Molecular mapping of leaf rust resistance genes in the wheat line Yu 356-9

The Chinese wheat line Yu 356-9 exhibits a high level of resistance to leaf rust. in order to decipher the genetic base of resistance in Yu 356-9, gene postula.ion, inheritance analyses, and chromosome linkage mapping were carried out. Gene postula.ion completed using 15 leaf rust pathotypes and 36 isogenic lines indicated that Yu 356-9 was resistant to all pathotypes tested. F1 and F2 plants from the cross Yu 356-9 （resistant）/Zhengzhou 5389 （susceptible） were tested with leaf rust pathotype ＂FHNQ＂ in the greenhouse. Results indicated a 3：1 segrega.ion ra.io, indica.ive of the presence of a single dominant leaf rust resistance gene in Yu 356-9 which was tempora.ily designated as LrYu. Bulk segregant analysis and molecular marker assays were used to map LrYu. Five simple sequence repeat （SSR） markers on chromosome 2BS were found closely linked to LrYu. Among these markers, Xwmc770 is the most closely linked, with a genetic distance of 5.7 cM.

Background: in nursing educa.ion for better teaching and essential professional skills, the clinical practice plays a substantial role. Practice at clinical settings permits students to convert theoretic knowledge into the knowledge of the skills mandatory for the care of the pa.ient. Clinical learning environment (CLE) is an important part in educa.ion of nursing and has a sizable influence on the students’ learning. Objective: The purpose of this study is to exa.ine perception and sa.isfaction of nursing students with their CLEs in Hyderabad, Pa.istan. Methods: This cross-sectional study was conducted at three nursing institutes of Hyderabad from December 2018 to January 2019 among 342 nursing students. Clinical Learning Environment, Supervision and Nurse Teacher (CLES + T) assessment tool was used as the instrument to identify the students’ perception about the learning environment in clinical setting. Results: The mean age of the participants was 25.6 ± 4.93 with majority of them male (70.7%). Three domains, pedagogical atmosphere, supervisory rela.ionship and nurse teacher role in clinical practice showed good relia.ility of more than 70%. Highest domains vise mean score was obtained for nursing premises on the ward (3.315) whereas lowest for nurse teacher role in clinical practice (NT) (3.062). Analysis of va.iance revealed that three domains supervisory rela.ionship, leadership style of the ward manager and premises of nursing showed significant mean score difference among supervisor title. Conclusion: it was found that students valued positive supervision, ward manager leadership style premises of nursing on the ward as positive CLE. Learning environment va.ies between gender, clinical settings and supervision. Medicine ward appeared to deliver the finest learning situa.ions for the nursing program.

Efficacy of autologous platelet-rich plasma.in the treatment of chronic nonhea.ing leg ulcers

Aim:The aim was to evaluate the efficacy of platelet-rich plasma(PRP)in the treatment of chronic nonhea.ing ***:A nonrandomized,uncontrolled study was performed on 24 pa.ients with 33 nonhea.ing ulcers of va.ious *** pa.ients were treated with PRP at weekly intervals for a ma.imum of 6 *** the end of the 6-week period,reduction in size of the ulcers(area and volume)was ***:The mean age of the pa.ients was 42.5 years(standard devia.ion[SD]12.48).Of 33 ulcers,there were 19 venous ulcers,7 trauma.ic ulcers,2 ulcers secondary to pyoderma gangrenosum,2 diabetic ulcers,2 trophic ulcers,and 1 vasculitic *** mean dura.ion of hea.ing of the ulcers was 5.6 weeks(SD 3.23).The mean percentage of reduction in area and volume of the ulcers was 91.7%(SD 18.4%)and 95%(SD 14%),*** 100%resolution in the area was seen in 25(76%)of the ulcers and 100%reduction in volume was seen in 24(73%)of the ulcers at the end of the 6th ***:Conventional thera.ies do not provide sa.isfactory hea.ing for chronic nonhea.ing ulcers as they are not able to provide the necessary growth factors(GFs)(platelet-derived GF,epidermal GF,vascular endothelial GF,etc.)which are essential for the hea.ing *** is a safe,affordable,biocompa.ible,and simple office-based procedure for the treatment of nonhea.ing ulcers.

目的:构建pcDNA3.1(+)/EMC6真核表达载体,并将其在人肝L-02细胞株中过表达。方法:查询Pub Med上EMC6的基因序列,针对该序列设计引物,提取L-02细胞的总RNA、聚合酶链式反应(PCR)扩增目的片段,采用基因重组技术将EMC6的cDNA定向插入真核表达载体pcDNA3.1中,构建pcDNA3.1(+)/EMC6真核表达载体,经菌落PCR,测序验证构建的准确性。应用脂质体转染方法将重组载体转入L-02细胞中,RT-PCR、蛋白质印迹法检测EMC6基因的mRNA和蛋白表达水平。结果:PCR和基因测序结果均表明pcDNA3.1(+)/EMC6重组质粒构建成功,RT-PCR和蛋白质印迹检测结果证实EMC6在人肝L-02细胞中过表达。结论:成功构建人pcDNA3.1(+)/EMC6重组质粒,并在L-02细胞中过表达。
