Hypoxia-mediated programmed cell death is involved in the formation of wooden breast in broilers

Background Wooden breast(WB)myopathy is a common myopathy found in commercial broiler chickens *** examination has revealed that WB myopathy is accompanied by damage to the pectoralis major(PM)***,the underlying mechanisms responsible for the formation of WB in broilers have not been fully *** study aimed to investigate the potential role of hypoxia-mediated programmed cell death(PCD)in the formation of WB *** Histological examination and biochemical analysis were performed on the PM muscle of the control(CON)and WB groups.A significantly increased thickness of the breast muscle in the top,middle,and bottom portions(Pth pathological structure damage of myofibers in the WB *** number of capillaries per fiber in PM muscle,and the levels of p O_(2) and s O_(2) in the blood,were significantly decreased(Pthe levels of p CO_(2) and tCO_(2) in the blood were significantly increased(Pting hypoxic conditions in the PM muscle of the WB *** further evaluated the PCD-related pathways including autophagy,apoptosis,and necroptosis to understand the consequence response to enhanced hypoxic conditions in the PM muscle of birds with *** ratio of LC3 II to LC3 I,and the autophagy-related factors HIF-1α,BNIP3,Beclin1,AMPKα,and ULK1 at the m RNA and protein levels,were all significantly upregulated(Pthat autophagy occurred in the PM muscle of the WB *** apoptotic index,as well as the expressions of Bax,Cytc,caspase 9,and caspase 3,were significantly increased(Ptly decreased(Pthe WB-affected PM muscle,indicating the occurrence of apoptosis mediated by the mitochondrial ***,the expressions of necroptosis-related factors RIP1,RIP3,and MLKL,as well as NF-κB and the pro-inflammatory cytokines tNF-α,IL-1β,and IL-6,were all significantly enhanced(Pthe WB-affected PM *** t

Optimizing electronic structure of Mo_(2)tiC_(2)t_(x) MXene through Nb doping for enhanced electrochemical performance

MXene is a promising electrode material for both high volumetric capacitance and high-rate performance in ***,the current study has mainly focused on the monometallic element ti_(3)C_(2)t_(x) MXene until now,while the bimetallic and multimetallic MXene have received comparatively less *** this work,we demonstrate that the electronic structure of the Mo_(2)tiC_(2)t_(x) MXene could be regulated by fine-tuning the content of doped Nb *** enhanced electron cloud density of surface–O termination and the electron spin of the Mo atoms in the Mo_(2)tiC_(2)t_(x) MXene,leads to the boost of electric double-layer capacitor(EDLC)and improvement of *** a consequence,the electrochemical performance of Nb-doped Mo_(2)tiC_(2)t_(x) MXene(Nb-0.3-MXene)demonstrates a capacitance of 398 F·cm^(−3),roughly doubling that of the pristine Mo_(2)tiC_(2)t_(x) MXene electrode at 197 F·cm^(−3) in the 3 M H_(2)SO_(4) *** the same time,the Nb-0.3-MXene could even maintain a capacitance of 82.75% at 200 mV·s−1,with high cyclic stability for 19,000 cycles at 10 A·g−***,Nb-0.3-MXene-based hybrid supercapacitors deliver a remarkable volumetric energy density of 48.1 W·h·L^(−1)at 230.7 W·L^(−1),and 34.4 W·h·L^(−1)at a high power density of 82.6 kW·L^(−1).there exists a balance between the volumetric capacitance and rate performance with different ratios of Nb atoms in the Nb-doped MXene due to the strong interaction between the Nb-doped MXene and the intercalated ***,optimizing the electronic structure of MXene through heteroatom doping is of great potential for enhanced supercapacitor performance.

Selenium-oxidizing Agrobacterium sp.t3F4 decreases arsenic uptake by Brassica rapa *** a native polluted soil

toxic arsenic(As)and trace element selenium(Se)are transformed by microorganisms but their complex interactions in soil-plant systems have not been fully *** Asand Se-oxidizing bacterium,Agrobacterium sp.t3F4,was applied to a native seleniferous As-polluted soil to investigate As/Se uptake by the vegetable Brassica rapa *** As-Se interaction as mediated by strain *** Se content in the aboveground plants was significantly enhanced by 34.1%,but the As content was significantly decreased by 20.5% in the t3F4-inoculated pot culture compared to the control(Pt was shown in treatment with additional 5 mg/kg of Se(IV)in *** addition,the As contents in roots were significantly decreased by more than 35% under t3F4 or Se(IV)treatments(Pterium interaction in a soil simulation experiment showed that the bioavailability of Se significantly increased and As was immobilized with the addition of the t3F4strain(Pthermore,an As/Se co-exposure hydroponic experiment demonstrated that As uptake and accumulation in plants was reduced by increasing Se(IV)*** 50% growth inhibition concentration(IC50)values for As in plants were increased about one-fold and two-fold under co-exposure with 5 and 10μmol/L Se(IV),*** conclusion,strain t3F4 improves Se uptake but decreases As uptake by plants via oxidation of As and Se,resulting in decrease of soil As bioavailability and As/Se competitive absorption by *** provides a potential bioremediation strategy for Se biofortification and As immobilization in As-polluted soil.

Inhalable metal–organic framework-mediated cuproptosis combined with PD-L1 checkpoint blockade for lung metastasis synergistic immunotherapy

Cuproptosis shows enormous application prospects in lung metastasis ***,the glycolysis,Cu^(+)efflux mechanisms,and insufficient lung drug accumulation severely restrict cuproptosis ***,an inhalable poly(2-(N-oxide-N,N-diethylamino)ethyl methacrylate)(OPDEA)-coated copper-based metal–organic framework encapsulating pyruvate dehydrogenase kinase 1 siRNA(siPDK)is constructed for mediating cuproptosis and subsequently promoting lung metastasis immunotherapy,namely *** inhalation,OMP shows highly efficient lung accumulation and long-term retention,ascribing to the OPDEA-mediated pulmonary mucosa *** tumor cells,OMP is degraded to release Cu2+under acidic condition,which will be reduced to toxic Cu^(+)to induce cuproptosis under glutathione(GSH)***,siPDK released from OMP inhibits intracellular glycolysis and adenosine-5ʹ-triphosphate(AtP)production,then blocking the Cu^(+)efflux protein AtP7B,thereby rendering tumor cells more sensitive to OMP-mediated ***,OMP-mediated cuproptosis triggers immunogenic cell death(ICD)to promote dendritic cells(DCs)maturation and CD8^(+)t cells ***,OMP-induced cuproptosis up-regulates membrane-associated programmed cell death-ligand 1(PD-L1)expression and induces soluble PD-L1 secretion,and thus synergizes with anti-PD-L1 antibodies(aPD-L1)to reprogram immunosuppressive tumor microenvironment,finally yielding improved immunotherapy ***,OMP may serve as an efficient inhalable nanoplatform and afford preferable efficacy against lung metastasis through inducing cuproptosis and combining with aPD-L1.

Argatroban promotes recovery of spinal cord injury by inhibiting the PAR1/JAK2/StAt3 signaling pathway

Argatroban is a synthetic thrombin inhibitor approved by *** and Drug Administration for the treatment of ***,whether it plays a role in the repair of spinal cord injury is *** this study,we established a rat model of t10 moderate spinal cord injury using an NYU Impactor ModerⅢand performed intraperitoneal injection of argatroban for 3 consecutive *** results showed that argatroban effectively promoted neurological function recovery after spinal cord injury and decreased thrombin expression and activity in the local injured spinal *** sequencing transcriptomic analysis revealed that the differentially expressed genes in the argatroban-treated group were enriched in the JAK2/StAt3 pathway,which is involved in astrogliosis and glial scar *** blotting and immunofluorescence results showed that argatroban downregulated the expression of the thrombin receptor PAR1 in the injured spinal cord and the JAK2/StAt3 signal *** also inhibited the activation and proliferation of astrocytes and reduced glial scar formation in the spinal *** together,these findings suggest that argatroban may inhibit astrogliosis by inhibiting the thrombin-mediated PAR1/JAK2/StAt3 signal pathway,thereby promoting the recovery of neurological function after spinal cord injury.

Effectively managing extensive,multi-source,and multi-level real-scene 3D models for responsive retrieval scheduling and rapid visualization in the Web environment is a significant challenge in the current development of real-scene 3D applications in *** this paper,we address this challenge by reorganizing spatial and temporal information into a 3D geospatial *** introduces the Global 3D Geocoding System(G_(3)DGS),leveraging neighborhood similarity and uniqueness for efficient storage,retrieval,updating,and scheduling of these models.A combination of G_(3)DGS and non-relational databases is implemented,enhancing data storage scalability and ***,a model detail management scheduling strategy(tLOD)based on G_(3)DGS and an importance factor t is *** with mainstream commercial and open-source platforms,this method significantly enhances the loadable capacity of massive multi-source real-scene 3D models in the Web environment by 33%,improves browsing efficiency by 48%,and accelerates invocation speed by 40%.

Resource recycling from waste-water and sludge is an important part of the 14th Five-Year Plan in *** emerging titanium-based coagulants have drawn growing attentions due to their strong coagulation capability in water purification and value-added ti-loaded sludge *** and recovery of the high value-added sludge into functional nanomaterials is highly significant for both sludge reduction and environmental *** present study was carried out to investigate the recycle of the coagulated ti-loaded sludge to produce functional C/tiO_(2)composites as the anode materials for lithium-ion batteries(LIBs).It is the first time that the application of the ti-loaded wastewater sludge derived C/tiO_(2)was evaluated for *** experimental results showed that the carbon coating through in-situ carbonization of the sludge produced the C/tiO_(2)composites with a high specific surface area,stable structural integrity,and excellent electrochemical properties that would facilitate Li+diffusion in long-term LIBs *** C/tiO_(2)composites calcinated from the polytitanium sulfate-coagulated sludge at 800℃(N_(2))exhibited the best electrochemical performance during the cycling tests(601 m Ah/g at 100 m A/g after 200 cycles).the research work demonstrates the promising prospect of the recycle and value-added utilization of the ti-loaded sludge in the production of high-performance C/tiO_(2)composites for energy storage *** study provides a new way for the management and reuse of ti-loaded waste-sludge.

the selection of important factors in machine learning-based susceptibility assessments is crucial to obtain reliable susceptibility *** this study,metaheuristic optimization and feature selection techniques were applied to identify the most important input parameters for mapping debris flow susceptibility in the southern mountain area of Chengde City in Hebei Province,China,by using machine learning *** total,133 historical debris flow records and 16 related factors were *** support vector machine(SVM)was first used as the base classifier,and then a hybrid model was introduced by a two-step ***,the particle swarm optimization(PSO)algorithm was employed to select the SVM model ***,two feature selection algorithms,namely principal component analysis(PCA)and PSO,were integrated into the PSO-based SVM model,which generated the PCA-PSO-SVM and FS-PSO-SVM models,*** statistical metrics(accuracy,recall,and specificity)and the area under the receiver operating characteristic curve(AUC)were employed to evaluate and validate the performance of the *** results indicated that the feature selection-based models exhibited the best performance,followed by the PSO-based SVM and SVM ***,the performance of the FS-PSO-SVM model was better than that of the PCA-PSO-SVM model,showing the highest AUC,accuracy,recall,and specificity values in both the training and testing *** was found that the selection of optimal features is crucial to improving the reliability of debris flow susceptibility assessment ***,the PSO algorithm was found to be not only an effective tool for hyperparameter optimization,but also a useful feature selection algorithm to improve prediction accuracies of debris flow susceptibility by using machine learning *** high and very high debris flow susceptibility zone appropriately covers 38.01%of the study area,where debris flow may occur under intens
