Prediction of clinically actionable genetic alterations from colorectal cancer histopathology images using deep learning

BACKGROUND Identifying genetic mutations in cancer patients have been increasingly important because distinctive mutational patterns can be very informative to determine the optimal therapeutic strategy. Recent studies have shown that deep learning-based molecular cancer subtyping can be performed directly from the standard hematoxylin and eosin(H&E) sections in diverse tumors including colorectal cancers(CRCs). Since H&E-stained tissue slides are ubiquitously available, mutation prediction with the pathology images from cancers can be a time-and cost-effective complementary method for personalized *** To predict the frequently occurring actionable mutations from the H&E-stained CRC whole-slide images(WSIs) with deep learning-based *** A total of 629 CRC patients from The Cancer Genome Atlas(TCGA-COAD and TCGA-READ) and 142 CRC patients from Seoul st. Mary Hospital(SMH) were included. Based on the mutation frequency in TCGA and SMH datasets, we chose APC, KRAS, PIK3CA, SMAD4, and TP53 genes for the study. The classifiers were trained with 360 × 360 pixel patches of tissue images. Testicular fine-needle aspiration for the assessmen＇ of intratesticular hormone concentrations

Measurement of intratesticular sex steroid concentrations in men informs both the development of male hormonal contraceptives and the understanding of male infertility. Given the challenges of using invasive techniques to measure testicular hormone physiology, our group has used a minimally-invasive fine-needle aspiration technique to measure intratesticular hormones in normal healthy men. Herein, we present a post-hoc analysis of the safety and efficacy of testicular fine-needle aspiration （FNA） completed as part of six clinical trials. From 2001 through 2011, a total of 404 procedures were conducted among 163 research volunteers, 85.9% of which were successful in obtaining sufficient fluid for the measurement of intratesticular steroid concentrations. Pain was the most common side effect, with 36.8% of procedures associated with moderate procedural pain and 4.7% with severe procedural pain. Postprocedural pain was uncommon and abated within a few days. Mild local bruising occurred with 14.9% of procedures. Two serious adverse events （0.5%） required surgical intervention. The risk of an adverse event was not associated with age, body mass index, testicular size, or the volume of fluid aspirated. Testicular FNA to obtain fluid for measurement of intratesticular steroid concentrations frequently causes mild to moderate procedural pain, but serious adverse events occur rarely. Testicular FNA has been instrumental for defining human intratesticular hormone physiology and is a minimally-invasive, safe, effective method for obtaining fluid for research on testicular physiology and pathology.

Fatigue failures cost approximately 4% of the United states' gross domestic product(GDP). The design of highly fatigue-resistant materials is always in demand. Different from conventional strategies of alloy design, high-entropy alloys(HEAs) are defined as materials with five or more principal elements, which could be solid solutions. This locally-disordered structure is expected to lead to unique fatigue-resistant properties. In this review, the studies of the fatigue behavior of HEAs during the last five years are summarized. The four-point-bending high-cycle fatigue coupled with statistical modelling, and the fatigue-crack-growth behavior of HEAs, are reviewed. The effects of sample defects and nanotwins-deformation mechanisms on four-point-bending high-cycle fatigue of HEAs are discussed in detail. The influence of stress ratio and temperature on fatigue-crack-growth characteristics of HEAs is also discussed. HEAs could exhibit comparable or greater fatigue properties, relative to conventional materials. Finally,the possible future work regarding the fatigue behavior of HEAs is suggested.

Rapid climate change-induced collapse of hunter-gatherer societies in the lower Mississippi River valley between ca. 3300 and 2780 cal yr BP

Hunter-gatherer communities in the American Southeast reached an apogee of social and political complexity in the period between ca. 4200 and 3000 cal yr BP. In the lower Mississippi Valley(LMV) the Poverty Point culture defined this period of socio-political elaboration. However, following a significant period of climate change that led to exceptional flooding and a major reorganization of the course of the Mississippi River, this culture collapsed beginning ca. 3300–3200 cal yr BP and the LMV was abandoned for the subsequent 500 years. In this study, we use data from the jaketown site in the Yazoo Basin of west-central Mississippi to refine the chronology of the climate event that caused the collapse of the Poverty Point culture. A large flood buried Poverty Point-era occupation deposits at jaketown around 3310 cal yr BP. Lateral migration of the Mississippi River during flooding led to inundation of the Yazoo Basin and re-occupation of ancient river courses. A coarse sand stratum topped by a more than a meter-thick fining upward sediment package marks a crevasse deposit caused by a rupture of the natural levee at jaketown. This levee breach was part of a larger pattern of erratic flooding throughout the LMV and is associated with major landscape evolution and the abandonment of Poverty Point sites within the valley. Early Woodland peoples re-colonized the crevasse surface after ca. 2780 cal yr BP. Following this event, the jaketown site and the eastern Yazoo Basin witnessed a period of landscape stability that lasts to this day. These archaeological data demonstrate how climate change and natural disasters can lead to socio-political dissolution and reorganization even in relatively small-scale hunter-gatherer populations.
