All-in-one treatments represent a paradigm shift in future *** example,inflammatory bowel disease(IBD)is mainly diagnosed by endoscopy,which could be applied for not only on-site monitoring but also the intestinal lesion-targeted spray of injectable ***,molecular conjugation to the hydrogels would program both lesion-specific adhesion and drug-free *** study validated this concept of all-in-one treatment by first utilizing a well-known injectable hydrogel that underwent efficient solution-to-gel transition and nanomicelle formation as a translatable *** properties enabled spraying of the hydrogel onto the intestinal walls during ***,peptide conjugation to the hydrogel guided endoscopic monitoring of IBD progress upon adhesive gelation with subsequent moisturization of inflammatory lesions,specifically by *** peptide was designed to mimic the major component that mediates intestinal interaction with Bacillus subtilis flagellin during IBD ***,the peptide-guided efficient adhesion of the hydrogel nanomicelles onto Toll-like receptor 5(TLR5)as the main target of flagellin binding and *** peptide binding potently suppressed inflammatory signaling without drug loading,where TLR5 and Notch-1 operated collaboratively through downstream actions of tumor necrosis *** results were produced using a human colorectal cell line,clinical IBD patient cells,gut-on-a-chip,a mouse IBD model,and pig experiments to validate the translational utility.

The extract of Celtis choseniana Nakai alleviates testosterone-in-duced benign prostatic hyperplasia through inhibiting 5αreductase type 2 and the Akt/NF-κB/AR pathway

Benign prostatic hyperplasia(BPH)is a chronic male disease characterized by the enlarged *** choseniana Nakai(***)is medicinally used to alleviate pain,gastric disease,and lung *** this study,the effect of *** extract on BPH was investigated using testosterone-induced *** Dawley rats were divided into five groups:control,BPH(testosterone 5 mg·kg^(−1)),Fina(finasteride 2 mg·kg^(−1)),and ***(50 and 100 mg·kg^(−1)).After four weeks of TP treatment with finasteride or ***,prostate weights and DHT levels were *** addition,the prostates were histopathologically examined and measured for protein kinase B(Akt)/nuclear factor-κB(NF-κB)/AR signaling,proliferation,apoptosis,and ***-state weight and epithelial thickness were reduced in the *** groups compared with that in the BPH *** extract of *** acted as a 5αreductase inhibitor,reducing DHT levels in the ***,the extract of *** blocked the activation of p-Akt,nuclear NF-κB activation and reduced the expression of AR and PSA compared with ***,the ex-pression of Bax,PARP-1,and p53 increased,while the expression of bcl-2 *** present study demonstrated that ***-ana extract alleviated testosterone-induced BPH by suppressing 5αreductase and Akt/NF-κB activation,reducing AR signaling and in-ducing apoptosis and autophagy in the *** results suggested that *** probably contain potential herbal agents to alleviate BPH.

Porous titanium(Ti)scaffolds have been extensively utilized as bone substitute scaffolds due to their superior biocompatibility and excellent mechanical ***,naturally formed TiO2 on the surface limits fast *** biomolecules have been widely utilized to overcome this issue;however,homogeneous porous Ti scaffolds could not simultaneously deliver multiple biomolecules that have different release *** this study,functionally graded porous Ti scaffolds(FGPTs)with dense inner and porous outer parts were fabricated using a two-body combination and densification *** with growth factor(BMP-2)and antibiotics(TCH)exhibited suitable mechanical properties as bone substituting material and presented good structural *** release of BMP-2 was considerably prolonged,whereas the release of TCH was comparable to that of homogenous porous titanium scaffolds(control group).The osteogenic differentiation obtained using FGPTs was maintained due to the prolonged release of *** antimicrobial properties of these scaffolds were verified using *** in terms of prior release *** addition,various candidates for graded porous Ti scaffolds with altered pore characteristics were presented.

Objective:To evaluate the effects of an aqueous extract of Protaetia brevitarsis(AEPB)on the growth of zebrafish and preosteoblast MC3T3-E1 ***:The effects of AEPB on the linear growth and the expression of growth-related genes in zebrafish and MC3T3-E1 cells were assessed using various molecular ***,the involvement of the mammalian target of rapamycin(mTOR)pathway in AEPB-induced growth was investigated by employing the mTOR inhibitor ***:AEPB administration led to a significant and dose-dependent increase in zebrafish larvae growth over ***,AEPB treatment upregulated the expression of growth hormone-1(GH-1),insulin-like growth factor-1(IGF-1),growth hormone receptor-1(GHR-1),and cholecystokinin-a(CCKA)in ***,AEPB stimulated the expression and release of IGF-1 and accelerated mTOR expression in MC3T3-E1 *** addition,rapamycin hindered AEPB-induced linear growth in zebrafish larvae and suppressed the expression of growth-promoting genes by inhibiting mTOR ***:AEPB shows growth-promoting effects by upregulating growth-related genes and activating the mTOR signaling *** investigations are warranted to elucidate its mechanisms of action and explore its potential application in the development of growth-enhancing supplements for various purposes.

Morbidity analysis of left hepatic trisectionectomy for hepatobiliary disease and live donor

Background:Despite remarkable advances in surgical techniques and perioperative management,left hepatic trisectionectomy(LHT)remains a challenging procedure with a somewhat high postoperative morbidity rate compared with less-extensive *** study aimed to analyze the short-and long-term outcomes of LHT and identify factors associated with the postoperative morbidity of this technically demanding surgical ***:The medical records of 53 patients who underwent LHT between June 2005 and October 2019 at a single institution were retrospectively *** independent prognostic factor of postoperative morbidity was analyzed using the logistic regression ***:Hepatocellular carcinoma was the most common indication for surgery(n=21),followed by hilar cholangiocarcinoma(n=14),intrahepatic cholangiocarcinoma(n=10),and other pathologies(including colorectal liver metastasis,hepatolithiasis,gallbladder cancer,living donor,hemangioma,and multilocular biliary cyst;n=8).The rates of postoperative morbidities of Clavien-Dindo grade 3 or higher and 90-day mortality were 39.6% and 1.9%,*** 1-,3-,and 5-year overall survival rates were 81.1%,61.4%,and 44.6%,*** analysis revealed that preoperative jaundice[hazard ratio(HR)=6.15,95%confidence interval(CI):1.57-24.17,P=0.009]and operative time>420 min(HR=4.66,95%CI:1.27–17.17,P=0.021)were independent predictors of postoperative ***:The in-hospital mortality of LHT surgery can be minimalized by a reliable preoperative evaluation of liver function and selection of the dominant anatomic features of right posterior sector,active and appropriate preoperative management for obstructive cholangitis and compensatory hypertrophy of the future remnant posterior sector,and the experience of the surgeon.

Glucocorticoid pulse therapy in an elderly patient with post-COVID-19 organizing pneumonia:A case report

BACKGROUND Pulmonary fibrosis often occurs as a sequel of coronavirus disease 2019(COVID-19);however,in some cases,it can rapidly progress,similar to the acute exacerbation of interstitial lung *** are the standard treatment for severe COVID-19 pneumonia requiring oxygen supply;however,the post-COVID-19 efficacy of high-dose steroid therapy remains ***,we presented a case of an 81-year-old man who developed acute respiratory failure after COVID-19 and was treated with glucocorticoid pulse *** SUMMARY An 81-year-old man with no respiratory symptoms was admitted due to a diabetic *** had been previously treated for COVID-19 pneumonia six weeks ***,upon admission,he suddenly complained of dyspnea and required a high-flow oxygen *** simple chest radiography and computed tomography(CT)revealed diffuse ground-glass opacities and consolidation in both ***,repeated sputum tests did not identify any infectious pathogens,and initial broad-spectrum antibiotic therapy did not result in any clinical improvement with the patient having an increasing oxygen *** patient was diagnosed with post-COVID-19 organizing ***,we initiated glucocorticoid pulse therapy of 500 mg for three days followed by a tapered dose on hospital day(HD)*** three days of pulse treatment,the patient's oxygen demand *** patient was subsequently discharged on HD 41,and chest radiography and CT scans have almost normalized nine months after *** Glucocorticoid pulse therapy may be considered when the usual glucocorticoid dose is ineffective for patients with COVID-19 sequelae.

Calcium-dependent proteasome activation is required for axonal neurofilament degradation

Even though many studies have identified roles of proteasomes in axonal degeneration, the mo- lecular mechanisms by which axonal injury regulates proteasome activity are still unclear. In the present study, we found evidence indicating that extracellular calcium influx is an upstream regulator of proteasome activity during axonal degeneration in injured peripheral nerves. In degenerating axons, the increase in proteasome activity and the degradation of ubiquitinated proteins were sig- nificantly suppressed by extracellular calcium chelation. In addition, electron microscopic findings revealed selective inhibition of neurofilament degradation, but not microtubule depolymerization or mitochondrial swelling, by the inhibition of calpain and proteasomes. Taken together, our findings suggest that calcium increase and subsequent proteasome activation are an essential initiator of neurofilament degradation in Wallerian degeneration.

Prognostic implications of FGFR1 and MYC status in esophageal squamous cell carcinoma

AIM To investigate the clinicopathological features and prognostic implications of combined MYC and fibroblast growth factor receptor 1(FGFR1) status in esophageal squamous cell carcinomas(ESCCs). METHODS All patients with ESCC(n = 180) underwent surgical resection at Seoul National University Hospital sometime between 2000 and 2013. A tissue microarray was constructed using cores obtained from representative tumor areas of formalin-fixed, paraffin-embedded tissue blocks. FGFR1 and MYC copy numbers were quantified using fluorescence in situ hybridization. The level of MYC expression was determined using immunohistochemistry. FGFR1 and MYC amplification status was compared between primary and metastatic lymph nodes. Univariate and multivariate survival analyses were performed according to adjuvant therapy *** FGFR1 and MYC amplifications were observed in 21.4%(37/173) and 54.2%(91/168) of patients, respectively, while MYC expression was observed in 58.9%(106/180) of patients. There was a positive correlation between MYC amplification and overexpression(P = 0.002). Although FGFR1 amplification was not associated with MYC amplification or expression, 12.3%(20/163) of patients exhibited both FGFR1 amplification and MYC expression. There was also a correlation in FGFR1 amplification status between matched primary tumors and metastatic lymph nodes(P < 0.001). MYC expression was higher in ESCCs with p T1(P < 0.001) and in those with no lymph node metastasis(P = 0.023). MYC expression was associated with prolonged diseasefree survival(P = 0.036) and overall survival(OS)(P = 0.017) but was not an independent prognostic factor. FGFR1 amplification was an independent predictor for prolonged OS in all patients(P = 0.029) and in those who did not receive adjuvant therapy(P = 0.013). Combined FGFR1 amplification and MYC expression predicted better OS in patients who did not receive adjuvant therapy(P = 0.034) but not in those who did receive adjuvant ***

Two-dimensional van der Waals(2D vdW)material-based heterostructure devices have been widely studied for high-end electronic applications owing to their heterojunction *** this study,we demonstrate graphene(Gr)-bridge heterostructure devices consisting of laterally series-connected ambipolar semiconductor/Gr-bridge/n-type molybdenum disulfide as a channel material for field-effect transistors(FET).Unlike conventional FET operation,our Gr-bridge devices exhibit nonclassical transfer characteristics(humped transfer curve),thus possessing a negative differential *** phenomena are interpreted as the operating behavior in two series-connected FETs,and they result from the gate-tunable contact capacity of the Gr-bridge ***-value logic inverters and frequency tripler circuits are successfully demonstrated using ambipolar semiconductors with narrow-and wide-bandgap materials as more advanced circuit applications based on non-classical transfer ***,we believe that our innovative and straightforward device structure engineering will be a promising technique for future multi-functional circuit applications of 2D nanoelectronics.
