Potato is the thirdmost important food crop in *** pathogens threaten sustainable crop production but can be controlled,in many cases,through the deployment of disease resistance genes belonging to the family of nucleotide-binding,leucine-rich-repeat(NLR)*** identify effective disease resistance genes in established varieties,we have successfully established SMRT-AgRenSeq in tetraploid potatoes and have further enhanced the methodology by including dRenSeq in an approach that we term *** inclusion of dRenSeq enables the filtering of candidates after the association analysis by establishing a presence/absence matrix across resistant and susceptible varieties that is translated into an F1 *** a SMRT-RenSeq-based sequence representation of the NLRome from the cultivar Innovator,SMRT-AgRenSeq-d analyses reliably identified the late blight resistance benchmark genes Rpi-R1,Rpi-R2-like,Rpi-R3a,and Rpi-R3b in a panel of 117 varieties with variable phenotype *** benchmark genes were identified with an F1 score of 1,which indicates absolute linkage in the *** method also identified nine strong candidates for Gpa5 that controls the potato cyst nematode(PCN)species Globodera pallida(pathotypes Pa2/3).Assuming that NLRs are involved in controlling many types of resistances,SMRT-AgRenSeq-d can readily be applied to diverse crops and pathogen systems.

Predicting Ailanthus altissima presence across a managed forest landscape in southeast Ohio

Background: The negative impacts of the exotic tree, Ailanthus altissima(tree-of-heaven, stink tree), is spreading throughout much of the Eastern United States. when forests are disturbed, it can invade and expand quickly if seed sources are ***: we conducted studies at the highly dissected Tar Hollow State Forest(THSF) in southeastern Ohio USA,where Ailanthus is widely distributed within the forest, harvests have been ongoing for decades, and prescribed fire had been applied to about a quarter of the study area. Our intention was to develop models to evaluate the relationship of Ailanthus presence to prescribed fire, harvesting activity, and other landscape characteristics, using this Ohio location as a case study. Field assessments of the demography of Ailanthus and other stand attributes(e.g., fire, harvesting, stand structure) were conducted on 267 sample plots on a 400-m grid throughout THSF,supplemented by identification of Ailanthus seed-sources via digital aerial sketch mapping during the dormant season. Statistical modeling tools Random Forest(RF), Classification and Regression Trees(CART), and Maxent were used to assess relationships among attributes, then model habitats suitable for Ailanthus ***: In all, 41 variables were considered in the models, including variables related to management activities, soil characteristics, topography, and vegetation structure(derived from LiDAR). The most important predictor of Ailanthus presence was some measure of recent timber harvest, either mapped harvest history(CART) or LiDARderived canopy height(Maxent). Importantly, neither prescribed fire or soil variables appeared as important predictors of Ailanthus presence or absence in any of the models of the ***: These modeling techniques provide tools and methodologies for assessing landscapes for Ailanthus invasion, as well as those areas with higher potentials for invasion should seed sources become available. Adjuvant chemotherapy for intrahepatic cholangiocarcinoma: approaching clinical practice consensus?

Background:Intrahepatic cholangiocarcinoma(ICC)is rare with limited evidence-based *** retrospective study evaluates the use of chemotherapy in patients with resected ***:The Surveillance Epidemiology and End Results(SEER)program database was used to identify patients with resected *** were stratified by date of diagnosis(2000-2004,2005-2009,2010-2014),T,and N *** logistic regression models identified predictors of chemotherapy ***-Meier and Cox proportional hazard models were used to identify survival ***:One thousand and two hundred twenty-three patients met inclusion *** utilization increased over time(33%to 41%,P≤0.05).Chemotherapy use increased in lymph node(LN)positive patients[32%to 60%in 2010-2014;(P≤0.05)and T3/T4 disease(40%to 60%in 2010-2014;P≤0.01)],but not in patients with LN negative or T1/T2 *** positivity was associated with utilization of chemotherapy in 2005-2009 and *** survival increased from 32 to 41 months(P≤0.05).In LN positive patients,chemotherapy was associated with a decreased hazard ratio of death(P≤0.05)and T3/T4 disease was associated with an increased hazard ratio of death(P≤0.05).Conclusions:Adjuvant chemotherapy use in ICC has *** LN positive or patients with T3/T4 tumors are receiving chemotherapy,which may explain the improvement in overall survival.

Scale-up of biomass conversion using 1-ethyl-3-methylimidazolium acetate as the solvent

This scale-up study demonstrated the feasibility of an ionic liquid(IL)pretreatment process at 40 kg scale,using the IL 1-ethyl-3-methylimidazolium acetate([C2C1Im][OAc])as the *** pretreatment was followed by enzymatic hydrolysis through which the process efficiency for biomass conversion to monomeric sugars was *** results show that 43 wt%of switchgrass was dissolved in IL after 2 h of pretreatment at 160℃ with 15 wt%solid loading.A 120 h enzymatic hydrolysis of the pretreated switchgrass results in 96%glucan and 98%xylan conversion.[C2C1Im][OAc]pretreatment has been successfully scaled up to 40 kg with improved sugar titers and yields relative to bench scale(6 kg).The mass flow of the overall process was established and the major scale-up challenges of the process were identified.

In recent years,there were two reported outbreaks of food borne illness associated with *** presence of melamine and its related compounds in milk,feed,and other foods has resulted in the need for reliable methods for the detection and accurate quantification of this class of *** sample pretreatment for melamine in a complex matrix usually involves a liquid extraction by a polar solvent,followed by a further clean-up with solid phase *** of melamine and related compounds are commonly carried out by liquid or gas chromatographic methods conjugated with mass *** innovative screening methods,which use antibodies,molecularly imprinted polymers,capillary electrophoresis,and gold nanoparticles,are also used to develop assays and biosensors to ***,many of these methods have been hindered by matrix effects,the solubility of melamine-cyanuric acid complex,and background *** article reviews recent developments for detecting melamine and discusses future directions.

Carbon pool trends and dynamics within a subtropical peatland during long-term restoration

Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is a novel beta-coronavirus that has emerged and caused a global pandemic of coronavirus disease 2019(COVID-19).1,2 Clinical manifestations of COVID-19 are age dependent,with children appearing to be less vulnerable than adults to severe disease.3,4 It has been reported that a subset of children and adults without previous infection had higher levels of humoral and cellular immunity to SARS-CoV-2,which may influence disease severity.5,6,7,8 Another study found that children who had never been exposed to SARS-CoV-2 had higher levels of cross-reactive antibodies to the spike glycoprotein subunit S2 of SARS-CoV-2 than adults.9 These findings raise the hypothesis that children may have a higher degree of preexisting cross-reactive immunity that provides some level of protection from severe *** this study,we determined antibody responses to SARS-CoV-2 in adults after infection and compared these responses in both children and adults with no prior exposure to ***,we mapped antibody epitopes elicited after infection,and cross-reactive antibody epitopes without prior SARS-CoV-2 infection,at high resolution using SARS-CoV-2 spike protein peptide arrays.

以花生[野生种(Arachis helodes)和栽培品种(Arachis hypogaea ***-5)],小麦(Triticum aestivum *** King)和高粱(Sorghum bicolor ***-2)的幼苗叶子为材料,在持续干燥脱水下(空气中)跟踪测定其叶绿素荧光诱导曲线的变化,发现曲线变化可分为三个阶段:1)开始荧光升高形成峰型(Peak pattern)荧光曲线;2)荧光继续攀升但峰形渐失,变为高原型(Plateau pattern)荧光曲线;3)继续脱水荧光跌落而形成平台型(Platform pattern)曲线。这三个变化阶段不仅存在于陆生植物(如作物)中,而且也证明存在于海藻中,故十分可能是植物界的普遍规律。重要发现在于:荧光跌落前和跌落后的植物光合器处两种完全不同的状态,跌落之前荧光的变化在复水时可以得到恢复,而跌落后则一般不能恢复,表明此时光合器已发生了不可逆的变化(伤害)。我们推测荧光的跌落或粹灭可能是光合膜上的蛋白复合体结构发生了变化。当荧光开始跌落时叶子的含水百分数是一个有价值的生理指标,我们称为临界含水百分数
