Population genetic structure and diversity of high value vulnerable medicinal plant Acorus calamus in India using RAPD and chloroplast microsatellite markers

Acorus calamus is a highly valued medicinal plant with global distribution used in several drugs of health care systems. We evaluated the genetic diversity and population structure of 50 populations of A. calamus from different geographical regions in India through RAPD and chloroplast microsatellite markers. From the total screened 82 RAPD primers and 18 cpssR primers, 10 RAPD and nine cpssRs were found polymorphic. The selected 10 RAPD primers produced a total of 96 reproducible bands, out of which 65 were polymorphic （67.70%）. Whereas, the selected nine cpssR markers produced 26 alleles and all of them were polymorphic. The mean genetic diversity （H T ） among populations using RAPD （H T = 0.263） and cpssR （H T = 0.530） markers was higher in comparison to the mean genetic diversity within populations. Mean coefficient of gene differentiation （G sT ） between the populations was also high for both RAPD （G sT = 0.830） and cpssR markers （G sT = 0.735）, whereas the estimated gene flow was very low for RAPD （Nm = 0.102） and for cpssR （Nm = 0.179）. AMOvA analysis revealed that more genetic variation resided among the populations than within populations. significant differences （p 0.001） were observed between the populations and individuals within the populations. Cluster analysis of RAPD and cpssR data using UPGMA algorithm based on Nei’s genetic similarity matrix placed the 50 populations into two main clusters. The implication of the results of this study in devising strategy for conservation of A. calamus is discussed.

Animal models of human colorectal cancer:Current status, uses and limitations

AIM: To make orthotopic colon cancer murine modelsa more clearly understood subject. The orthotopic tumor models have been found to be more relevant in replicating the human disease process as compared to heterotopic models, many techniques for making orthotopic colorectal murine models have been ***: We evaluated the current literature for various reported orthotopic colon cancer models to understand their techniques, advantages and limitations. An extensive literature review was performed by searching the National Library of Medicine Database(Pub Med) using Me sH terms animal model; colon cancer; orthotopic model; murine model. Twenty studies related to colon cancer orthotopic xenograft model were evaluated in detail and discussed here. REsULTs: The detailed analysis of all relevant reports on orthotopic model showed tumor take rate between 42%-100%. While models using the enema technique and minimally invasive technique have reported development of tumor from mucosa with tumor take rate between 87%-100% with metastasis in 76%-90%.CONCLUsION: Over the years, the increased understanding of the murine models of human colon cancer has resulted in the development of various models. Each reported model has some limitations. These latest models have opened up new doors for continuing cancer research for not only understanding the colon cancer pathogenesis but also aid in the development of newer chemotherapeutic drugs as they mimic the human disease closely.

BACkGROUND The management of high-grade pancreatic trauma is *** To review our single-institution experience on the surgical management of blunt and penetrating pancreatic *** A retrospective review of records was performed on all patients undergoing surgical intervention for high-grade pancreatic injuries [American Association for the surgery of Trauma(AAsT) Grade Ⅲor greater] at the Royal North shore Hospital in sydney between January 2001 and December 2022. Morbidity and mortality outcomes were reviewed, and major diagnostic and operative challenges were *** Over a twenty-year period, 14 patients underwent pancreatic resection for highgrade injuries. seven patients sustained AAsT Grade Ⅲinjuries and 7 were classified as Grades Ⅳ or Ⅴ. Nine underwent distal pancreatectomy and 5 underwent pancreaticoduodenectomy(PD). Overall, there was a predominance of blunt aetiologies(11/14). Concomitant intra-abdominal injuries were observed in 11 patients and traumatic haemorrhage in 6 patients. Three patients developed clinically relevant pancreatic fistulas and there was one in-hospital mortality secondary to multi-organ failure. Among stable presentations, pancreatic ductal injuries were missed in two-thirds of cases(7/12) on initial computed tomography imaging and subsequently diagnosed on repeat imaging or endoscopic retrograde cholangiopancreatography. All patients who sustained complex pancreaticoduodenal trauma underwent PD without mortality. The management of pancreatic trauma is evolving. Our experience provides valuable and locally relevant insights into future management *** We advocate that high-grade pancreatic trauma should be managed in high-volume hepatopancreato-biliary specialty surgical units. Pancreatic resections including PD may be indicated and safely performed with appropriate specialist surgical, gastroenterology, and interventional radiology support in tertiary centres.

This work was aimed to investigate the changes brought about in the polymer polycarbonate irradiated to different doses of γ-radiation. Yellowing of the samples with the increase of γ-absorbed dose was observed. The changes in the optical properties were studied by recording Uv-visible absorbance spectra of the pristine and irradiated polycarbonate films. A simultaneous coexistence of direct and indirect band gaps was observed. The indirect band gap values were found lower in comparison to the corresponding values of direct band gap in the pristine and γ-irradiated poly-carbonate. Both types of the optical band gap energies had decreasing tendency with the increasing γ-radiation dose. Urbach energy was also determined from the tail of absorption edge which was found to have increasing tendency with progressive γ-radiation dose. Increase in carbon cluster size with the increasing γ absorbed dose was also shown. This increase in the number of carbon atoms (N) in a cluster can be correlated to the optical energy band gap (Eg). Moreover, the FTIR spectra of pristine and irradiated PC samples suggest chain scissoring with apparently the elimination of carbon di/monoxide.
