Ron recep.or-dep.ndent gene regulation of Kup.fer cells during endotoxemia

BACKGROUND: Ron recep.or tyrosine kinase signaling in macrop.ages, including Kup.fer cells and alveolar macrop.ages,sup.resses endotoxin-induced p.oinflammatory cytokine/chemokine p.oduction. Further, we have also identified genes from Ron rep.ete and Ron dep.ete livers that were differentially exp.essed during the p.ogression of liver inflammation associated with acute liver failure in mice by microarray *** imp.rtant genes and signaling p.thways have been identified downstream of Ron signaling during p.ogression of inflammation by this ap.roach, the p.ecise role that Ron recep.or p.ays in regulating the transcrip.ional landscap. in macrop.ages, and p.rticular in isolated Kup.fer cells, has still not been ***: Kup.fer cells were isolated from wild-typ.(TK+/+)and Ron tyrosine kinase deficient(TK-/-) mice. Ex vivo, the cells were treated with lip.p.lysaccharide(Lp.) in the p.esence or absence of the Ron ligand, hep.tocyte growth factor-like p.otein(HGFL). Microarray and qRT-p.R analyses were utilized to identify alterations in gene exp.ession between ***: Microarray analyses identified genes exp.essed differentially in TK+/+ and TK-/- Kup.fer cells basally as well as after HGFL and Lp. treatment. Interestingly, our studies identified Mefv, a gene that codes for the anti-inflammatory p.otein p.rin, as an HGFL-stimulated Ron-dep.ndent ***, lip.calin 2, a p.oinflammatory gene, which is induced by Lp., was significantly sup.ressed by HGFL *** results were validated by qRT-p.R studies on Kup.fer cells treated with Lp. and ***: The studies herein suggest a novel mechanism whereby HGFL-induced Ron recep.or activation p.omotes the exp.ession of anti-inflammatory genes while inhibiting genes involved in inflammation with a net effect of diminished inflammation in macrop.ages.

In vitro investigation of the effect of dairy p.op.onibacteria on rumen p.,lactic acid and volatile fatty acids

Ruminal acidosis is a p.evalent disorder in ruminants such as dairy cows and feedlot beef cattle, caused p.imarily by the inclusion of a high p.rcentage of readily fermentable concentrates in the diet. The disorder p.esents as an accumulation of lactic acid, a decrease of p. in the rumen and a subsequent imbalance of the rumen fermentation p.ocess with detrimental imp.cts on the animal＇s health and p.oductivity. Dairy p.op.onibacteria, a group.of bacteria characterised by utilization of lactic acid as the favoured carbon source, with p.op.onic acid p.oduced as a by-p.oduct, were evaluated in this study as p.tential direct-fed microbials for use in controlling ruminal acidosis. Acidosis was simulated by introduction of high concentrations of lactic acid into rumen fluid samp.es and a multi-strain in vitro analysis was conducted, whereby changes in p. and lactic acid metabolism were comp.red in identical acidified rumen samp.es, following inoculation with various p.op.onibacteria. This was followed by a study to evaluate the effect of bacterial inoculation dosage on acid metabolism. The results indicated that lactic acid levels in the rumen fluid were significantly reduced, and p.op.onic acid and acetic acid concentrations both significantly increased, following addition of p.op.onibacteria. Significant ＇between strains＇ differences were observed, with p.op.onibacterium acidop.op.onici 341, p.op.onibacterium freudenreichfi CSCC 2207, p.op.onibacterium jensenfi NCFB 572 and p. jensenii 702 each p.oducing more rap.d reduction of lactic acid concentration than p. freudenreich# CSCC 2206, p. acidop.op.onici ATCC 25562 and p.op.onibacterium thoenii ATCC 4874. Furthermore, the efficacy of this ap.lication was dosage related, with the rates of reduction in lactic acid levels and p.oduction of p.op.onic acid, both significantly greater for the higher （10^10 cfu mL-1） comp.red with lower （10^5 cfu mL-1） dosage inoculation. Statistical analysis of recombination p.op.rties of the boronoxygen defect in p.typ. Czochralski silicon

This p.p.r p.esents the ap.lication of lifetime sp.ctroscop. to the study of carrier-induced degradation ascribed to the boron-oxygen （BO） defect. Sp.cifically, a large data set of p.typ. silicon samp.es is used to investigate two imp.rtant asp.cts of carrier lifetime analysis： ① the methods used to extract the recombination lifetime associated with the defect and ② the underlying assump.ion that cartier injection does not affect lifetime comp.nents unrelated to the defect. The results demonstrate that the cap.ure cross section ratio associated with the donor level of the BO defect （kl） vary widely dep.nding on the sp.cific method used to extract the defect-sp.cific recombination lifetime. For the data set studied here, it was also found that illumination used to form the defect caused minor, but statistically significant changes in the surface p.ssivation used. This violation of the fundamental assump.ion could be accounted for by ap.lying ap.rop.iate curve fitting methods, resulting in an imp.oved estimate of k1 （11.90±0.45） for the fully formed BO defect when modeled using the donor level alone. Illumination also ap.eared to cause a minor, ap.arently injectionindep.ndent change in lifetime that could not be attributed to the donor level of the BO defect alone and is likely related to the accep.or level of the BO defect. While sp.cific to the BO defect, this study has imp.ications for the use of lifetime sp.ctroscop. to study other carrier induced defects. Finally, we demonstrate the use of a unit-less regression goodness-of-fit metric for lifetime data that is easy to interp.et and accounts for rep.atability error.

AIMS The p.rp.se of this study was to determine the relationship.between p.lmonary vein (p.) electrical activation during atrial fibrillation (AF) and after cardioversion into sinus rhythm. Methods and Results Electrograms were recorded using a circular map.ing catheter during AF and after cardioversion in 53 p.s from 41 p.tients. Geograp.ically distributed hybrid testing & collaboration between geotechnical centrifuge and structures laboratories

Distributed Hybrid Testing（DHT） is an exp.rimental technique designed to cap.talise on advances in modern networking infrastructure to overcome traditional laboratory cap.city limitations. By coup.ing the heterogeneous test ap.aratus and comp.tational resources of geograp.ically distributed laboratories, DHT p.ovides the means to take on comp.ex, multi-discip.inary challenges with new forms of communication and collaboration. To introduce the op.ortunity and p.acticability afforded by DHT, here an exemp.ar multi-site test is addressed in which a dedicated fibre network and suite of custom software is used to connect the geotechnical centrifuge at the University of Cambridge with a variety of structural dynamics loading ap.aratus at the University of Oxford and the University of Bristol. While centrifuge time-scaling p.events real-time rates of loading in this test, such exp.riments may be used to gain valuable insights into p.ysical p.enomena, test p.ocedure and accuracy. These and other related exp.riments have led to the develop.ent of the real-time DHT technique and the creation of a flexible framework that aims to facilitate future distributed tests within the UK and beyond. As a further examp.e, a real-time DHT exp.riment between structural labs using this framework for testing across the Internet is also p.esented.
