Metabolic and performance responses of male runners wearing 3 types of footwear: Nike Vaporfly 4%, Saucony Endorphin racing flats, and their own shoes

Purpose We compared running economy(RE)and 3-km time-trial(TT)variables of runners wearing Nike Vaporfly 4%(VP4),Saucony Endorphin lightweight racing flats(FLAT),and their habitual running(OWN)*** Eighteen male recreational runners(age=33.5±11.9 year(mean±SD),peak oxygen uptake(VO_(2peak))=55.8±4.4 mL/kg·min)attended 4 sessions approximately 7 days *** first session consisted of a VO_(2peak) test to inform subsequent RE speeds set at 60%,70%,and 80%of the speed eliciting *** subsequent sessions,treadmill RE and 3-km TTs were assessed in the 3 footwear conditions in a randomized,counterbalanced crossover *** Oxygen consumption(mL/kg·min)was less in VP4(from 4.3%to 4.4%,p≤0.002)and FLAT(from 2.7%to 3.4%,p≤0.092)*** across intensities,with a non-significant difference between VP4 and FLAT(1.0%-1.7%,p≥0.292).Findings related to energy cost(W/kg)and energetics cost of transport(j/kg·m)were ***43-km TT performance(11:07.6±0:56.6 mm:ss)was enhanced *** by 16.6 s(2.4%,p=0.005)and *** by 13.0 s(1.8%,p=0.032).The 3-km times between OWN and FLAT(0.5%,p=0.747)were *** runners(n=11,61%)ran their fastest TT in *** Overall,VP4 improved laboratory-based RE measures in male recreational runners at relative speeds compared to OWN,but the RE improvements in VP4 were not significant *** runners exhibited better treadmill TT performances in VP4(61%)***(22%)and OWN(17%).The variability in RE(-10.3%to 13.3%)and TT(-4.7%to 9.3%)improvements suggests that responses to different types of shoes are individualized and warrant further investigation.

An integrative multivariate approach for predicting functional recovery using magnetic resonance imaging parameters in a translational pig ischemic stroke model

Magnetic resonance imaging(MRI)is a clinically relevant,real-time imaging modality that is frequently utilized to assess stroke type and ***,specific MRI biomarkers that can be used to predict long-term functional recovery are still a critical ***,the present study sought to examine the prognostic value of commonly utilized MRI parameters to predict functional outcomes in a porcine model of ischemic *** was induced via permanent middle cerebral artery *** 24 hours post-stroke,MRI analysis revealed focal ischemic lesions,decreased diffusivity,hemispheric swelling,and white matter *** deficits including behavioral abnormalities in open field and novel object exploration as well as spatiotemporal gait impairments were observed at 4 weeks *** graphical models identified specific MRI outputs and functional recovery variables,including white matter integrity and gait performance,that exhibited strong conditional *** correlation analysis revealed a prognostic relationship between lesion volume and white matter integrity and novel object exploration and gait ***,these analyses may also have the potential of predicting patient recovery at chronic time points as pigs and humans share many anatomical similarities(e.g.,white matter composition)that have proven to be critical in ischemic stroke *** study was approved by the University of Georgia(UGA)Institutional Animal Care and Use Committee(IACUC;Protocol Number:A2014-07-021-Y3-A11 and 2018-01-029-Y1-A5)on November 22,2017.

Harmony Search(HS)algorithm is highly effective in solving a wide range of real-world engineering optimization ***,it still has the problems such as being prone to local optima,low optimization accuracy,and low search *** address the limitations of the HS algorithm,a novel approach called the Dual-Memory Dynamic Search Harmony Search(DMDS-HS)algorithm is *** main innovations of this algorithm are as follows:Firstly,a dual-memory structure is introduced to rank and hierarchically organize the harmonies in the harmony memory,creating an effective and selectable trust region to reduce approach blind ***,the trust region is dynamically adjusted to improve the convergence of the algorithm while maintaining its global search ***,to boost the algorithm’s convergence speed,a phased dynamic convergence domain concept is introduced to strategically devise a global random search ***,the algorithm constructs an adaptive parameter adjustment strategy to adjust the usage probability of the algorithm’s search strategies,which aim to rationalize the abilities of exploration and exploitation of the *** results tested on the Computational Experiment Competition on 2017(CEC2017)test function set show that DMDS-HS outperforms the other nine HS algorithms and the other four state-of-the-art algorithms in terms of diversity,freedom from local optima,and solution *** addition,applying DMDS-HS to data clustering problems,the results show that it exhibits clustering performance that exceeds the other seven classical clustering algorithms,which verifies the effectiveness and reliability of DMDS-HS in solving complex data clustering problems.

Impacts of 1-Aminocyclopropane-1-Carboxylic Acid as a Late Post-Bloom Thinner on Fruit Set, Yield, and Fruit Quality in “Gala” and “Fuji” Apples

A few post-bloom thinners are available for apple (Malus domestica Borkh.) and the prospects for additional thinners appear to be limited. Application of blossom thinners may present a risk of overthining in the areas where weather is less predictable. Thus, we studied the impacts of various rates of 1-aminocyclopropane-1-carboxylic acid (ACC) in three strains of “Fuji” apple and different rates of ACC and one rate of Ethrel in “Buckeye Gala” apple, when fruitlet diameter was about 20 mm, on fruit set, yield and fruit quality attributes at harvest, and return bloom in southwest Idaho in the Intermountain West region, USA. In 2013, application of Ethrel at 300 mg·L-1 did not affect fruit set, fruit weight, diameter (D), length(L), or L/D ratio 34 days after application, or yield, fruit weight, color, russet, and starch degradation pattern (SDP) at harvest, while application of ACC at 150 mg·L-1 or higher reduced fruit set by 19% to 34% in “Buckeye Gala” apple. In this cultivar, application of ACC at 350 mg·L-1 significantly increased fruit weight, diameter and length 34 days after application, and increased fruit weight, color, and SDP at harvest time. Application of ACC at all rates reduced total yield per tree in “Buckeye Gala”. Application of ACC at 300 mg·L-1 significantly reduced fruit set but applications of 150 mg·L-1 or 300 mg·L-1 ACC did not affect yield or quality attributes of “Sun Fuji” apple in Sunny Slope area in 2013. Application of ACC reduced fruit set and slightly increased fruit size in “Top Export Fuji” in 2013. Application of ACC at 600 mg·L-1 significantly reduced fruit set in “Aztec Fuji” apple in 2014. Application of ACC in a season never reduced bloom density (return bloom) of the next season. Overall, we conclude that ACC is an excellent tool as a late-season post-bloom fruit thinner and can be effective when applied

Activation of Plasma Membrane NADPH Oxidase and Generation of H_2O_2 Mediate the Induction of PAL Activity and Saponin Synthesis byEndogenous Elicitor in Suspension-Cultured Cells of Panax ginseng

Endogenous elicitor, termed cellulase-degraded cell wall (CDW), was prepared from the cell wall of suspension-cultured ginseng (Panax ginseng C.A. Meyer) cells via cellulase degradation. CDW activated the NADPH oxidase activity of isolated plasma membranes and stimulated in vivo H2O2 generation in ginseng cell suspensions. CDW also increased the activity of phenylalanine ammonia lyase (PAL), expression of a P. ginseng squalene epoxidase (sqe) gene and saponin synthesis. NADPH oxidase inhibitors inhibited both in vitro NADPH oxidase activity and in vivo H2O2 generation. Induction of PAL activity, saponin synthesis and sqe gene expression were all inhibited by such inhibitor treatments and reduced by incubation with catalase and HA scavengers. These data indicate that activation of NADPH oxidase and generation of H2O2 are essential signalling events mediating defence responses induced by the endogenous elicitor(s) present in CDW.

AIM: To investigate modulation of antagonist and agonist binding to adenosine A1receptors by MgCl2and 5'-guanylimidodiphosphate (Gpp（NH）p) using rat brain membranes and the A1antagonist [3H]-8-cyclopentyl-1,3-dipropylxanthine （[3H]DPCPX） and the A1agonist [3H]-2-chloro-N6-cyclopentyladenosine （[3H]CCPA）. METHODS: Parallel saturation and inhibition studies were performed using well-characterised radioligand binding assays and a Brandel Cell Harvester. RESULTS: MgCl2produced a concentration-dependent decrease （44%）, whereas Gpp（NH）p increased [3H]DPCPX binding （19%）. In [3H]DPCPX competition studies, agonist affinity was 1.5-14.6-fold higher and 4.6-10-fold lower in the presence of 10 mmol/L MgCl2and 10 μmol/L Gpp（NH）p respectively; antagonist affinity was unaffected. The decrease in agonist affinity with increasing Gpp（NH）p concentrations was due to a reduction in the proportion of binding to the high affinity receptor state. In this study, we report the fabrication of engineered iron oxide magnetic nanoparticles (MNPs)functionalized with anti-human epidermal growth factor receptor type 2 (HER2) antibody totarget the tumor antigen HER2. The Fc-directed conjugation of antibodies to the MNPs aidstheir efficient immunospecific targeting through free Fab portions. The directional specificity ofconjugation was verified on a macrophage cell line. Immunofluorescence studies on macrophagestreated with functionalized MNPs and free anti-HER2 antibody revealed that the antibodymolecules bind to the MNPs predominantly through their Fc portion. Different cell lines with different HER2 expression levels were used to test the specificity of our functionalized nanoprobe formolecular targeting applications. The results of cell line targeting demonstrate that these engineered MNPs are able to differentiate between cell lines with different levels of HER2 expression.

The impact of COVID-19 on clinical outcomes of burn patients

Background:Multiple studies have shown the SARS-CoV-2 virus(COVID-19)to be associated with deleterious outcomes in a wide range of *** impact of COVID-19 has not been well investigated among burned *** suspect that patients will have worsened respiratory and thrombotic complications,ultimately leading to increased *** objective of this study is to determine the impact a concurrent infection of COVID-19 has on clinical outcomes after a burn ***:This is a retrospective,propensity matched,cohort *** examined a de-identified database of electronic medical records of over 75 million patients across 75 health care associations in the United States for patients treated for thermal burns from 1 january 2020,to 31 july 2021,and those who also were diagnosed with COVID-19 infection within one day before or after injury based on International Classification of Disease,tenth revision(ICD-10)*** participants included adults who were treated for a burn injury during the study ***:We included 736 patients with burn injury and concomitant COVID-19 infection matched to 736 patients with burn injury and no concurrent COVID-19 infection(total 1472 patients,mean age 36.3±24.3).We found no significant increase in mortality observed for patients with concurrent COVID-19(OR 1.203,95%CI 0.517-2.803;p=0.6675).We did observe significant increase in infections(OR 3.537,95%CI 2.798-4.471;p=0.0001),thrombotic complications(OR 2.342,95%CI 1.351-4.058;p=0.0018),as was the incidence of hypertrophic scarring(OR 3.368,95%CI 2.326-4.877;p=0.0001).Conclusions:We observed that concurrent COVID-19 infection was associated with an increase in infections,thrombosis and hypertrophic scarring but no increase in mortality in our cohort of burn patients.
