the COVID-19 pandemic caused by SARS-CoV-2 virus is an ongoing global health *** cases of COVID-19 and the rare cases of COVID-19 vaccine-induced-thrombotic-thrombocytopenia(VItt)are both associated with thrombosis and thrombocytopenia;however,the underlying mechanisms remain inadequately *** infection and vaccination utilize the spike protein receptor-binding domain(RBD)of *** found that intravenous injection of recombinant RBD caused significant platelet clearance in *** investigation revealed the RBD could bind platelets,cause platelet activation,and potentiate platelet aggregation,which was exacerbated in the Delta and Kappa *** RBD–platelet interaction was partially dependent on theβ3 integrin as binding was significantly reduced inβ3−/−***,RBD binding to human and mouse platelets was significantly reduced with relatedαIIbβ3 antagonists and mutation of the RGD(arginine-glycine-aspartate)integrin binding motif to RGE(arginine-glycine-glutamate).We developed anti-RBD polyclonal and several monoclonal antibodies(mAbs)and identified 4F2 and 4H12 for their potent dual inhibition of RBD-induced platelet activation,aggregation,and clearance in vivo,and SARS-CoV-2 infection and replication in Vero E6 *** data show that the RBD can bind platelets partially thoughαIIbβ3 and induce platelet activation and clearance,which may contribute to thrombosis and thrombocytopenia observed in COVID-19 and *** newly developed mAbs 4F2 and 4H12 have potential not only for diagnosis of SARS-CoV-2 virus antigen but also importantly for therapy against COVID-19.

Platelets are small, versatile blood cells that are critical for hemostasis/thrombosis. local platelet accumulation is a known contributor to proinflammation in various disease states. However, the anti-inflammatory/immunosuppressive potential of platelets has been poorly explored. Here, we uncovered, unexpectedly, desialylated platelets (dPlts) down-regulated immune responses against both platelet-associated and -independent antigen challenges. Utilizing multispectral photoacoustic tomography, we tracked dPlt trafficking to gut vasculature and an exclusive Kupffer cell-mediated dPlt clearance in the liver, a process that we identified to be synergistically dependent on platelet glycoprotein Ibα and hepatic Ashwell–Morell receptor. Mechanistically, Kupffer cell clearance of dPlt potentiated a systemic immunosuppressive state with increased anti-inflammatory cytokines and circulating CD4+ regulatory t cells, abolishable by Kupffer cell depletion. last, in a clinically relevant model of hemophilia A, presensitization with dPlt attenuated anti-factor VIII antibody production after factor VIII ( infusion. As platelet desialylation commonly occurs in daily-aged and activated platelets, these findings open new avenues toward understanding immune homeostasis and potentiate the therapeutic potential of dPlt and engineered dPlt transfusions in controlling autoimmune and alloimmune diseases.

Mitochondriopathy inspired adenosine triphosphate(AtP) depletions have been recognized as a powerful way for controlling tumor growth. Nevertheless, selective sequestration or exhaustion of AtP under complex biological environments remains a prodigious challenge. Harnessing the advantages of in vivo self-assembled nanomaterials, we designed an Intracellular AtP Sequestration(IAS) system to specifically construct nanofibrous nanostructures on the surface of tumor nuclei with exposed AtP binding sites,leading to highly efficient suppression of bladder cancer by induction of mitochondriopathy-like ***, the reported transformable nucleopeptide(NlS-FF-t) self-assembled into nuclear-targeted nanoparticles with AtP binding sites encapsulated inside under aqueous conditions. By interaction with KPNA2, the NlS-FF-t transformed into a nanofibrous-based AtP trapper on the surface of tumor nuclei,which prevented the production of intracellular energy. As a result, multiple bladder tumor cell lines(t24,EJ and Rt-112) revealed that the half-maximal inhibitory concentration(IC50) of NlS-FF-t was reduced by approximately 4-fold when compared to NlS-t. Following intravenous administration, NlS-FF-t was found to be dose-dependently accumulated at the tumor site of t24 xenograft mice. More significantly, this IAS system exhibited an extremely antitumor efficacy according to the deterioration of t24 tumors and simultaneously prolonged the overall survival of t24 orthotopic xenograft mice. together, our findings clearly demonstrated the therapeutic advantages of intracellular AtP sequestration-induced mitochondriopathy-like damages, which provides a potential treatment strategy for malignancies.

Effect of slag composition on the cleanliness of 28MnCr5 gear steel in the refining processes

the equilibrium reaction between CaO-Al2O3-SiO2-MgO slag and 28MnCr5 molten steel was calculated to obtain the suitable slag composition which is effective for decreasing the oxygen content in molten steel. the dissolved oxygen content [O] in molten steel un- der different top slag conditions was calculated using a thermodynamic model and was measured using an electromotive force method in slag-steel equilibrium experiments at 1873 K. the relations among [O], the total oxygen content （t.O）, and the composition of the slag were investigated. the experimental results show that both [O] and t.O decrease with decreasing SiO2 content of the slag and exhibit different trends with the changes in the CaO/Al2O3 mass ratio of the slag. Increasing the CaO/Al2O3 mass ratio results in a decrease in [O] and an in- crease in t.O. to ensure that t.O ≤ 20 ppm and [O] ≤ 10 ppm, the SiO2 content should be controlled to 〈5wt%, and the CaO/AI203 mass ratio should be in the range from 1.2 to 1.6.

本文采用情感评定任务和自定步速阅读任务对外语谎言的情感与认知加工进行研究。结果显示:1)与母语谎言相比,中国英语学习者对外语谎言的情感评定整体较弱,但对外语利他谎言情感效价评定与母语无显著差异;2)与母语谎言相比,外语谎言的加工时间显著增加,且外语语境下谎言与真言之间加工时间的关系模式与母语不同。结果表明,外语谎言加工发生了部分情感钝化反应,支持了外语的弱具身性和谎言的极差属性。另外,本文基于BIA模型和双过程加工理论构建了外语情感语言认知加工模型,以阐释外语谎言的认知加工机制。本研究的发现还可为谎言研究方法和外语教学实践提供参考。

洞庭湖是我国最大的通江湖泊之一,为探究洞庭湖溶解性有机物(DOM)组成、(DOC)收支与江湖水力连通间的内在关联,于2018年5月、7月、9月和11月在洞庭湖湘、资、沅、澧4支流和长江荆南松滋、虎渡、藕池3个入湖口以及洞庭湖出湖口城陵矶采集水样,并探究洞庭湖流域DOM的组成分异特征及其对江湖水力连通的响应.结果表明,入湖河流和城陵矶DOM吸收系数a254和陆源类腐殖酸组分C1在7月最高.DOC由入湖口(2.4±1.3mg)/l下降至出湖口(2.1±0.3)mg/l,而a254由入湖口(9.7±2.2)m^(-1)上升至出湖口(10.1±1.7)m^(-1).微生物培养前的DOC浓度为(2.4±1.0)mg/l,微生物降解后的DOC浓度为(2.1±0.9mg)/l,DOC的微生物活性,即BDOC最高值出现在澧水为50.3%.5月、7月、9月和11月入湖的DOC通量分别为(63.7±43.6)×10^(3)t,(60.1±7.3)×10^(3)t,(27.6±3.6)×10^(3)t,(31.9±13.9)×10^(3)t.洞庭湖因自身水力滞留时间短,导致DOM在湖内运移过程中较少被降解,全湖DOM来源组成受流域不同季节来水条件影响较大.

Background:Black locust(Bl,Robinia pseudoacacia)is considered a promising tree species for reforestation due to its great ability to fix ***,after two or three coppice-harvesting rotations,the productivity of Bl *** soil microbial communities are affected and how these groups correlate with the nitrogen mineralization process across multi-generation stands remains ***:We investigated the composition and structure of free-living nitrogen-fixing microorganisms(diazotrophs)by sequencing the marker gene nifH and compared these results to levels of soil nitrogen mineralization in the bulk soil and rhizosphere in black locust plantations on Mount tai,***:the results showed multi-generation Bl coppice plantations decreased the total soil nitrogen(N),soil phosphorus(P),soil microbial biomass N(MBN),soil microbial biomass C(MBC),soil nitrification rate(Rn),soil ammonification rate(Ra),and net soil N mineralization rate(Rm),but significantly increased the concentration of soil NH_(4þ)-N to maintain sufficient NO_(3)^(-)*** dominant species in bulk soil and rhizosphere changed from Rhodopseudomonas(22.62%and 15.76%),unclassified_c_Alphaproteobacteria(22.37%and 29.28%),unclassified_o_Rhizobiales(15.40%and 13.31%),Bradyrhizobium(12.00%and 11.74%)in seedling plantations to Bradyrhizobium(45.95%and 47.86%)and Rhodopseudomonas(43.56%and 41.84%)in coppice plantations,*** test and Redundancy analysis(RDA)revealed that Rn,Ra,and Rm were the most important factors shaping the diazotrophic ***:Our results suggest that the multi-generation Bl coppice plantation can homogenize soil diazotrophic communities,which is mainly regulated by the available N loss caused by nitrogen *** the management technology of coppice plantations will provide more beneficial external consumption.

Multi-h C波段导航信号调制方式及性能评估

近年来,l波段(1164~1610 MHz)频谱拥挤日渐严重,我国北斗卫星导航系统可用资源明显不足,基于频谱资源的不可再生性,探索研究新频段迫在眉睫。鉴于目前C波段导航信号设计尚不成熟,各国对C波段卫星导航信号体制也都还处于探索研究阶段,我国有必要预先开展C波段卫星导航信号研究。处于C波段中5010~5030 MHz的卫星导航信号具有较少的频谱干扰、较低的电离层误差等显著优势,但仅有的20 MHz带宽对诸如射电天文业务(Radio Astronomy Service,RAS)和微波着陆系统(Microwave landing System,MlS)等相邻业务频段的射频兼容性要求极为严苛。首先提出将多调制指数(Multi-h)的正交频分复用-连续相位调制(Orthogonal Frequency Division Multiplexing-Continuous Phase Modulation,OFDM-CPM)联合调制方式应用于C波段卫星导航;其次,在分析关键参数对Multi-h OFDM-CPM信号功率谱特性影响后,选取关联长度l为64、调制指数h为0.5的单调制指数(Single-h)OFDM-CPM(10)信号和h为[1/16,3/16]的Multi-h OFDM-CPM(10)信号作为C波段候选导航信号;最后,对包括兼容性、码跟踪精度、抗多径干扰在内的基础导航性能指标进行评估。研究表明,OFDM-CPM调制方式能很好兼顾C波段信号的带外约束性和导航性能要求,OFDM-CPM(10)h=[1/16,3/16]信号在所述各个基础导航性能指标上均表现最佳,在兼容性方面能弥补OFDM-CPM(10)h=0.5信号的略微不足。然而考虑到多调制指数会增加接收机复杂度,因此在C波段导航中,只有当接收机采用低复杂度检测算法时,Multi-h OFDM-CPM信号才会是比Single-h OFDM-CPM信号更佳的选择。

Oil charging history and pathways of the Ordovician carbonate reservoir in the tuoputai region,tarim Basin,NW China

Geochemical analysis of 64 oils sampled from an Ordovician carbonate reservoir in the tuoputai region was undertaken to study the composition of molecular markers. All the oils have similar geochemical characteristics and belong to a single oil family. they are presumed to derive from the same source kitchen and have similar oil charging history. A histogram of homogenization temperatures(th) of aqueous inclusions in reservoir rocks shows a bimodal distribution pattern, indicating that the Ordovician reservoir has been charged twice. Coupling the measured th(°C) with the burial and geothermal histories reconstructed using 1D basin modeling, we relate the homogenization temperature to the relevant geological ages: i.e.,425–412 and 9–4 Ma, corresponding to the Middle to late Silurian and the Miocene to Pliocene, respectively. the oil filling orientation and pathways are traced using molecular indicators related to alkyldibenzothiophenes and benzo[b]naphthothiophenes. the oil charging orientation is from south to north generally. It can be predicted that the Ordovician reservoirs were sourced from a kitchen located to the south of the tuoputai region, most probably between the Awati and Manjiaer Depressions. traps located in the southern side of the tuoputai region, along the oil charging pathways, should therefore be preferred oil exploration targets.
