B-cell and antibody responses to SARS-CoV-2:infection,vaccination,and hybrid immunity

The emergence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)in 2019 prompted scientific,medical,and biotech communities to investigate infection-and vaccine-induced immune responses in the context of this pathogen.B-cell and antibody responses are at the center of these investigations,as neutralizing antibodies(nAbs)are an important correlate of protection(COP)from infection and the primary target of SARS-CoV-2 vaccine *** addition to absolute levels,nAb longevity,neutralization breadth,immunoglobulin isotype and subtype composition,and presence at mucosal sites have become important topics for scientists and health policy *** recent pandemic was and still is a unique setting in which to study de novo and memory B-cell(MBC)and antibody responses in the dynamic interplay of infection-and vaccine-induced *** also provided an opportunity to explore new vaccine platforms,such as mRNA or adenoviral vector vaccines,in unprecedented cohort *** with the technological advances of recent years,this situation has provided detailed mechanistic insights into the development of B-cell and antibody responses but also revealed some unexpected *** this review,we summarize the key findings of the last 2.5 years regarding infection-and vaccine-induced B-cell immunity,which we believe are of significant value not only in the context of SARS-CoV-2 but also for future vaccination approaches in endemic and pandemic settings.

Identification and Characterization of Reduced Epicuticular Wax Mutants in Rice

Epicuticular wax forms the outermost protective barrier of the aerial surfaces of land plants, working in concert with other components of the plant cuticle to prevent uncontrolled loss of water and to provide protection against an array of external environmental stress. In this study, chemically- mutagenized populations of rice （Oryza sativa L.） derived from approximately 4 750 M2 families were screened for adhesion of water droplets resulting in a wet leaf/glossy （wig） phenotype. Mutants were identified in 11 independently-derived M2 families. Scanning electron microscopy analysis confirmed the association of the wig phenotype with changes in the epicuticular wax crystals of these plants. The phenotypes of five mutants （7-17A, 26.1, 524.2, 680.2, and 843.1） were confirmed to be the result of single recessive gene mutation. Evaluation of mutants from 3 （6-1A, 7-17A, and 11-39A） of 11 M2 families revealed significant reductions （〉 50%） in surface wax content and increases in cuticle membrane permeability.

Clinical strategies to enhance nerve regeneration

The slow rate of nerve regeneration after injury or recon- struction remains a clinical problem because it prohibits the timely reinnervation of distant target muscles be- fore the irreversible degeneration of the neuromuscular junction and breakdown of muscle tissue. As such, high （proximal） nerve injuries result in the incomplete recovery of motor function and poor functional outcomes despite current and timely surgical management. Experimentally, several strategies have been shown to enhance nerve regen- eration and improve functional recovery in animal models but translation to clinical practice has not been realized. Two potential treatments, tacrolimus （immunosuppres- sant） and electrical stimulation are commonly used for other reconstructive indications and as such, are both read- ily available clinically. There is some evidence, which will be reviewed in subsequent sections, that these approaches may also be useful in enhancing neuronal regeneration.

Enamel biomimetics—fiction or future of dentistry?

Tooth enamel is a complex mineralized tissue consisting of long and parallel apatite crystals configured into decussating enamel rods. In recent years, multiple approaches have been introduced to generate or regenerate this highly attractive biomaterial characterized by great mechanical strength paired with relative resilience and tissue compatibility. In the present review, we discuss five pathways toward enamel tissue engineering,(i) enamel synthesis using physico-chemical means,(ii) protein matrix-guided enamel crystal growth,(iii) enamel surface remineralization,(iv) cell-based enamel engineering, and(v) biological enamel regeneration based on de novo induction of tooth morphogenesis. So far, physical synthesis approaches using extreme environmental conditions such as ph, heat and pressure have resulted in the formation of enamel-like crystal *** methods relying on enamel proteins as templating matrices have aided the growth of elongated calcium phosphate crystals. To illustrate the validity of this biochemical approach we have successfully grown enamel-like apatite crystals organized into decussating enamel rods using an organic enamel protein matrix. Other studies reviewed here have employed amelogeninderived peptides or self-assembling dendrimers to re-mineralize mineral-depleted white lesions on tooth surfaces. So far, cell-based enamel tissue engineering has been hampered by the limitations of presently existing ameloblast cell lines. Going forward, these limitations may be overcome by new cell culture technologies. Finally, whole-tooth regeneration through reactivation of the signaling pathways triggered during natural enamel development represents a biological avenue toward faithful enamel regeneration. In the present review we have summarized the state of the art in enamel tissue engineering and provided novel insights into future opportunities to regenerate this arguably most fascinating of all dental tissues.

Lophozonia tree cavities used for nesting by Slender-billed Parakeets(Enicognathus leptorhynchus)in the central valley of southern Chile:a potentially vanishing keystone resource

Background: The Slender-billed Parakeet(Enicognathus leptorhynchus) is a psittacine endemic to southern Chile and an obligate secondary cavity-nester. In the central valley of southern Chile, most(94%) of the known Slender-billed Parakeet nests have occurred in large, mature southern beech(Lophozonia obliqua) trees(locally known as "pellines"). As relicts of the original old-growth forests of southern Chile, most pellines have been lost due to extensive landclearing throughout the region, potentially threatening long-term persistence of the Slender-billed ***: We conducted our study in the central valley of southern Chile, near the city of Osorno during three consecutive nesting seasons(November–January, 2008–2011). Nest trees used by Slender-billed Parakeets were located by direct observation of parakeet activities and through interviews with local residents, some of whom were former parrot nest poachers. Nest cavities were accessed, inspected and measured using single-rope climbing techniques. We report means, standard errors, 95% confidence intervals and ranges for 11 cavity-related variables. We also report clutch sizes encountered in active nests, and age estimates of nest trees based on known growth rates of Lophozonia trees in southern Chile. Linear regressions were used to evaluate potential relationships between cavity-related variables and clutch ***: We located and measured 38 Lophozonia tree cavities used for nesting by Slender-billed Parakeets. Compared to those used by other psittacines, nest trees were relatively large, averaging 30.4 breast height of 134.5 ter ± 1.1 m in height with a mean diameter at ± 4.7 cm. Based on estimated annual diameincrement, ages of nest trees ranged from approximately 209–485 years. Nest cavities entrances averaged 12.5 ± 0.9 m in height above ground level. Emotion recognition and inhibitory control in manifest and pre-manifest huntington’s disease: evidence from a new Stroop task
