Objective Intestinal fibrosis is a refractory complication of inflammatory bowel disease(IBD).Tumor necrosis factor ligand-related molecule-1A(TL1A)is important for IBD-related intestinal fibrosis in a dextran sodium sulfate(DSS)-induced experimental colitis *** study aimed to explore the effects of TL1A on human colonic *** A trinitrobenzene sulfonic acid(TNBS)-induced experimental colitis model of LCK-CD2-TL1A-GFP transgenic(Tg)or wild-type(WT)mice was established to determine the effect and mechanism of TL1A on intestinal *** human colonic fibroblast CCD-18Co cell line was treated concurrently with TL1A and human peripheral blood mononuclear cell(PBMC)*** proliferation and activation of CCD-18Co cells were detected by BrdU assays,flow cytometry,immunocytochemistry and Western *** metabolism was tested by Western blotting and real-time quantitative polymerase chain reaction(RT-qPCR).Results The level of collagen metabolism in the TNBS+ethyl alcohol(EtOH)/Tg group was greater than that in the TNBS+EtOH/WT *** growth factor-β1(TGF-β1)and p-Smad3 in the TNBS+EtOH/Tg group were upregulated as compared with those in the TNBS+EtOH/WT *** proliferation of CCD-18Co cells was promoted by the addition of human PBMC supernatant supplemented with 20 ng/mL TL1A,and the addition of human PBMC supernatant and TL1A increased CCD-18Co proliferation by 24.4%at 24 ***1A promoted cell activation and increased the levels of COL1A2,COL3A1,and TIMP-1 in CCD-18Co *** of CCD-18Co cells with TL1A increased the expression of TGF-β1 and *** TL1A promotes TGF-β1-mediated intestinal fibroblast activation,proliferation,and collagen deposition and is likely related to an increase in the TGF-β1/Smad3 signaling pathway.

Natural killer T cell lymphoma(NKTCL)is highly aggressive,with advanced stage patients poorly responding to intensive *** explore effective and safe treatment for newly diagnosed advanced stage NKTCL,we conducted a phase ll study of anti-metabolic agent pegaspargase plus PD-1 antibody sintilimab(NCT04096690).Twenty-two patients with a median age of 51 years(range,24-74)were enrolled and treated with induction treatment of pegaspargase 2500 IU/m2 intramuscularly on day 1 and sintilimab 200 mg intravenously on day 2 for 6 cycles of 21 days,followed by maintenance treatment of sintilimab 200 mg for 28 cycles of 21 *** complete response and overall response rate after induction treatment were 59%(95%Cl,43-79%)and 68%(95%Cl,47-84%),*** a median follow-up of 30 months,the 2 year progression-free and overall survival rates were 68%(95%Cl,45-83%)and 86%(95%Cl,63-95%),*** most frequently grade 3/4 adverse events were neutropenia(32%,n=7)and hypofibrinogenemia(18%,n=4),which were manageable and led to no discontinuation of *** proportion score of PD-L1,peripheral blood high-density lipoprotein cholesterol,and apolipoprotein A-l correlated with good response,while PD-1 on tumor infiltrating lymphocytes and peripheral Treg cells with poor response to pegaspargase plus sintilimab *** conclusion,the chemo-free regimen pegaspargase plus sintilimab was effective and safe in newly diagnosed,advanced stage *** lipid profle and immunosuppressive signature contributed to treatment resistance,providing an alternative therapeutic approach dual targeting fatty acid metabolism and CTLA-4 in NKTCL.

The aim of this study was to investigate the role of DOPS and DOPS100 in attenuating immune dysfunction in subfertile mice. The results of the study revealed several noteworthy findings. First， DOPS and DOPS100 treatments led to significant improvement in weight gain and reversal of fatigue-related behaviors compared to the normal group. Second， both DOPS and DOPS100 showed effectiveness in reducing immune organ swelling， modulating immunoglobulin （IgA and IgM） levels， and restoring complement （C3c and C4） levels. In addition， they demonstrated a significant ability to enhance the integrity of the intestinal mechanical barrier by differentially upregulating the tight junction proteins Occludin and ZO-1. In addition， it was found that DOPS100 specifically enhanced the CD4+-Th17/Treg immune axis in the gut， as evidenced by increased expression of Foxp3 as well as decreased expression of RORγt， and further modulation of IL-10， IL-22， and IL-17A expression levels of inflammatory factors. These findings collectively suggest that DOPS100 holds significant potential in improving sub-healthy status by repairing the intestinal barrier， restoring local immune homeostasis， and activating the intestinal immune regulatory network. The study's outcomes provide valuable insights into the therapeutic implications of DOPS and DOPS100 in addressing immune dysfunction in sub-healthy conditions.

The development of freestanding and binder-free electrode is an effective approach to perform the inherent capacity of active materials and promote the mechanism study by minimizing the interference from additives. herein, we construct a freestanding cathode composed of MoS3/PPy nanowires（NWs） deposited on porous nickel foam（NF）(MoS3/PPy/NF) through electrochemical methods, which can work efficiently as sulfur-equivalent cathode material for Li-S batteries. The structural stability of the MoS3/PPy/NF cathode is greatly enhanced due to its significant tolerance to the volume expansion of MoS3during the lithiation process,which we ascribe to the flexible 3D framework of PPy NWs, leading to superior cycling performance compared to the bulk-MoS3/NF reference. Eliminating the interference of binder and carbon additives, the evolution of the chemical and electronic structure of Mo and S species during the discharge/charge was studied by X-ray absorption near-edge spectroscopy（XANES）. The formation of lithium polysulfides was excluded as the driving cathode reaction mechanism, suggesting the great potential of MoS3as a promising sulfur-equivalent cathode material to evade the shuttle effect for Li-S batteries. The present study successfully demonstrates the importance of structural design of freestanding electrode enhancing the cycling performances and revealing the corresponding mechanisms.

BACKGROUND:Sepsis is one of the main causes of mortality in intensive care units(ICUs).Early prediction is critical for reducing *** approximately 36%of sepsis occur within 24 h after emergency department(ED)admission in Medical Information Mart for Intensive Care(MIMIC-IV),a prediction system for the ED triage stage would be *** methods such as the quick Sequential Organ Failure Assessment(qSOFA)are more suitable for screening than for prediction in the ED,and we aimed to fi nd a light-weight,convenient prediction method through machine ***:We accessed the MIMIC-IV for sepsis patient data in the *** dataset comprised demographic information,vital signs,and synthetic *** Gradient Boosting(XGBoost)was used to predict the risk of developing sepsis within 24 h after ED ***,SHapley Additive exPlanations(SHAP)was employed to provide a comprehensive interpretation of the model's *** percent of the patients were randomly selected as the testing set,while the remaining patients were used for training with 10-fold ***:For 10-fold cross-validation on 14,957 samples,we reached an accuracy of 84.1%±0.3%and an area under the receiver operating characteristic(ROC)curve of 0.92±*** model achieved similar performance on the testing set of 1,662 *** values showed that the fi ve most important features were acuity,arrival transportation,age,shock index,and respiratory ***:Machine learning models such as XGBoost may be used for sepsis prediction using only a small amount of data conveniently collected in the ED triage *** may help reduce workload in the ED and warn medical workers against the risk of sepsis in advance.

2023年是全面预算绩效管理改革的收官之年,基于“中央-省级”“省级—市级—县(区)级”的跨层视角,以各级政府发布的相关政策文本和官员的访谈记录为研究素材,构建“制度—组织—个体”的分析框架,剖析全面预算绩效管理改革在政策结构、推进模式与官员行为间的衔接机制,重点识别全面预算绩效管理改革在各层级之间形成的结构效率及其阻滞性因素。研究发现:全面预算绩效管理制度体系的“形”已经基本形成,各级政府“点”的行动也初具成效,但要进一步实现推进过程中的“形”“实”相符与“点”“面”相接,重点还需依托各省实际情况,突破跨层政策推进的衔接机制,提升内在结构效率。

Objective This study aimed to investigate the potential relationship between urinary metals copper(Cu),arsenic(As),strontium(Sr),barium(Ba),iron(Fe),lead(Pb)and manganese(Mn)and grip *** We used linear regression models,quantile g-computation and Bayesian kernel machine regression(BKMR)to assess the relationship between metals and grip *** In the multimetal linear regression,Cu(β=−2.119),As(β=−1.318),Sr(β=−2.480),Ba(β=0.781),Fe(β=1.130)and Mn(β=−0.404)were significantly correlated with grip strength(Phe results of the quantile g-computation showed that the risk of occurrence of grip strength reduction was−1.007(95%confidence interval:−1.362,−0.652;Phen each quartile of the mixture of the seven metals was *** kernel function regression model analysis showed that mixtures of the seven metals had a negative overall effect on grip strength,with Cu,As and Sr being negatively associated with grip strength *** the total population,potential interactions were observed between As and Mn and between Cu and Mn(P_(interactions) of 0.003 and 0.018,respectively).Conclusion In summary,this study suggests that combined exposure to metal mixtures is negatively associated with grip ***,Sr and As were negatively correlated with grip strength levels,and there were potential interactions between As and Mn and between Cu and Mn.
