Ron receptor-dependent gene regulation of Kupffer cells during endotoxemia

BACKGROUND: Ron receptor tyrosine kinase signaling in macrophages, including Kupffer cells and alveolar macrophages,suppresses endotoxin-induced proinflammatory cytokine/chemokine production. Further, we have also identified genes from Ron replete and Ron deplete livers that were differentially expressed during the progression of liver inflammation associated with acute liver failure in mice by microarray *** important genes and signaling pathways have been identified downstream of Ron signaling during progression of inflammation by this approach, the precise role that Ron receptor plays in regulating the transcriptional landscape in macrophages, and particular in isolated Kupffer cells, has still not been ***: Kupffer cells were isolated from wild-type(TK+/+)and Ron tyrosine kinase deficient(TK-/-) mice. Ex vivo, the cells were treated with lipopolysaccharide(LPS) in the presence or absence of the Ron ligand, hepatocyte growth factor-like protein(HGFL). Microarray and qRT-PCR analyses were utilized to identify alterations in gene expression between ***: Microarray analyses identified genes expressed differentially in TK+/+ and TK-/- Kupffer cells basally as well as after HGFL and LPS treatment. Interestingly, our studies identified Mefv, a gene that codes for the anti-inflammatory protein pyrin, as an HGFL-stimulated Ron-dependent ***, lipocalin 2, a proinflammatory gene, which is induced by LPS, was significantly suppressed by HGFL *** results were validated by qRT-PCR studies on Kupffer cells treated with LPS and ***: The studies herein suggest a novel mechanism whereby HGFL-induced Ron receptor activation promotes the expression of anti-inflammatory genes while inhibiting genes involved in inflammation with a net effect of diminished inflammation in macrophages.

Contents of four active components in different commercial crude drugs and preparations of Danshen (Salvia miltiorrhiza)

AIM: To detect the contents of four active components of Salvia miltiorrhiza in various commercially available danshen crude drugs and preparations. METHODS: Commercially available danshen crude drugs from different sources, as well as danshen pills and intravenous injection preparations containing danshen alone or in combination with other herbs were collected. The composition of these danshen samples was analyzed using HPLC. Specifically, the amounts of magnesium tanshinoate B (MTB), danshensu, isotanshinone IIA, and ctyptotanshinone were determined. In some of these samples, the content of MTB was further confirmed by liquid chromatography-tandem mass spectrometer (LC-MS)/MS method. RESULTS: There were great variations in the amount of the four active ingredients in the commercially available danshen crude drugs and drug preparations in this study. Aftershock of the Wenchuan Earthquake in Sichuan Triggers Large Earthquake in Minxian County, Gansu

Because of the confining compression of the Pacific, Eurasian and Indian plates, the Chinese mainland is frequently stricken with earthquakes, especially in the Qinghai Tibet Plateau and surrounding areas and along the NS-trending tectonic zone from Yinchuan to Lanzhou, Chengdu, and finally to the Kunming tectonic belt （Fig. 1）. Historical records show that there are 14 earthquakes of 〉Ms 8 occurring in the two regions, eight of which occurred along the latter tectonic belt. The 2008 Wenchuan earthquake located on both the NS tectonic zone and the Longmenshan fault zone along the eastern margin of the Qinghai-Tibet Plateau.

Phylogenetic relationships of native and introduced Bemisia tabaci (Homoptera:Aleyrodidae) from China and India based on mtCOI DNA sequencing and host plant comparisons

Phylogenetic relationships for Bemisia tabaci were reconstructed by analysis of a ～780 bp fragment of the mitochondrial cytochrome oxidase I (mtCOI) gene with an emphasis on geographic range and distribution among eight eudicot plant families that are common hosts of B. tabaci worldwide to elucidate key phylogeographic linkages between populations extant in China (n=31) and India (n=34). Bootstrap values for the Maximum Parsimony tree were highly robust for all major nodes involving the major Asian clade, subgroups, and sister groups within, at 92%—100%. Between-clade distances for the Southeast Asia and three other major clades, e.g. from sub-Sahara Africa, North Africa-Mediterranean, and the Americas, were approximately >16% divergent. Two major Asian subgroups (I, II) were resolved, which represented populations indigenous to the region, comprising two (I a, I b) and five (II a—e) sister groups, respectively, which diverged by 11%. Two distinct populations from sunflower in Hyderabad grouped separately within the two Asian subgroups. All other populations grouped uniquely within Asian subgroup II or I. The "B" biotype was identified in 23 collections from China at 97.3%—99.5% nucleotide identity with "B" biotype reference sequences; it was not identified in collections from India. The majority of haplotypes were associated with 3—4 plant families, with one exception that for sister group IId (sesame, India), it might be monophagous. Thus, B. tabaci from the southeastern and near eastern regions of the Asian continent comprise of a large number of ancestral, richly divergent, mostly polyphagous populations. This region is therefore hypothesized to constitute an important Old World center of diversification for the B. tabaci complex, together with sub-Saharan Africa.

Next Generation Transcriptome Sequencing and Quantitative Real-Time PCR Technologies for Characterisation of the Bemisia tabaci Asia 1 mtCOI Phylogenetic Clade

A programme of functional genomics research is underway at the University of Greenwich,UK,to develop and apply genomics technologies to characterise an economically-important but under-researched Bemisia tabaci（Hemiptera：Aleyrodidae）,the Asia 1 mtCOI phylogenetic group.A comparison of this putative species from India with other important *** populations and insect species may provide targets for the development of more effective whitefly control *** a first step,next-generation sequencing（NGS）has been used to survey the transcriptome of adult female whitefly,with high quality RNA preparations being used to generate cDNA libraries for NGS using the Roche 454 Titanium DNA sequencing *** assemblies constructed from the resultant sequences（301 094 reads）using the software program CLC Genomics Workbench generated 3 821 core *** of a selection of these contigs with related sequences from other *** genetic groups has revealed good alignment for some genes（e.g.,HSP90）but misassemblies in other datasets（e.g.,the vitellogenin gene family）,highlighting the need for manual curation as well as collaborative international efforts to obtain accurate assemblies from the existing next generation sequence ***,data emerging from the NGS has facilitated the development of accurate and reliable methods for analysing gene expression based on quantitative real-time RT-PCR,illustrating the power of this approach to enable rapid expression analyses in an organism for which a complete genome sequence is currently lacking.

Influence of Soil Properties on Zinc Solubility Dynamics Under Different Redox Conditions in Non-Calcareous Soils

Zinc(Zn) deficiency in paddy soils is often a problem for rice *** can decrease metal availability in some noncalcareous soils through different mechanisms associated with soil redox *** experiments were performed in order to better understand the processes that governed the dynamics of Zn in non-calcareous paddy soils at varying redox potentials(Eh).Airdried non-calcareous soil samples collected from four different paddy field sites in the Philippines were submerged and incubated in a reaction cell with continuous stirring and nitrogen purging for 4 weeks,and then purged with compressed air for another week to reoxidize the *** Eh of the four soils started at 120 to 300 mV,decreased to —220 to —300 mV after 100 to 250 h of reduction,and was maintained at this low plateau for about 2 weeks before increasing again upon *** solubility showed contrasting patterns in the four soils,with two of the soils showing a decrease in soluble Zn as the Eh became low,probably due to zinc sulfide(ZnS) *** contrast,the other two soils showed that Zn solubility was maintained during the reduced phase which could be due to the competition with iron(Fe) for precipitation with *** in the relative amounts of S,Fe,and manganese(Mn) oxides in the four soils apparently influenced the pattern of Zn solubility after flooding.

Preparation and Properties of Poly(vinyl alcohol)/Chitosan Blend Bio-nanocomposites Reinforced by Cellulose Nanocrystals

The aim of this paper is to report the effect of the addition of cellulose nanocrystals（CNCs） on the mechanical, thermal and barrier properties of poly（vinyl alcohol）/chitosan（PVA/Cs） bio-nanocomposites films prepared through the solvent casting process. The characterizations of PVA/Cs/CNCs films were carried out in terms of X-ray diffraction（XRD）, transmission electron microscopy（TEM）, scanning electron microscopy（SEM）, thermogravimetric analysis（TGA and DTG）, oxygen transmission rate（OTR）, and tensile tests. TEM and SEM results showed that at low loading levels, CNCs were dispersed homogenously in the PVA/Cs matrix. The tensile strength and modulus in films increased from 55.1 MPa to 98.4 MPa and from 395 MPa to 690 MPa respectively, when CNCs content went from 0 wt% to 1.0 wt%. The thermal stability and oxygen barrier properties of PVA/Cs matrix were best enhanced at 1.0 wt% of CNCs loading. The enhanced properties attained by incorporating CNCs can be beneficial in various applications.
