 sporadic samples from 49 breast cancer patients using 1-Mb resolution bacterial artificial chromosome CGH arrays. A number of highly frequent genomic aberrations were discovered, which may act as "drivers" of tumor progression. Meanwhile, the genomic profiles of four "normal" breast tissue samples taken at least 2 cm away from the primary tumor sites were also found to have some genomic aberrations that recurred with high frequency in the primary tumors, which may have important implications for clinical therapy. Additionally, we performed class comparison and class prediction for various clinicopathological parameters, and a list of characteristic genomic aberrations associated with different clinicopathological phenotypes was compiled. Our study provides clues for further investigations of the underlying mechanisms of breast carcinogenesis.

The Thesaurus Conception of Russian Youth

There are deep-rooted traditions of researches on the youth problems in Russia, By their trends and purposes they partly concur with the traditions of the humanities in Europe and America. In Russia in different timesmit was the same way in the West-diverse youth concepts had been conveying and continue to express the society＇s expectations for new generations. This is in a sense a theoretical mirror of the natural process of generation change. Under modern conditions these concepts can be reduced to three directions： youth-＂no man＇s land＂, youth-social danger, youth-hope of society. At the same time youth theories have the mark of the socio-cultural contexts and contexts of the development of the humanities in Russia. In this article these similarities and distinctions will be examined.

The Plastid-Localized NAD-Dependent Malate Dehydrogenase Is Crucial for Energy Homeostasis in Developing Arabidopsis thaliana Seeds

In the absence of photosynthesis, ATP is imported into chloroplasts and non-green plastids by ATP/ADP transporters or formed during glycolysis, the latter requiring continuous regeneration of NAD＋, supplied by the plastidial isoform of NAD-MDH. During screening for T-DNA insertion mutants in the plNAD-MDH gene of Arabidopsis, only heterozygous plants could be isolated and homozygous knockout mutants grew only after complementation. These heterozygous plants show higher transcript levels of an alternative NAD＋-regenerating enzyme, NADH-GOGAT, and, remarkably, improved growth when ammonium is the sole N-source. In situ hybridization and GUS-histochemical stain- ing revealed that plNAD-MDH was particularly abundant in male and female gametophytes. Knockout plNAD-MDH pollen exhibit impaired tube growth in vitro, which can be overcome by adding the substrates of NADH-GOGAT. In vivo, knockout pollen is able to fertilize the egg cell. Young siliques of selfed heterozygous plants contain both green and white seeds corresponding to wild-type/heterozygous （green） and homozygous knockout mutants （white） in a （1：2）：1 ratio. Embryos of the homozygous knockout seeds only reached the globular stage, did not green, and developed to tiny wrinkled seeds. Complementation with the gene under the native promoter rescued this defect, and all seeds developed as wild-type. This suggests that a blocked major physiological process in plNAD-MDH mutants stops both embryo and endosperm development, thus avoiding assimilate investment in compromised offspring.

Priority Directions of National Policy of Regulation of Mutual Relations Between Youth and Mass Media： The Theoretical and Empirical Researches

The study was concerned with the problem of the Russian youth, with a special emphasis on the history of the theoretical and empirical researches devoted to the youth issues. The research delineates the term thesaurus and its connection with the formation of youth＇s outlook. The central focus of the research is social and cultural value orientations of the young Russian. The study presents a detailed analysis of impact that mass media plays in socialization of youth, its generations, its influence on public opinion formation and priority directions of national policy of regulation of mutual relations between youth and mass media. It attempts to examine the presence of mass media in youth＇s everyday life. The research provides a detailed analysis of contemporary state of the youth＇s trust to the mass media and degree of dependence on it. The study also presents a detailed analysis of the traces of mass media＇s influence on young Russians. The completed research is fundamental for the sociological and the philosophical generalisations that underline comparative research on the youth problems in Russia, its social and cultural value orientations and the influence on public opinion formation and priority directions of national policy of regulation of mutual relations between youth and mass media.

Plant triterpenoid saponins function as susceptibility factors to promote the pathogenicity of Botrytis cinerea

Impact of mine waters on chemical composition of soil in the Partizansk Coal Basin, Russia

Partizansk Coal Basin,located in the south of the Russian Far Eastern Region,was intensively mined from 1918 until *** it was mostly explored by underground excavation,the natural landscapes were transformed into anthropogenic *** the mines closed,ground subsidence occurred widely,especially in areas near the waste *** caused water tables to rise to the surface and pollute the *** of the hydrochemical composition of the mine waters were conducted in 2011-2013,and showed low alkalinity and average level of *** can be explained by the fact that while going up through soil mass,the mine waters lost their much of their pollutants due to soil buffering All mine water samples contain thermo-tolerant coliform bacteria *** that indicates a source of fresh fecal *** research indicates increased hydrocarbon ion concentrations in mine waters,especially in autumn,that resulted in accumulation of chromium and copper compounds,which can cause soil pollution.A strong relationship between the chemical composition of the mine waters and soil extracts was found within areas of unregulated groundwater discharge on the *** negative correlation between pH and content of metal compounds including chromium and copper was found at the 'Avangard' mine (r =-0.95);and between alkalinity and chromium content at the 'Glubokaya' mine (r =-0.94).
