DNA Extraction from a Single Seed for Marker-Assisted Selection in Squash

Marker-assisted selection is an important tool in squash (Cucurbita species) breeding. A seed-based genotyping system would not only allow selection of desirable individuals prior to planting, but also reduce the cost associated with leaf-derived DNA genotyping, such as the need for greenhouse facilities and ultra-low-temperature storage freezers. A robust seed-based genotyping system requires a non-destructive sampling method and DNA of sufficient quantity and quality for marker-assisted selection. In the current study, six cultivars representing Cucurbita pepo (Black Beauty and Yellow Crookneck), C. moschata (Butterbush and Fairytale), and C. maxima (Buttercup and Big Max) were used to develop a suitable seed-based genotyping system for squash. Seed chips for DNA extraction were sampled by removing 1/3 of the distal end, while the remnant seed-embryos were sowed to assess germination potential. Four extraction methods including two column-based commercial kits (CTAB and ENZA) and two detergent-based conventional methods (CTAB and SDS) were assessed for DNA quality and quantity. Utility of extracted DNA for downstream applications was tested by genotyping with SSR and SNP markers. There was no significant difference in germination percentage between whole and cut seeds across the six cultivars. The average DNA concentration across methods ranged from 11.6 ng/μL to 62.6 ng/μl, while the DNA quality (A260/280) ranged from 0.89 to 1.95. Although DNA was obtained for all the extraction methods, only EZNA and Favorgen methods yielded DNA of sufficient quality for marker-assisted selection.

A Systematic Review of Clinical Studies Using VEGF Inhibitors in the Treatment of Macular Edema from Diabetic Retinopathy

Purpose: To synthesize the present clinical evidence of efficacy and adverse events of commonly used anti-VEGF drugs for Diabetic Macular Edema. Methods: A systematic review was undertaken from the Medline, Biosis, CINAHL, Cochrane and Web of Science databases. Grey literature that consisted of lectures, seminars and conferences was also retrieved. The cut-off date was January 1 2014. A two-stage screening process was undertaken followed by a data extraction stage using the systematic review software EPPI. These were done by two reviewers. Heterogeneous meta-analysis was performed on the primary outcome which was change in macular thickness from baseline after injection. Side effects were tabulated. Results: From 846 articles that were initially screened, 18 papers were included in the data extraction stage. For all anti-VEGF treatments, the average decrease in macular thickness was 114.4 microns (95% CI: 66.8 - 162 μM). The average decrease in thickness from Lucentis (161.9 μM) was larger than that for Avastin (96.5 μM) but this was not statistically significant (p = 0.23). The most common complications were vitreous hemorrhage, endophthalmitis and retinal detachment. Vision threatening complications were rare but were reported regularly. Conclusions: The synthesized clinical evidence to date supports both of these treatments as efficacious and safe for diabetic macular edema (DME). There is a trend toward greater efficacy for Lucentis over Avastin but this is not statistically significant and will need a head-to-head RCT to assess accurately.

Development and Evaluation of Nano-Silica Dispersed Polyurethane Based Coatings for Improved Anti-Graffiti and Scratch Resistance

Polyurethane based anti-graffiti coatings have been developed by incorporating varying amounts of OH-functional Silicone modified polyacrylate. Optimization of Silicone polyacrylate concentration in neat PU was done by testing its performance for graffiti resistance. Tests which include static contact angle measurements with water and spray paint, visual inspection of stain removal and color change measurements were performed. It was observed that 5 wt % of Silicone polyacrylate in the neat PU showed 95% stain removability. These coatings showed excellent graffiti resistance but poor mechanical properties. Hence modification of these coatings was done by nano-particle incorporation. Nano-silica particle concentration was optimized for surface mechanical properties such as surface hardness, elastic modulus and scratch resistance. FEG-TEM revealed that nano-silica particles were uniformly distributed over the surface. 5 wt % nano-silica particle additions in PU-5 wt % silicone polyacrylate base showed superior mechanical properties than other concentrations. Color change measurements showed that nano-silica particle addition had no effect on its graffiti resistance. Hence PU-5 wt % silicone polyacrylate, 5 wt % nano-silica showed excellent graffiti resistance and mechanical properties.

A rabbit model of non- cirrhotic portal hypertension by repeated injections of E. coli through indwelling cannulation of the gastrosplenic vein

BACKGROUND: Non-cirrhotic portal hypertension is acommon cause of portal hypertension in developing coun-tries. To understand its etiopathogenesis we developed ananimal model by repeated portal endotoxemia inducedthrough the gastrosplenic ***: Twenty-nine rabbits (1.5-2.0 kg) were divid-ed into control (group n = 13) and experimental ( groupn = 16) groups. Heat killed E. coli were injected throughan indwelling cannula into the gastrosplenic vein in pre-sensitized animals. The animals were sacriflced at 1, 3 and6 ***: The mean portal pressure in group animalswas significantly (P < 0. 05) higher than in group at 1(17.5 ±3.4 vs 10.4±2.2 mmHg), 3 (17.8±1.3 vs7.2 +3.6mmHg), and 6 (19.8±3.1 vs 10.3±4.8 mmHg) ***, the mean splenic weight in group was signifi-cantly greater than in group (P <0.05). Histopathologi-cally, the spleen showed medullary congestion, hemosid-rin-laden macrophages and mild fibrosis. Histologically,the liver had normal parenchyma with mild portal lympho-cytic infiltrates and kupffer cell hyperplasia. No significantanomalies were detected by liver function ***: The rabbit model showed significantsplenomegaly with a persistent increase in portal pressureand mild fibrosis without hepatic parenchymal injury, quiteakin to non-cirrhotic portal fibrosis as seen in humans. Re-current intra-abdominal infection may play an importantrole in the pathogenesis of non-cirrhotic portal fibrosis.

Standardization of DNA Extraction Method from Mature Dried Leaves and ISSR-PCR Conditions for Melia dubia Cav. —A Fast Growing Multipurpose Tree Species

Melia dubia Cav. of family Meliaceae is a fast growing, high value tree species native to India. Isolating DNA from matured dried leaves of M. dubia was difficult due to accumulation of secondary metabolites, majorly polyphenolics, which resulted in dark brown to black colour of the pellet. In this study, a modified STE-(Sucrose, Tris-HCl and Ethylene Diamine Tetra Acetic Acid) CTAB (hexadecyltrimethylammonium bromide) method was standardized for removal of polyphenolics. The protocol developed yielded 200 - 1000 ng/μl of quality DNA without any impurities as evident by A260/280 ratio ranging from 1.75 - 2.0. It was also suitable for extracting quality DNA from other members of Meliaceae like Azadirachta indica and Melia azedarach. In downstream applications, the extracted DNA was used for PCR amplification by using ISSR and SSR markers. ISSR PCR conditions were optimized in a reaction volume of 25 μl, consisting of 30 ng of template DNA, 1.5 mM MgCl2, 200 μM of each of dNTPs and 2 U of Taq polymerase. The best amplification was observed and the same was applicable for SSR markers.

A resonance-mode piezoelectric device for measurement of respiratory mechanics

This paper presents the design of a novel oscillometry device for the measurement of respiratory mechanics based on piezoelectric bimorph actuator technology. To predict performance for measurement of human respiratory mechanics, a dynamic model was developed based on a bimorph piezoelectric actuator driving a linear resistance mesh screen including subject’s respiratory impedance loads, and realistic breathing noise. Model performance was also validated in a prototype device. We found that while breathing noise substantially lowered SNR, the model could produce sufficient pressure and flow for acceptable SNRs exceeding 35 dB, and accuracies exceeding 99%. Satisfactory accuracy could be achieved with load impedance errors less than 3%. Maintaining the air-gap around the oscillating mesh with a resistance against the leak greater than 0.38 cmH2O/L/s maintained good performance, with an acceptable 4 dB decrease to SNR. Moreover, this work provides multiple solutions to host higher amounts of noise and nonlinearities. These results indicate that the development of an accurate lightweight portable single frequency FOT device is feasible.

The drug L-DOPA has been widely used against Parkinson's disease and is extracted from *** to the increasing demand of this drug,new plant sources need to be discovered in addition to the existing *** paper embodies results on Mucuna monosperma,which can be a promising candidate for *** seed powder of this plant contains 5.48%of(dry weight)the drug and when the seeds were soaked in distilled water,content was increased to 6.58%.Different elicitors when added,enhanced the drug level in seed up to 11.8%.The possible rationale behind this increase was confirmed by increase in tyrosinase activity in the *** of L-DOPA was confirmed using various analytical techniques as HPLC,HPTLC and *** work demonstrates a potential candidate plant as a source for L-DOPA when a novel method was adopted as described here.

背景:克罗恩病(CD)和肠结核(ITB)是有着相似表型的慢性肉芽肿性疾病,因此,找到一种生物标志物来鉴别这两种疾病是必要的。本研究旨在对ITB和CD的结肠标本进行全基因表达分析。同时,为了评价调节性T细胞的作用,我们对血清标本及结肠标本进行FOXP3 mRNA定量检测。方法:收集33名研究对象(CD患者、ITB患者及正常对照)血清和结肠组织配对样本,提取标本中的总RNA。采用Illumina HumanWG-6 BeadChip试剂盒对3组研究对象中的6种总RNA样本进行人类全基因组表达芯片分析。采用real-time PCR对具有合格RNA样本的病例(CD 4例,ITB 5例,正常对照4例)血清和组织标本进行FOXP3 mRNA表达水平检测。结果:在CD和ITB患者中,分别有92和382个基因表达上调1.5倍,91和256个基因表达下调1.5倍。过氧化物酶体增殖物介导的PPARγ信号通路在CD中下调最为显著(P<0.005);此外,IL4/5/6信号通路和Toll样受体信号通路也存在显著差异表达,上调2倍以上。而在ITB患者上调最为显著是补体活化通路尤其是经典通路。ITB患者结肠组织标本中FOXP3 mRNA表达水平明显高于CD患者(4.7062.21 vs 1.4860.31,P=0.016)。结论:结肠黏膜FOXP3 mRNA表达可作为鉴别ITB与CD的一项标志物。ITB患者补体激活通路的上调提示ITB的发病机制与肺结核相似。CD患者中PPARγ通路的下调提示PPARγ依赖的微生物屏障功能可能成为新的治疗靶点。

Nepeta cataria L., commonly known as catmint or catnip, belongs to the family “Lamiaceae” and is indigenous to Europe and Asia. The essential oil of this species is known for the richness and diversity of nepetalactones (NPL) which are used as mosquito/insect repellents in perfumery and cosmetic industries. Reports on Indian catmint germplasm are very meager and warrants exploration of its commercial potential as a natural, non-toxic source of insect repellents. With this objective, commercial open-pollinated seeds of catmint collected from its native, temperate habitat in Himalayas were introduced in the tropical plains. Subsequent to adaptation to a new zone we were able to isolate nineteen individual plants based on plant growth. Hydrodistillation of the fresh herb yielded essential oil in the range of 0.01% to 0.2%. Gas Chromatography (GC) and GC-Mass Spectrometry (GC-MS) analyses of the oil revealed the dominance of monoterpene hydrocarbon, namely, 4aα,7α,7aα NPL (1) isomer (84%). The other two isomers of nepetalactone, 4aα,7α,7aβ NPL (2) and 4aα,7β,7aα NPL (3) were also present, although in very less amounts (1.0% and 1.6%, respectively).
