Effects of Melandrium firmum methanolic extract on testosterone-induced benign prostatic hyperplasia in Wistar rats

Benign prostatic hyperplasia （BPH） is an age-related disease of unknown aetiology characterized by prostatic enlargement coincident with distinct alterations in tissue histomorphology. Instead of therapeutic agents that can cause severe side effects, plant extracts are frequently used to treat BPH. In this study, we investigated whether the Melandrium firmum methanolic extract （MFME） improves BPH, using the testosterone propionate （TP）-induced BPH rat model. Castration was performed via the scrotal route under sodium pentobarbital anaesthesia. BPH in castrated rats was generated via daily subcutaneous injections of TP （3 mg kg-1） dissolved in corn oil, for 4 weeks. MFME was administered daily by oral garage at a dose of 200 mg kg-1 for 4 weeks, along with the TP injections. The control group received injections of corn oil subcutaneously. At the scheduled termination of the experiment, all rats were killed and their prostates weighed; the relative prostate weight （prostate/body weight ratio） was calculated, and histomorphological changes in the prostate were examined. Additionally, we measured the levels of testosterone and dihydrotestosterone （DHT） in the serum and the prostate. Experimentally induced BPH led to marked decreases in the relative prostate weight and the DHT levels in the serum and the prostate. Histologically, BPH was evident in the ventral lobe of the prostate, and MFME treatment suppressed the severity of the lesions. These results indicate that MFME effectively inhibits the development of BPH induced by testosterone in a rat model. Further studies will be needed to identify the compound（s） responsibility for inducing the protective effect against BPH and determine its mechanism of action,

Treatment of pediatric average-risk medulloblastoma using craniospinal irradiation less than 2500 cGy and chemotherapy: single center experience in Korea

Background:Although craniospinal irradiation (CSI) of 2340 cGy plus tumor booster with chemotherapy have been established as a standard treatment of childhood average-risk (AvR) medulloblastoma (MBL) in Western counties,there are a few recent reports in outcomes ofAvR MBL using this strategy in Korean and other Asian *** investigated the outcome of the Korean children with AvR MBL who were treated with CSI <2500 cGy and ***:Between January 2001 and December 2010,clinical characteristics and outcomes of 42 patients who were diagnosed with AvR MBL postoperatively and treated with radiation including CSI <2500 cGy and chemotherapy in Seoul National University Children's Hospital were ***:Their median age was 9 years (range:3-18.8),and 29 were *** subtypes were classic type in 28 patients,nodular/desmoplastic in 7,and large cell/anaplastic (LCA) in *** the patients received adjuvant radiotherapy (CSI with median 2340 cGy and booster) and multiagent chemotherapy as the first-fine *** a median follow-up of 54 months,12 patients experienced relapse or progression of the *** 3-and 5-year disease-free survival (DFS) rates were 78.0%±6.5% and 75.0%±6.9%,respectively,and overall survival (OS) rates were 85.3%±5.6% and 76.8%±6.9%,*** LCA subtype was associated with poorer DFS (P=0.023) and OS (P=0.008),compared with non-LCA ***:The outcomes of children and adolescents with AvR MBL treated with radiation including CSI <2500 cGy and chemotherapy,are compatible to those in Western countries;however,the LCA subtype has a poor outcome with this strategy.

Influence of forest road on breeding of tits in artificial nest boxes

A study was conducted to determine the influence of forest road on breeding of tits in artificial nest boxes in deciduous, coniferous and mixed forests in the Gwanak Arboretum （37° 25′ 05＂ N, 126° 56′ 85＂ E） of Seoul National University, Anyang, Korea from November 2002 to June 2003. Three tits species, varied tit （Parus varius）, marsh tit （P. palustris） and great tit （P. major）, breeding in artificial t nest boxes were investigated on number of breeding pairs, cultch size, and egg measurement. Resuls showed that the breeding pairs of varied tit was more in 75-150 m area than in 0-75m area from forest road for all the three study sites, and the clutch size and egg measurements （weight, Major axis and Minor axis） of varied tit was also higher in the area of 75-150 m than in the area of 0-75 m, while no differences in number of breeding pairs and clutch size were found for marsh tit and great tit between the two areas. Egg measurement of great tit was also higher in forest interior area than in forest edge area. It is concluded that varied tit were most significantly influenced by forest road, followed by great tit, whereas marsh tit were not influenced by forest road. Artificial nest box is roved to be good for cavity nester in disturbed areas by human activities. Supply of artificial nest can help population protection and management of bird species.

MRI of gastric carcinoma:Results of T and N-staging in an in vitro study

AIM： To determine the accuracy of 1.5-T magnetic resonance imaging （MRI） in the evaluation of gastric wall invasion and perigastric lymph node metastasis in gastric adenocarcinoma.METHODS： Twenty resected gastric specimens containing 20 tumors were studied with a 1.5-T MR system using a commercial head surface coil. MR scanning was performed with a T1 weighted image （TR/TE = 500/20）, and a T2 weighted image （TR/TE = 2500/90）. MR findings were compared with pathologic findings.RESULTS： A T1-weighted image demonstrated three layers in the normal gastric wall. All of the gastric tumors were well demonstrated by lesions and location. In a MRI findings of gastric wall invasion, there was 1 case of T1, 7 of T2, 11 of T3. Pathologic results of resected specimens included 3 cases of pT1, 4 of pT2, and 12 of pT3. The accuracy of T staging with MRI was 74% （14 of 19）. MRI findings of lymph node metastasis included 6 cases of NO, 13 cases of N1. The accuracy of the N staging with MRI was 47% （9 of 19）.CONCLUSION： MRI has a high diagnostic accuracy in the evaluation of the T staging of gastric cancerin vitro and thus potentially enables preoperative histopathologic staging.

Background:Prenatal maternal diet may influence disease susceptibility in offspring with specific genetic *** hypothesized that interactions between prenatal antioxidant intake and polymorphisms in immunity genes influence respiratory tract infection (RTI) susceptibility in infants at 12 months of ***:This study included 550 *** the Cohort for Childhood Origin of Asthma and Allergic Diseases (COCOA) birth cohort study,prenatal maternal diet was assessed by administering a food frequency ***' cord blood was genotyped for CD14 (rs2569190),TLR4 (rs1927911),and GSDMB (rs4794820) polymorphisms by the TaqMan ***:Higher prenatal intake of total fruit and vegetables (FV) was associated with the decreased risk of RTI in offspring (P-trend=0.0430).In children with TT genotype at rs2569190,a higher prenatal intake of vitamins A and C,fruits,and total FV decreased RTI risk (P-trend <0.05),while in infants with TC+CC genotype,a higher prenatal intake of fruit increased RTI risk (P-trend <0.05).When analyzing the 3 genotypes,children with TT genotype at rs2569190 were more protected against RTIs compared with those with CC genotype with respect to vitamin C and fruits [odds ratio (OR)=5.04 and OR=10.30,respectively].In children with CC genotype at rs1927911,RTI risk showed a dose-response association with a higher prenatal intake of vitamin C (P for interaction<0.05).A higher prenatal intake of fruits and total FV reduced RTI risk in infants with GA+AA genotype of rs4794820 (P for interaction<0.05).Conclusion:Prenatal antioxidant intake may reduce RTI risk in infants and this relationship may be modified by CD14,TLR4,and GSDMB polymorphisms.

Long-term culture-induced phenotypic difference and efficient cryopreservation of small intestinal organoids by treatment timing of Rho kinase inhibitor

To investigate a suitable long-term culture system and optimal cryopreservation of intestinal organoid to improve organoid-based therapy by acquiring large numbers of *** were isolated from jejunum of C57BL/6 mouse. Two hundred crypts were cultured in organoid medium with either epidermal growth factor/Noggin/R-spondin1 (ENR) or ENR/CHIR99021/VPA (ENR-CV). For subculture, organoids cultured on day 7 were passaged using enzyme-free cell dissociation buffer (STEMCELL Technologies). The passage was performed once per week until indicated passage. For cryopreservation, undissociated and dissociated organoids were resuspended in freezing medium with or without Rho kinase inhibitor subjected to different treatment times. The characteristics of intestinal organoids upon extended passage and freeze-thaw were analyzed using EdU staining, methyl thiazolyl tetrazolium assay, qPCR and time-lapse live cell *** established a three-dimensional culture system for murine small intestinal organoids using ENR and ENR-CV media. Both conditions yielded organoids with a crypt-villus architecture exhibiting Lgr5+ cells and differentiated intestinal epithelial cells as shown by morphological and biochemical analysis. However, during extended passage (more than 3 mo), a comparative analysis revealed that continuous passaging under ENR-CV conditions, but not ENR conditions induced phenotypic changes as observed by morphological transition, reduced numbers of Lgr5+ cells and inconsistent expression of markers for differentiated intestinal epithelial cell types. We also found that recovery of long-term cryopreserved organoids was significantly affected by the organoid state, i.e., whether dissociation was applied, and the timing of treatment with the Rho-kinase inhibitor Y-27632. Furthermore, the retention of typical morphological characteristics of intestinal organoids such as the crypt-villus structure from freeze-thawed cells was observed

Random lasing detection of structural transformation and compositions in silk fibroin scaffolds

In tissue engineering, microstructure and material composition of tissue scaffolds have major influences on the proliferation and differentiation of cells in the scaffolds. However, once tissue scaffolds implanted, it is extremely difficult to monitor the change of their microstructure and compositions during tissue regeneration. Here, we report how random lasing can be utilized to non-invasively monitor the structure and composition of scaffolds. We hypothesize that morphological and compositional change of silk fibroin (SF) scaffolds can be conveniently detected based on random lasing responses. Engineered SF scaffolds with hydroxyapatite (HAP) nanoparticles and controlled pore alignment were fabricated, and their random lasing responses were analyzed depending on the concentration of HAP nanoparticles and the degree of,:internal pore alignment. We also examined the real-time random lasing responses of porous SF scaffolds by applying a compressive force to' the scaffolds. Introduction of HAP nanoparticles lowered the lasing thresholds and narrowed the random lasing (RL) width dramatically, which :is likely due to the increase in heterogeneity in both refractive index and physical arrangement within the SF and HAP composites. The strong dependency of RL response on pore alignment was also measured and validated by numerical calculation with the finite element method (FEM).Finally, real-time monitoring of RL on compressed scaffolds demonstrated the possibility of using RL as a monitoring tool for structural change of SF scaffolds in vivo.

Neuroprotective effects of tamsulosin on intracerebral hemorrhage

The alpha1 adrenoreceptor antagonist tamsulosin is primarily used to treat benign prostatic hypertrophy. In this study, we intragastrically administered rats with 0.01,0.1 and 1 mg/kg tamsulosin to investigate the effects of tamsulosin on memory function, and hippocampal cell apoptosis and proliferation following intracerebral hemorrhage. Step-down avoidance task results showed that tamsulosin treatment markedly alleviated intracerebral hemorrhage- induced short-term memory impairment. Terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling results showed that tamsulosin treatment markedly suppressed intracerebral hemorrhage-induced caspase-3 expression in the hippocampal CA1 region and cell proliferation in the hippocampal dentate gyrus. Tamsulosin treatment at a dose of 0.01 mg/kg exhibited the most potent inhibitory effects on cell apoptosis and proliferation. These findings suggest that tamsulosin treatment facilitates the recovery of rat memory function by inhibiting hippocampal cell apoptosis and proliferation followincl intracerebral hemorrhaQe.

Association between cytochrome CYP17A1, CYP3A4, and CYP3A43 polymorphisms and prostate cancer risk and aggressiveness in a Korean study population

In this study, we evaluated genetic variants of the androgen metabolism genes CYP17A1, CYP3A4, and CYP3A43 to determine whether they play a role in the development of prostate cancer （PCa） in Korean men. The study population included 240 pathologically diagnosed cases of PCa and 223 age-matched controls. Among the 789 singie-nucleotide polymorphism （SNP） database variants detected, 129 were reported in two Asian groups （Han Chinese and Japanese） in the HapMap database. Only 21 polymorphisms of CYP17A1, CYP3A4, and CYP3A43 were selected based on linkage disequilibrium in Asians （d = 1）, locations （SNPs in exons were preferred）, and amino acid changes and were assessed. In addition, we performed haplotype analysis for the 21 SNPs in CYP17A1, CYP3A4, and CYP3A43 genes. To determine the association between genotype and haplotype distributions of patients and controls, logistic analyses were carried out, controlling for age. Twelve sequence variants and five major haplotypes were identified in CYP17A1. Five sequence variants and two major haplotypes were identified in CYP3A4. Four sequence variants and four major haplotypes were observed in CYP3A43. CYP17A1 haplotype-2 （Ht-2）（odds ratio [OR], 1.51; 95% confidence interval [Cl], 1.04-2,18） was associated with PCa susceptibility. CYP3A4 Ht-2 （OR. 1.87; 95% CI. 1.02-3.43） was associated with PCa metastatic potential according to tumor stage, rs17115149 （OR： 1.96; 95% Ch 1.04-3.68） and CYP17A1 Ht-4 （OR： 2.01; 95% Ch 1.07-4.11） showed a significant association with histologic aggressiveness according to Gleason score. Genetic variants of CYP17A1 and CYP3A4 may play a role in the development of PCa in Korean men.
