Knockout of butyrophilin subfamily 1 member A1(BTn1A1) alters lipid droplet formation and phospholipid composition in bovine mammary epithelial cells

Background: Milk lipids originate from cytoplasmic lipid droplets(LD) that are synthesized and secreted from mammary epithelial cells by a unique membrane-envelopment process. Butyrophilin 1 A1(BTn1 A1) is one of the membrane proteins that surrounds LD, but its role in bovine mammary lipid droplet synthesis and secretion is not well ***: The objective was to knockout BTn1 A1 in bovine mammary epithelial cells(BMEC) via the CRISPR/Cas9 system and evaluate LD formation, abundance of lipogenic enzymes, and content of cell membrane phospholipid(PL) species. Average LD diameter was determined via Oil Red O staining, and profiling of cell membrane phospholipid species via liquid chromatography-tandem mass spectrometry(LC-MS/MS).Results: Lentivirus-mediated infection of the Cas9/sg RnA expression vector into BMEC resulted in production of a homozygous clone BTn1 A1^((-/-)). The LD size and content decreased following BTn1 A1 gene knockout. The m RnA abundance of fatty acid synthase(FASn) and peroxisome proliferator-activated receptor-gamma(PPARG) was downregulated in the BTn1 A1^((-/-))clone. Subcellular analyses indicated that BTn1 A1 and LD were co-localized in the cytoplasm. BTn1 A1 gene knockout increased the percentage of phosphatidylethanolamine(PE) and decreased phosphatidylcholine(PC), which resulted in a lower PC/PE ***: Results suggest that BTn1 A1 plays an important role in regulating LD synthesis via a mechanism involving membrane phospholipid composition.

The paper presents results of studies of the formation of phases during the solid-state synthesis in the[(na_(0.5)Bi_(0.5)T_(0.80)Ba_(0.20)]Ti_(1-y)B_(y)TO_(3)system of solid solutions with B-site *** substitutions by zirconium,tin and ion complexes(In_(0.5)nb_(0.5)T and(Fe_(0.5)nb_(0.5)T have been *** has been found that the synthesis is a multi-step process associated with the formation of a number of intermediate phases(depending on the compositions and calcination temperatures).Single-phase solid solutions have been produced at the calcination temperatures in the interval 1000–1100℃.An increase in the substituting ions concentration leads to a linear increase of the crystal cell *** the same time,the tolerance factor gets reduced boosting the stability of the antiferroelectric phase as compared to that of the ferroelectric phase.

Optical actuation of a micromechanical photodiode via the photovoltaic-piezoelectric effect

Radiation pressure and photothermal forces have been previously used to optically actuate micro/nanomechanical structures fabricated from semiconductor piezoelectric materials such as gallium arsenide(GaAs).In these materials,coupling of the photovoltaic and piezoelectric properties has not been fully explored and leads to a new type of optical actuation that we call the photovoltaic-piezoelectric effect(PVPZ).We demonstrate this effect by electrically measuring,via the direct piezoelectric effect,the optically induced strain in a novel torsional *** micron-scale torsional resonator is fabricated from a lattice-matched single-crystal molecular beam epitaxy(MBE)-grown GaAs photodiode *** find that the strain depends on the product of the electro-optic responsivity and piezoelectric constant of *** photovoltaic-piezoelectric effect has important potential applications,such as in the development of configurable optical circuits,which can be used in neuromorphic photonic chips,processing of big data with deep learning and the development of quantum circuits.

Dissipation of chlorpyrifos, pendimethalin, and thiophanate-methyl pesticides in compost based-versus peat-moss based-biomixture of biobeds

Biobed is a smart bioremediation system used to treat point-source pesticide *** is the main component of biobeds,and pesticide dissipation is affected by its *** study aimed to compare the effectiveness of compost-based(C)versus peat-moss-based(P)biomixtures of biobeds on tested pesticide *** concentrations(25,50,and 75 mg/kg)of chlorpyrifos,pendimethalin,and thiophanate methyl were added separately and as a mix to both biomixtures C and *** data showed the effect of biomixture type on the dissipation of the tested *** chlorpyrifos,its dissipation rate in biomixture P was more rapid than that in biomixture *** result was confirmed by the mineralization kinetic experiment,since 25%of the initial ^(14)C-chlorpyrifos concentration accumulated in the form ^(14)CO_(2) in biomixture P compared to only 14%in biomixture *** addition,the chlorpyrifos dissipation rate was influenced by the initial concentration when applied individually in biomixture *** contrast,biomixture C was more effective at pendimethalin dissipation than biomixture P,since>76%of pendimethalin was dissipated in biomixture C versus 67%in biomixture P at the same incubation *** thiophanate-methyl,the abilities of both biomixtures C and P were similar and less efficient than those of the other tested *** addition of the three tested pesticides to biomixture C only had a positive effect on both chlorpyrifos and thiophanate-methyl dissipation,while pendimethalin dissipation was similar when applied separately or as a *** activity was stimulated by the addition of separately or mixed pesticides to biomixtures C and P as measured by dehydrogenase activity.

Recent advances in the understanding of deltaic deposits provide new tools for the study and analysis of deltaic deposits in shallow epicontinental *** the introduction of sequence stratigraphic concepts,meter-scale coarsening and thickening upward successions have been considered as“parasequences”originated by high-frequency sea-level ***,recent studies enhanced the importance of wave-aided low-dense hyperpycnal flows in transporting fine-grained sediments in shallow shelfal *** poorly-known(and at the same time very common)types of delta,known as hyperpycnal littoral deltas(HLD),develop very low gradient progradational units,controlled by changes in the sediment supply instead of sea level *** small-scale progradational units are very common in shallow epicontinental seas like the Lower Cretaceous Agrio Formation in the neuquén *** study provides a first detailed analysis of hyperpycnal littoral deltas from the Agua de la Mula Member(upper Hauterivian-lower Barremian)of the Agrio *** unit has been studied in three locations near Bajada del Agrio locality in the central part of the neuquén Basin,*** sandy facies,three heterolithic facies,three muddy facies and four calcareous facies were *** facies analysis,three facies associations could be determined,corresponding to offshore/prodelta,distal ramp delta and proximal ramp *** three stratigraphic sections discussed in this study are internally composed of several small-scale sequences showing a coarsening and thickening upward pattern,transitionally going from muddy to sandy wave-dominated facies,and ending with calcareous bioclastics levels on *** small-scale sequences are interpreted as delta front deposits of wave-influenced hyperpycnal littoral deltas,punctuated by calcareous intervals accumulated during periods of low sediment *** is interpreted that the development of hyperpycnal littoral deltas could have been

The n-formyl peptide receptors: contemporary roles in neuronal function and dysfunction

n-formyl peptide receptors(FPRs)were first identified upon phagocytic leukocytes,but more than four decades of research has unearthed a plethora of non-myeloid roles for this receptor *** are expressed within neuronal tissues and markedly in the central nervous system,where FPR interactions with endogenous ligands have been implicated in the pathophysiology of several neurodegenerative diseases including Alzheimer's disease and Parkinson's disease,as well as neurological cancers such as *** the homeostatic function of FPRs in the nervous system is currently undefined,a variety of novel physiological roles for this receptor family in the neuronal context have been posited in both human and animal *** developments in recent years have implicated FPRs in the process of neurogenesis and neuronal differentiation which,upon greater characterisation,could represent a novel pharmacological target for neuronal regeneration therapies that may be used in the treatment of brain/spinal cord injury,stroke and *** review aims to summarize the recent progress made to determine the physiological role of FPRs in a neuronal setting,and to put forward a case for FPRs as a novel pharmacological target for conditions of the nervous system,and for their potential to open the door to novel neuronal regeneration therapies.

1997年12月至1998年2月,我们对南极半岛席尔瓦角善于飞翔海鸟与企鹅的取食关联性进行了研究,同时调查了取食集团中主要鸟种的食性。发现每个取食集团中有35.6-37.0只善于飞翔的海鸟,其中几乎都有纹颊企鹅群(Pygoscelis antarctica),黑背鸥(Larus dominicanus)、灰贼鸥(Catharacta maccormicki)、花斑鹱(Daptioncapensis)和巨鹱(Macronectes giganteus)是各集团中最常见的鸟类。各取样单元内有相关性的种数随季节变化而减少,一些种类的减少与特定的物候期有关。南极磷虾(Euphausia superba)是绝大部分飞翔海鸟的主要食物,研究发现黑背鸥与纹颊企鹅所捕食的南极磷虾的大小最为接近。飞翔海鸟的觅食行为表明:在海面上短时停留的飞翔海鸟也能够成功捕捉到磷虾,这可能与磷虾躲避企鹅的捕食有关。

Semi-exact Solutions of Konwent Potential

In this work we study the quantum system with the symmetric Konwent potential and show how to find its exact solutions. We find that the solutions are given by the confluent Heun function. The eigenvalues have to be calculated numerically because series expansion method does not work due to the variable z ≥ 1. The properties of the wave functions depending on the potential parameter A are illustrated for given potential parameters V_0 and a. The wave functions are shrunk towards the origin with the increasing |A|. In particular, the amplitude of wave function of the second excited state moves towards the origin when the positive parameter A decreases. We notice that the energy levels ε_i increase with the increasing potential parameter |A| ≥ 1, but the variation of the energy levels becomes complicated for |A| ∈(0, 1), which possesses a double well. It is seen that the energy levels ε_i increase with |A| for the parameter interval A ∈(-1, 0), while they decrease with |A| for the parameter interval A ∈(0, 1).
