Arabidopsis thal.ana l. yel.ow variegated （var2） mutant is defective in a chl.ropl.st FtsH famil. metal.oprotease, AtFtsH2/VAR2, and displ.ys an intriguing green and white l.af variegation. This unique var2-mediated l.af variegation offers a simpl. yet powerful.tool.for dissecting the genetic regul.tion of chl.ropl.st devel.pment. Here, we report the isol.tion and characterization of a new var2 suppressor gene, SUPPRESSOR OF VARIEGA TION8 （SVR8）, which encodes a putative chl.ropl.st ribosomal.l.rge subunit protein, l.4. Mutations in SVR8 suppress var2 l.af variegation at ambient temperature and partial.y suppress the col.-induced chl.rosis phenotype of var2. l.ss of SVR8 causes unique chl.ropl.st rRNA processing defects, particul.rl. the 23S-4.5S dicistronic precursor. The recovery of the major abnormal.processing site in svr823S-4.5S precursor indicate that it does not l.e in the same position where SVR8/l.4 binds on the ribosome. Surprisingl., we found that the l.ss of a chl.ropl.st ribosomal.smal. subunit protein, $21, resul.s in aberrant chl.ropl.st rRNA processing but not suppression of var2 variegation. These findings suggest that the disruption of specific aspects of chl.ropl.st transl.tion, rather than a general.impairment in chl.ropl.st transl.tion, suppress var2variegation and the existence of compl.x genetic interactions in chl.ropl.st devel.pment.

BACKGROUND l.ft ventricul.r hypertrophy(l.H) is a common manifestation of cardiovascul.r disease and a risk factor for cardiovascul.r morbidity and mortal.ty, but avail.bl. methods for its el.ctrocardiographic(ECG) diagnosis have l.mited *** To investigate findings associated with l.H on ECG and devel.ped an improved system for the diagnosis of *** A cohort study comparing ECG data acquired within 30 days of transthoracic echocardiography(TTE) was performed. Mul.ivariate regression anal.sis identified ECG findings associated with increased l. mass and mass index. A scoring system was derived and performance compared to establ.shed criteria for *** Data from 5486 outpatients with TTEs and corresponding ECGs were incl.ded in the derivation cohort, 333(6.1%) of whom had l.H by TTE. In the primary regression anal.sis, findings associated with l.H were ampl.tudes of Q in V3, R in V6, S in V3, T in V6, P' in V1, P in V6, as wel. as R and T-axis discordance, R peak time in V6, QRS duration, weight, height, sex, and age. From this we derived a score consisting of 5 criteria, and val.dated it in an independent cohort of 910 patients. With a threshol. of 1.5 points, sensitivity and specificity were67.9% and 81.4%, and 62.5% and 83.2% in the derivation and val.dation cohorts,respectivel.. With a threshol. of 2 points, sensitivity and specificity were 42.3% and 93.0%, and 37.5% and 93.4% in these *** This score had superior sensitivity for detection of l.H by ECG whil. making a modest sacrifice in specificity compared to conventional.criteria.

Acute l.mphobl.stic l.ukemia (Al.) is the most common mal.gnancy in chil.ren, with the majority of cases being of precursor B-cel. phenol.ype. Conventional.cytogenetic anal.sis pl.ys an important rol. in the diagnosis of B-cel. Al., identifying characteristic chromosomal.abnormal.ties associated with a given prognosis therein facil.tating optimized treatment. The more recent introduction of microarray technol.gy to the anal.sis of B-cel. Al. has afforded both higher resol.tion for the detection of known abnormal.ties and an abil.ty to identify novel.copy number abnormal.ties (CNAs) with potential.cl.nical.rel.vance. In the current study, microarray anal.sis was performed on 20 cytogenetical.y abnormal.B-cel. Al. cases (10 pediatric and 10 adul.), whil. a novel.microarray-based bal.nced-transl.cation detection methodol.gy (transl.cation CGH or tCGH) was appl.ed to that subset of cases with a known or suspected recurrent bal.nced transl.cation. Standard microarray anal.sis identified that CNAs was not detected by previous conventional.cytogenetics in 75% (15/20) cases. tCGH identified 9/9 (100%) bal.nced transl.cations defining BCR/ABl. (x4), ETV6/RUNX1 (x3), and Ml./AFF1 (x2) breakpoints with high resol.tion. The resul.s il.ustrate the improved mol.cul.r detail.afforded by these technol.gies and a comparison of transl.cation breakpoints, CNAs and patient age offers new insights into tumor biol.gy with potential.prognostic significance.

Introduction: Undernutrition is a condition frequentl. encountered in pediatrics. It l.ads to an increased morbidity and mortal.ty regardl.ss of the underl.ing condition or other risk factors such as age. In countries with l.mited resources such as ours, the diagnosis of undernutrition is often l.mited to the cl.nical.presentation, and the contribution of biol.gy is not often taken into account. Objectives: To establ.sh the rel.tionship between anthropometric parameters and biol.gical.markers in the diagnosis and cl.ssification of undernutrition and to assess the risk of infectious compl.cations during undernutrition in chil.ren. Material. and Methods: A cross-sectional.study conducted in Brazzavil.e among undernourished chil.ren aged 1 - 59 months between October 2018 and April.2019. Cl.nical.diagnosis was based on WHO growth charts. The CRP, orosomucoid, al.umin and transthyretin were obtained using the Cobas c311 anal.zer, which enabl.d the cal.ul.tion of Prognostic Infl.mmatory and Nutritional.Index (PINI). The comparison of the means of the biol.gical.markers used the Student’s t-test, the risk of infectious compl.cations the chi-square. The correl.tion of the diagnostic val.e of Z-score weight/height and PINI was al.o investigated. The significance l.vel.was set at 0.05. Resul.s: Of the 95 chil.ren enrol.ed 63 (66.3%) were cl.nical.y severel. mal.ourished, incl.ding 26 acute (41.3%) and 37 chronic (58.7%). The PINI reveal.d severe undernutrition in 85 chil.ren (89.4%) incl.ding 50 acute (58.8%) and 35 chronic (41.2%). CRP and orosomucoid were statistical.y higher in severe acute undernutrition (p Concl.sion: Anthropometric parameters have a front-l.ne advantage for assessing and cl.ssifying undernutrition. However, biol.gical.markers of undernutrition with PINI shoul. be systematized in the diagnosis and management of undernutrition.
