Transcriptional co-activators: emerging roles in signaling pathways and potential therapeutic targets for diseases

Specific cell states in metazoans are established by the symphony of gene expression programs that necessitate intricate synergic interactions between transcription factors and the *** of these regulatory molecules is associated with cell state transitions,which in turn is accountable for diverse maladies,including developmental disorders,metabolic disorders,and most significantly,cancer.A decade back most transcription factors,the key enablers of disease development,were historically viewed as‘undruggable’;however,in the intervening years,a wealth of literature validated that they can be targeted indirectly through transcriptional co-activators,their confederates in various physiological and molecular *** co-activators,along with transcription factors,have the ability to initiate and modulate transcription of diverse genes necessary for normal physiological functions,whereby,deregulation of such interactions may foster tissue-specific disease ***,it is essential to analyze how these co-activators modulate specific multilateral processes in coordination with other *** proposed review attempts to elaborate an in-depth account of the transcription co-activators,their involvement in transcription regulation,and context-specific contributions to pathophysiological *** review also addresses an issue that has not been dealt with in a comprehensive manner and hopes to direct attention towards future research that will encompass patient-friendly therapeutic strategies,where drugs targeting co-activators will have enhanced benefits and reduced side *** insights into currently available therapeutic interventions and the associated constraints will eventually reveal multitudes of advanced therapeutic targets aiming for disease amelioration and good patient prognosis.

Persistent gastritis induced by Helicobacter pylori(***)is the strongest known risk factor for gastric cancer(GC).*** is prevalent in about 50%world's population,while it causes cancer in less than 2%of exposed individuals.1 Our studies found that *** infection can induce oncogenic properties in AGS cells by deregulating multiple factors associated with the cell cycle,apoptosis,and other important events associated with cancer progression.

Dynamics of the phase-change material GeTe across the structural phase transition

Results of inelastic neutron scattering experinients and ab initio molecular dynamics simulations for GeTe 一 the parent compound of phase-change materials are reported. The inelastic neutron spectra of GeTe powder samples have been determined in the temperature range extending from 300 to 700 K. The phonon peaks undergo thermal shifts resulting from anharmonic effects being weaker for acoustic than optic modes. A small concentration of free charge carries arising from the presence of Ge-vacancies was found not to affect significantly the neutron weighted phonon densities of states of GeTe. The spectral pattern changes qualitatively across the structural phase transition, but the local structure of GeTe remains hardly affected, as confirmed by the analysis of temperature dependence of the pairdistribution function obtained from ab initio molecular dynamics investigations. The present theoretical studies support in a wide extent our experimental observations and also those provided by local probe methods.

Objective:To evaluate the antioxidant activity of aqueous extract of Atoringa oleifeta ***) young leaves by in vivo as well as in vitro ***:In vitro study included estimation of total phenolic,total ilavonol,total flavonoid and total antioxidant power(FRAP assay).Tn addition, in vivo study was done with the identified most effective dose of 200 nig/kg of its lyophilized powder on normal and diabetic *** effect on different oxidative free radical scavenging enzymes,viz,superoxide dismutase(SOD),catalase(CAT),glutathione-S-transferase(GST),lipid peroxide(LPO) contents were ***:Significant increase in activities of ***, GST while,a decrease in LPO content was ***,total phenolic,flavonoid and ilavonol contents in the extract were found to be 120 mg/g of CAK,40.5 mg/g of QEK and 12.12 mg/g of QE,*** the other *** assay results of *** leaves was(85.00±5.00)μM of Fe^+/g of extract ***:The significant antioxidant activities of *** leaves from both in vivo as well as in vitro studies suggests that the regular intake of its leaves through diet can protect normal as well as diabetic patients against oxidative damage.

Performance of the health system in China and Asia as measured by responsiveness

Objectives: To examine differences in health system responsiveness across different sectors in China and to compare to other Asian countries. Methods: The World Health Survey was implemented in a nationally representative sample in China and 10 additional Asian countries from 2002-2003. Face-to-face interviews were conducted to gather health care utilization and health systems responsiveness data. Results: Overall health system responsiveness in China was better for the inpatient than the outpatient health system. Differences were seen by domain, with prompt attention and respectful treatment performing better than the other domains. Differences in responsiveness were seen by socio- demographic characteristics, with women and younger respondents rating inpatient systems, whereas men and higher educated respondents rated outpatient systems, more responsive. Conclusions: As populations age, health care systems will come under more pressures – responsiveness can be used by governments to guide policy and system improvement efforts when resources are limited. In China, reforms might prioritize outpatient system responsiveness.

In vitro and in silico studies on fibrinolytic activity of nattokinase： A clot buster from Bacillus sp.

BACKGROUND： Nattokinase （NK） is a serine protease enzyme of the subtilisin family. It exhibits a strong fibrinolytic activity. The fibrinolytic enzymes from Bacillus sp. have attracted interest as thrombolytic agents because of their efficiency in the fibrinolytic process including plasmin activation. METHODS： In the present study, VIT garden soil was collected and subjected to isolation process in order to screen for the NK production. Screening for NK enzyme was performed by radial caseinolytic assay. The production of NK enzyme was done in two different production medium for comparative studies. The NK enzyme was purified by gel permeation chromatography. The activity of the purified NK was checked by clot lysis and casein digestion assay. To investigate the structural basis of NK and fibrinogen interaction and also to identify the best binding mode, molecular dynamics and docking studies were performed. RESULTS： Based on the morphological and biochemical characterization, the isolate was identified as Bacillus sp. The overall purification fold of NK was about 3 with the specific activity of 664U/mg and 9.9% yield. Homogeneity of the purified enzyme was analyzed and confirmed by the single band obtained in SDS-PAGE. Molecular weight of the purified protease was estimated as 25 kDa. Purified NK enzyme exhibited 97% of effective clot lysis activity. The NK was docked in to the knob region of the fibrinogen at its binding site using Dock server. A total of 26 residues of fibrinogen and 29 residues of NK constitute the interface region. However, 9 residues offibrinogen （THR238, MET264, LYS266, ARG275, THR277, ALA279, ASN308, MET310, and LYS321） and 8 residues ofNK （GLY61, SER63, THR99, PHE189, LEU209, TYR217, ASN218, and MET222） are involved in intact binding. CONCLUSIONS： A significant amount of NK enzyme was obtained from Bacillus sp. Regional planning starts with the basic assumption of the existence of political,geographic, economic,and social regions within a nation or a group of *** and soil differences are pronounced in different parts of a country demanding different types of economic and social *** reasons,political situation,language differences,ethnic diversities and most important the economic considerations all dictate that we choose a smaller area and decide the optimum planning *** the beginning it may appear that this policy of treating a portion of the space(district,metropolitan area,river basin,State or a combination of States,etc.) as the proper planning unit goes against the very formulation of overall national plans and the concept of national *** this does not have to be that way.A national plan cannot take into account the best possible strategy for regional development in its broad *** the other hand,a regional plan by its very definition possesses some question of inter-regional conflict. Whereas these conflicts are often over-emphasized and used against the concept and implementation or regional plans,enough is not said about how we can resolve these conflicts through inter-regional cooperation and at the same time maximize the potentialities of development of respective *** is true the benefits of this type of cooperation are often preached by politicians in different *** in reality it is seldom pursued primarily due to the absence of scientific studies which can show how this cooperation will lead to increased benefits for the interested regions and the nation as a *** objective of this paper is to discuss some of the analytical techniques that can be applied for such a purpose.

The objective of this presentation is to discuss the impact of Globalization on Urban-Regional system in the developing countries. Although the emphasis will be more on the urban sector, some observation will be made on the rural sector since both of them are intricately related. I shall start with urban growth patterns of different cities in Asia, Africa and Latin America vis-a-vis cities in Europe and U.S. More emphasis will be given to World Cities/Mega Cities and the theoretical reasons of the growth and decay of these cities will be given in the framework Paul Krugman's theory of New Geography. Then I shall address the issue of the impact of globalization on the cultural, social, economic and political characteristics of this transformation particularly for developing countries. I shall also discuss how knowledge and information system affects this transformation. Special attention will be paid on inequality and environmental degradation caused in urban center by globalization. Some recommendation will be given how to change the governance, decision making process, institutions and use international and inter regional co-operation to mitigate the negative externality and increase the benefit of globalization I shall refer to some theoretical and quantitative techniques in Urban and Regional Science which can be used for analyzing this transformation and forecasting the future trend.

工程师要求在使用混凝土骨料之前通过一些快速的方法对骨料的碱-硅反应进行测试。调查发现,存在两种截然不同的测试方法:第一种在较短的时间内查明骨料的碱-硅反应;第二种对骨料-水泥反应产生的长期膨胀进行评估,并设定可接受范围。标准、规范常忽视它们的不同之处而只有一种方法被详细说明。众所周知,当混凝土置于强碱溶液中,添加适量的火山灰可抑制因碱-硅反应引起的混凝土膨胀。许多评估骨料碱-硅反应的可用方法都被实践证明。在本文中,我建议从碱-硅反应及膨胀的机理来检验这些测试方法,测试重点应当放在骨料碱-硅反应自身上而非可接受范围上。碱-硅反应及产生膨胀压力的化学机理表明,将试样放在适量离子浓度溶液中[最好是Ca（OH）2溶液]最有利于测试,可通过适量添加中性离子（如碱盐）制得高浓度溶液。从碱-硅反应机理中获得一种简单易行的骨料测试评价方法,该方法已通过丹尼斯骨料测试并且制定了标准。初步实验表明,日本中性长石、英国杂砂岩、瑞士流纹岩、挪威斑岩和比利时硅化石灰石的反应在24h内可检测完成,这是检测骨料碱-硅反应最快方法之一。
