Assessment of spermatogenesis in camels via seminiferous tubule staging and testosterone profiling

目标树经营对红锥生长及根际土壤养分与酶活性的影响

土壤养分和酶是影响林木生长的重要因素，林木生长速率可以直接反映不同经营模式实施的效果。研究红锥在不同目标树经营密度下根际土壤养分和酶之间的关系及对林木生长的影响，可为红锥大径材的经营培育提供参考依据。以13年生的红锥人工林为研究对象，采取目标树经营密度为60 株/hm2、120 株/hm2、180 株/hm2、传统间伐和不间伐5种不同处理，分析经营4年后的红锥生长及根际土壤中养分和酶的影响。结果显示：1.与传统间伐相比，120 株/hm2的目标树密度显著提高了红锥4年的胸径和单株材积生长量。2.间伐能提高红锥根际土壤养分含量及酶活性，但会导致pH值和全氮下降。其中目标树密度为120 株/hm2下红锥根际土壤中的有机质、速效氮、全磷、速效磷含量及过氧化氢酶、蔗糖酶和脲酶活性最高且显著高于传统间伐，分别高出23.61%、37.67%、57.89%、50.39%、14.12%、28.81%和34.21%。酸性磷酸酶活性在60 株/hm2的目标树密度中最高。全钾和速效钾含量在传统间伐中最高。3.相关性分析和通径分析表明：土壤养分与土壤酶密切相关，其中有机质、速效氮、全磷和速效磷与胸径和材积生长量还呈现出显著的正相关。综上所述，目标树经营有效释放了红锥的生长空间，有利于根际土壤养分积累和提高酶活性，从而加快红锥生长速度，其中目标树密度为120 株/hm2的经营效果最佳。

Background The prevalence of understory removal and anthropogenic nitrogen(N) deposition has significantly altered the ecological processes of forest ecosystems at both regional and global scales. However, it remains a pressing challenge to understand how N deposition and understory removal affect leaf nutrient dynamics, nutrient resorption, litter decomposition, and their linkages for better managing forest ecosystems under nutrient imbalances induced by N enrichment. To address this research gap, a field manipulation experiment was carried out in a subtropical Cunninghamia lanceolata plantation with four treatments including: control(CK), canopy N addition(CN), understory removal(UR), and canopy N addition plus understory removal(CN × UR). Green and senesced leaf N and phosphorus(P) concentrations, N and P resorption efficiencies, litter decomposition, and their correlations were *** The results revealed that the average N concentrations of green early and late leaves in UR were increased by 6.61 and 18.89% compared to CK. UR had the highest whereas CN had the lowest P concentrations in green leaves across the two sampling seasons. Following this, UR, leaf type, season, and their interactions significantly affected leaf N, P, and N:P(P < 0.05). The highest leaf N resorption(32.68%) and P resorption efficiencies(63.96%) were recorded in UR. Litter decomposition was significantly retarded in UR(P < 0.01) relative to CN. The regression analysis demonstrated that leaf nutrient status was significantly interconnected with leaf nutrient resorption efficiencies. 植物入侵可通过正或负的植物-土壤反馈效应改变土壤的生物和非生物性质,从而影响入侵栖息地的土壤理化性质。许多入侵物种的凋落物分解可增加土壤养分,降低本地植物多样性,并导致进一步的植物入侵。关于入侵植物凋落物在不同土壤类型及深度分解及反馈效应的研究依然很少。本研究旨在明确入侵植物南美蟛蜞菊(Sphagneticola trilobata)凋落物在不同土壤类型和不同土壤深度条件下的分解情况及其对本地近缘植物蟛蜞菊(***)生理生长的影响。将装有南美蟛蜞菊凋落物的尼龙袋加入到不同深度(即0、2、4和6 cm)的砂土、营养土和粘土中,经6个月的分解后,回收凋落物袋并计算分解速率,随后在凋落物分解处理后的土壤中种植本地蟛蜞菊,并在生长期结束时测量其生理生态指标。研究结果表明,所有处理土壤类型中,凋落物在土壤深度为2和4 cm处分解后显著增加了土壤养分,而对本地蟛蜞菊的叶片叶绿素、叶氮含量等生长指标表现为负效应。因此,入侵植物南美蟛蜞菊凋落物分解对土壤养分表现为正的反馈效应,而对本地植物蟛蜞菊的生长表现为负效应。我们的研究结果还表明,入侵植物的凋落物分解对土壤和本地物种的影响还因凋落物分解所在的土壤深度而显著不同。未来的研究应侧重于入侵栖息地中更多本地和入侵物种的植物-土壤反馈效应,以及更多土壤类型和土壤深度的入侵植物凋落物效应。

Determination of Chikungunya Virus RNA by Reverse Transcription Loop-Mediated Isothermal Amplification (RT-LAMP) Assay

As a matter of fact, infectious diseases hamper everyone’s life and produce a lifelong threat to everyone neutral of age, sex, lifestyle and socio-economic status. Nowadays, come into the sight of Chikungunya viral (CHIKV) infection injured many Asian and African countries, also deliberated threat in rising countries and also low socio-economic countries. CHIKV is a positive-sense, enveloped single-stranded, RNA virus belonging to the genus Alphavirus, family Togaviridae. As the Dengue & Chikungunya viruses are spread simultaneously at the same time, so it is tough to identify them. In our resource-limited countries, swift detection of CHIKV by RT-LAMP is the simplest molecular technique in low-equipment settings without the use of any expensive decoration. Heat-treated centrifuged and uncentrifuged samples were used in this study and they showed the same result (100%). Different instruments like heat block, water bath, conventional thermal cycler & real-time thermal cycler were used to amplify the CHIKV RNA and they indicated that 100% samples were identified by all four instruments. The amplified products were visualized by turbidity test, color change by HNB, step-ladder band pattern in agarose gel electrophoresis and amplification curve in real-time thermal cycler naked eye, here the results also showed 100% samples were determined by all visualized methods. Reverse transcription loop-mediated isothermal amplification (RT-LAMP) is a recent technique for amplifying RNA under limited temperature, with high tangibility, quickness and competency. To identify the CHIKV RNA Reverse transcription loop-mediated isothermal amplification was fabricated and validated, and the results were also compared with reverse transcription polymerase chain reaction (RT-PCR). The sensitivity was 95.71% and specificity was 100%, these results indicate that RT-LAMP is a feasible method for quick detection of CHIKV RNA.
