The outer blood-retina barrier(oBRB),crucial for the survival and the proper functioning of the overlying retinal layers,is disrupted in numerous diseases affecting the retina,leading to the loss of the photoreceptors and ultimately of *** study the oBRB and/or its degeneration,many in vitro oBRB models have been developed,notably to investigate potential therapeutic strategies against retinal ***,to this day,most of these pathologies are untreatable,especially once the first signs of degeneration are *** cure those patients,a current strategy is to cultivate in vitro a mature oBRB epithelium on a custom membrane that is further implanted to replace the damaged native *** a description of the oBRB and the related diseases,this review presents an overview of the oBRB models,from the simplest to the most ***,we propose a discussion over the used cell types,for their relevance to study or treat the *** designed for in vitro applications are then examined,by paying particular attention to the design evolution in the last years,the development of pathological models and the benefits of co-culture models,including both the retinal pigment epithelium and the ***,this review focuses on the models developed for in vivo implantation,with special emphasis on the choice of the material,its processing and its characterization,before discussing the reported pre-clinical and clinical trials.

Percoll Gradient Optimization for Blood CD133+ Stem Cell Recovery

Circulating CD133+ stem cells from the peripheral blood have been shown to be able to differentiate into numerous cell lineages. However, adults have only a small number of these circulating stem cells. The aim of the present study was to assess a new isolation and enrichment technique for CD133+ stem cells from peripheral blood with the use of Percoll density gradients. Our results demonstrated the presence of two large mononuclear bands when whole blood was centrifuged with 48% and 50% Percoll concentrations. Flow cytometric analysis (FACS) revealed a major CD133+ enrichment at the 48% Percoll concentration in one of the two bands. Further culture of these cells resulted in the formation of multiple colony-forming units. Our results suggest an advantage from using a simple Percoll gradient for successful CD133+ cell recovery, which could aid in differentiation and transplantation protocols.

Machine learning (ML) is a software solution with the ability of making predictions without prior explicit programming, aiding in the analysis of large amounts of data. These algorithms can be trained through supervised or unsupervised learning. Cardiology is one of the fields of medicine with the highest interest in its applications. They can facilitate every step of patient care, reducing the margin of error and contributing to precision medicine. In particular, ML has been proposed for cardiac imaging applications such as automated computation of scores, differentiation of prognostic phenotypes, quantification of heart function and segmentation of the heart. These tools have also demonstrated the capability of performing early and accurate detection of anomalies in electrocardiographic exams. ML algorithms can also contribute to cardiovascular risk assessment in different settings and perform predictions of cardiovascular events. Another interesting research avenue in this field is represented by genomic assessment of cardiovascular diseases. Therefore, ML could aid in making earlier diagnosis of disease, develop patient-tailored therapies and identify predictive characteristics in different pathologic conditions, leading to precision cardiology.

Chemical and minero-petrographical changes on granulite rocks affected by weathering processes

The purpose of this work is to study the weathering processes of the granulite rocks of the Serre Massif (southern Calabria,Italy) using a multidisciplinary approach based on field studies,geochemical modeling,and minero?petrographical *** granulite rocks are plagioclase-rich with minor amphibole,clinopyroxene,orthopyroxene,biotite,and garnet and their texture are *** reaction path modeling was performed to simulate the evolution of groundwaters upon interaction with local granulite by means of the software package EQ3/6,version *** were performed in kinetic (time) mode under a closed system at a constant temperature of 11.5℃,(which reproduces the average temperature of local area) and fixing the fugacity of CO2 at 10-2.34 bar (mean value).During the most advanced stage of weathering the main mineralogical changes are: partial destruction and transformation of biotite and plagioclase associated with neoformation of ferruginous products and secondary clay minerals producing a change in the origin rock *** secondary solid phases observed during the geochemical modeling (kaolinite,vermiculite and ferrihydrite) are similar to those found in this natural ***,the soil-like material mainly characterized by mostly sand to gravel grain-size fractions is the final result of the weathering processes.

Effect of Sn precursor on the synthesis of SnO2 and Sb-doped SnO2 particles via polymeric precursor method

SnO2 and Sb-doped SnO2 particles were synthesized using the polymeric precursor method with different Sn salt precursors： SnCl2.2H2O, SnCl4.5H20, or Sn citrate. Sb2O3 was used as the precursor of Sb, and the molar ratio of nsn：nsb was held constant. FTIR and TGA/DTA were used to examine the influence of the Sn precursor on the formation and thermal decomposition of the Sn and Sn-Sb complexes. The calcination products obtained from heating the Sn and Sn-Sb complexes at 500℃ in air were analyzed using XRD and TEM analysis. The results revealed that the SnO2 and Sb-doped SnO2 formation temperatures depended on the nature of the Sn precursor. The calcination products were found to be SnO2 and Sb-doped SnO2 particles, which crystallized in a tetragonal cassiterite structure with a highly preferred （110） planar orientation. The Sn precursor and the presence of Sb in the SnO2 matrix strongly influenced the crystallinity and lattice parameters.

Rat cardiomyocyte H9c2(2-1)-based sulforhodamine B assay as a promising in vitro method to assess the biological component of effluent toxicity

The treatment of wastewaters is crucial to maintain the ecological status of receiving waters,and thereby guarantee the protection of aquatic life and human *** quality evaluation is conventionally based on physicochemical parameters,but increasing attention has been paid to integrate physicochemical and biological ***,the regulatory use of fish in biological testing methods has been subject to various ethical and cost concerns,and in vitro cell-based assays have thus become an important topic of ***,the present study intends:(a) to evaluate the efficiency of two different sample pre-concentration techniques (lyophilisation and solid phase extraction) to assess the toxicity of municipal effluents on rat cardiomyoblast H9c2(2-1) cells,and (b) maximizing the use of the effluent sample collected,to estimate the environmental condition of the receiving *** gathered results demonstrate that the H9c2(2-1) sulforhodamine B-based assay is an appropriate in vitro method to assess biological effluent toxicity,and the best results were attained by lyophilising the sample as *** to its response,the H9c2(2-1) cell line might be a possible alternative in vitro model for fish lethal testing to assess the toxicity of municipal *** physicochemical status of the sample suggests a high potential for eutrophication,and iron exceeded the permissible level for wastewater discharge,possibly due to the addition of ferric chloride for wastewater *** general,the levels of carbamazepine and sulfamethoxazole are higher than those reported for other countries,and both surpassed the aquatic protective values for long-term exposure.

Differential diagnosis of uterine vascular anomalies:Uterine pseudoaneurysm as a cause of massive hemorrhage

In this article,we comment on the paper by Kakinuma et al published *** focus specifically on the diagnosis of uterine pseudoaneurysm,but we also review other uterine vascular anomalies that may be the cause of life-threating hemorrhage and the different causes of uterine *** artery pseudoaneurysm is a complication of both surgical gynecological and nontraumatic *** hemorrhage is the consequence of the rupture of the *** artery pseudoaneurysm can develop after obstetric or gynecological procedures,being the most frequent after cesarean or vaginal deliveries,curettage and even during ***,there are several cases described unrelated to pregnancy,such as after conization,hysteroscopic surgery or laparoscopic *** is the clinical manifestation and it can be life-threatening so suspicion of this vascular lesion is essential for early diagnosis and ***,there are other uterine vascular anomalies that may be the cause of severe hemorrhage,which must be taken into account in the differential *** tomography angiography and embolization is supposed to be the first therapeutic option in most of them.

Bioluminescent nanopaper for rapid screening of toxic substances

Environmental pollution is threatening human health and ecosystems as a result of modern agricultural techniques and industrial progress. A simple nanopaper-based platform coupled with luminescent bacteria Aliivibrio Jischeri （A. Jischeri） as a bio-indicator is presented here, for rapid and sensitive evaluation of contaminant toxicity. When exposed to toxicants, the luminescence inhibition of A. Jischeri-decorated bioluminescent nanopaper （BLN） can be quantified and analyzed to classify the toxicity level of a pollutant. The BLN composite was characterized in terms of morphology and functionality. Given the outstanding biocompatibility of nanocellulose for bacterial proliferation, BLN achieved high sensitivity with a low cost and simplified procedure compared to conventional instruments for laboratory use only. The broad applicability of BLN devices to environmental samples was studied in spiked real matrices （lake and sea water）, and their potential for direct and in situ toxicity screening was demonstrated. The BLN architecture not only survives but also maintains its function during freezing and recycling processes, endowing the BLN system with competitive advantages as a deliverable, ready-to-use device for large-scale manufacturing. The novel luminescent bacteria-immobilized, nanocelullose-based device shows outstanding abilities for toxicity bioassays of hazardous compounds, bringing new possibilities for cheap and efficient environmental monitoring of potential contamination.

Rockfall localization from seismic polarization considering multiple triaxial geophones and frequency bands

Boulder/rock mass movements generate ground vibrations that can be recorded by geophone ***,there are two methods applied to rockfall trajectory reconstruction or rockfall seismic *** method uses seismic wave arrival times and is achieved by minimizing the differences in signal arrival times between multiple stations by grid map *** other method uses seismic polarization and is achieved by calculating eventsource back azimuths from the seismic polarizations of rockfall *** this study,we proposed the use of an overdetermined matrix for joint localization based on the polarization *** overdetermined matrix considers the contributions of all geophones in the network,and at each geophone is assigned a different weight according to the recorded signal qualities and the reliability of the calibrated back *** method shows a great advantage relative to the case in which only two sensors are ***,we suggested three marker parameters for proper frequency band selection in back azimuth calculations:energy,rectilinearity,and a special permanent frequency band(SPF).We found that the back azimuths calculated with energy and an SPF are generally close to the real back azimuths measured in the field,while the SPF is limited by seismic attenuation due to a long-distance *** localization results of rockfalls were validated by using field camera videos and in situ *** typical cases and 43 artificially released rockfalls are presented in this *** proposed method provides an interesting way to locate rockfall events and track rockfall trajectories and avoids the difficulties of obtaining accurate arrival times,as required by the arrival times method.
