glioblastoma,a rare,and highly lethal form of brain cancer,poses significant challenges in terms of therapeutic resistance,and poor survival rates for both adult and paediatric patients *** advancements in brain cancer research driven by a technological revolution,translating our understanding of glioblastoma pathogenesis into improved clinical outcomes remains a critical unmet *** review emphasises the intricate role of receptor tyrosine kinase signalling pathways,epigenetic mechanisms,and metabolic functions in glioblastoma tumourigenesis and therapeutic *** also discuss the extensive efforts over the past two decades that have explored targeted therapies against these *** therapeutic approaches,such as antibody-toxin conjugates or CAR T cell therapies,offer potential by specifically targeting proteins on the glioblastoma cell *** strategies incorporating protein-targeted therapy and immune-based therapies demonstrate great promise for future clinical ***,gaining insights into the role of cell-of-origin in glioblastoma treatment response holds the potential to advance precision medicine *** these challenges is crucial to improving outcomes for glioblastoma patients and moving towards more effective precision therapies.

An Adaptive Sequential Replacement Method for Variable Selection in Linear Regression Analysis

With the rapid development of DNA technologies, high throughput genomic data have become a powerful leverage to locate desirable genetic loci associated with traits of importance in various crop species. However, current genetic association mapping analyses are focused on identifying individual QTLs. This study aimed to identify a set of QTLs or genetic markers, which can capture genetic variability for marker-assisted selection. Selecting a set with k loci that can maximize genetic variation out of high throughput genomic data is a challenging issue. In this study, we proposed an adaptive sequential replacement (ASR) method, which is considered a variant of the sequential replacement (SR) method. Through Monte Carlo simulation and comparing with four other selection methods: exhaustive, SR method, forward, and backward methods we found that the ASR method sustains consistent and repeatable results comparable to the exhaustive method with much reduced computational intensity.

Comparison of REML and MINQUE for Estimated Variance Components and Predicted Random Effects

Linear mixed model (LMM) approaches have been widely applied in many areas of research data analysis because they offer great flexibility for different data structures and linear model systems. In this study, emphasis is placed on comparing the properties of two LMM approaches: restricted maximum likelihood (REML) and minimum norm quadratic unbiased estimation (MINQUE) with and without resampling techniques being included. Bias, testing power, Type I error, and computing time were compared between REML and MINQUE approaches with and without Jackknife technique based on 500 simulated data sets. Results showed that MINQUE and REML methods performed equally regarding bias, Type I error, and power. Jackknife-based MINQUE and REML greatly improved power compared to non-Jackknife based linear mixed model approaches. Results also showed that MINQUE is more time-saving compared to REML, especially with the use of resampling techniques and large data set analysis. Results from the actual cotton data analysis were in agreement with our simulated results. Therefore, Jackknife-based MINQUE approaches could be recommended to achieve desirable power with reduced time for a large data analysis and model simulations.

Beliefs and Practices Regarding Spirituality as a Healthcare Strategy

Objective: This study examined the perceptions and practices of medical personnel regarding the spirituality in the treatment management plan of their patients. Methods: A spirituality survey was administered to doctors and nurses, soliciting responses about their beliefs and willingness to include spirituality practices in treating patients within their facility. Respondents were asked to give their views on daily practices regarding the use of spirituality in patient care and patient response to treatment. Results: The doctors’ responses suggest that they believe that solutions to medical problems, patient recovery, and coping are related to mind and body stress issues that surgery and drugs may not necessarily cure. The emphasis on self-care for some of these medical problems can put the patient back in control and eliminate the use of excessive drugs in the treatment process. Chances for health and well being of patients are improved if good medical practices are combined with a strong mental and psychological disposition to provide a vital foundation for directly confronting diseases. Conclusion: Spiritual engagement helps to support a psychological outlook that is necessary for fighting diseases and coping adequately with life’s tragedies. This approach provides a well-balanced attack on disease and increases the patient’s chances of coping and surviving.

Tetracycline-inducible protein expression in pancreatic cancer cells: Effects of Capg overexpression

AIM: To establish stable tetracycline-inducible pancreatic cancer cell lines. METHODS: Suit-2, MiaPaca-2, and Panc-1 cells were transfected with a second generation reverse tetracycline-controlled transactivator protein (rtTA2S-M2), under the control of either a cytomegalovirus (CMV) or a chicken β-actin promoter, and the resulting clones were ***: Use of the chicken (β-actin) promoter proved superior for both the production and maintenance of doxycycline-inducible cell lines. The system proved versatile, enabling transient inducible expression of a variety of genes, including gST-P, CYP2E1, S100A6, and the actin capping protein, Capg. To determine the physiological utility of this system in pancreatic cancer cells, stable inducible Capg expressors were established. Overexpressed Capg was localised to the cytoplasm and the nuclear membrane, but was not observed in the nucleus. High Capg levels were associated with enhanced motility, but not with changes to the cell cycle, or cellular proliferation. In Capg-overexpressing cells, the levels and phosphorylation status of other actin-moduating proteins (Cofilin and Ezrin/Radixin) were not altered. However, preliminary analyses suggest that the levels of other cellular proteins, such as ornithine aminotransferase and enolase, are altered upon Capg induction. CONCLUSION: We have generated pancreatic-cancer derived cell lines in which gene expression is fully controllable.

Experience with Thoracic Aortic Stent-grafting at the Royal Brisbane and Women’s Hospital: Outcomes from the First Decade

Objectives: To describe thoracic endovascular aortic repair (TEVAR) outcomes at the Royal Brisbane and Women’s Hospital between 2001-2010. Design: Prospective cohort study. Results: We successfully treated 95 of 97 patients for a 98% procedural success rate. Of the treated patients, 68 (72%) were males and mean age was 61 ± 17 years. Average follow up was 3.6 ± 2.0 years. Pathologies treated were: dissection (n = 35), aneurysmal disease (n = 32), traumatic disease (n = 19), coarctation (n = 5) and miscellaneous (n = 4). There was one peri-procedural myocardial infarction. Renal complications occurred in 7 patients (3 with doubling creatinine and 4 requiring temporary renal replacement therapy). Stroke occurred in 2 patients and paresis in 2 patients (permanent in one). Six patients died during index hospitalization and 17 deaths occurred during follow-up, 2 of which were confirmed secondary to aortic pathology. Age (HR 1.08 per year, p ghteen of these re-interventions in 16 patients were related to the management of complications of TEVAR of which 13 were for endoleaks;eight type I, four type II, one type III. Conclusion: TEVAR can be used to treat thoracic aortic pathology but questions remain regarding long-term durability.
