Alpha-fetoprotein-targeted reporter gene expression imaging in hepatocellular carcinoma

Hepatocellular carcinoma (HCC) is one of the most common cancers in Eastern Asia, and its incidence is increasing globally. Numerous experimental models have been developed to better our understanding of the pathogenic mechanism of HCC and to evaluate novel therapeutic approaches. Molecular imaging is a convenient and up-to-date biomedical tool that enables the visualization, characterization and quantification of biologic processes in a living subject. Molecular imaging based on reporter gene expression, in particular, can elucidate tumor-specific events or processes by acquiring images of a reporter gene’s expression driven by tumor-specific enhancers/promoters. In this review, we discuss the advantages and disadvantages of various experimental HCC mouse models and we present in vivo images of tumor-specific reporter gene expression driven by an alpha-fetoprotein (AFP) enhancer/promoter system in a mouse model of HCC. The current mouse models of HCC development are established by xenograft, carcinogen induction and genetic engineering, representing the spectrum of tumor-inducing factors and tumor locations. The imaging analysis approach of reporter genes driven by AFP enhancer/promoter is presented for these different HCC mouse models. Such molecular imaging can provide longitudinal information about carcinogenesis and tumor progression. We expect that clinical application of AFP-targeted reporter gene expression imaging systems will be useful for the detection of AFP-expressing HCC tumors and screening of increased/decreased AFP levels due to disease or drug treatment.

Combined silicosis and mixed dust pneumoconiosis with rapid progression: A case report and literature review

BACKGROUND Rapidly progressive pneumoconiosis(RPP) occasionally occurs in coal workers, particularly those with high exposure to silica. Here, we report the case of a 64-year-old male miller with RPP. CASE SUMMARY The patient had a persistent cough for one month and had been clinically diagnosed with pulmonary tuberculosis in 2011. He worked in a stone processing factory from the ages of 20 through 37 and has owned his own mill for the past 25 years. His chest radiograph showed significant increases in the size and number of lung nodules since his last follow-up in 2013. By percutaneous needle lung biopsy, the nodular lesions showed diffuse infiltration of phagocytic macrophages and birefringent crystals by polarizing microscopy. He was finally diagnosed with RPP of mixed dust pneumoconiosis combined with silicosis. CONCLUSION In this case, mixed dust pneumoconiosis with silicosis might be accelerated by persistent exposure to graindust from working in a mill environment.

AIM： To examine whether trans-10,cis-12 CLA （t10C12） or cis-9,trans-11 CLA （c9t11） inhibits heregulin （HRG）-β- stimulated cell growth and HRG-β-ErbB3 signaling in HT-29 cells. METHODS： We cultured HT-29 cells in the absence or presence of the CLA isomers and/or the ErbB3 ligand HRG-β. MTT assay, [^3H]thymidine incorporation, Annexin V staining, RT-PCR, Western blotting, immunoprecipitation, and in vitro kinase assay were performed. RESULTS： HRG-β increased cell growth, but did not prevent DNA t10c12-induced growth inhibition. T10C12 inhibited DNA synthesis and induced apoptosis of HT-29 cells, whereas c9t11 had no effect. T10c12 decreased the levels of ErbB1, ErbB2, and ErbB3 proteins and transcripts in a dose-dependent manner, whereas c9t11 had no effect. Immunoprecipitation/ Western blot studies revealed that t10c12 inhibited HRG- β-stimulated phosphorylation of ErbB3, recruitment of the p85 subunit of phosphoinositide 3-kinase （PI3K） to ErbB3, ErbB3-associated PI3K activities, and phosphorylation of Akt. However, c9t11 had no effect on phospho Aid： levels. Neither t10c12 nor c9t11 had any effect on HRG-β-induced phosphorylation of ERK-1/2. CONCLUSION： These results indicate that the inhibition of HT-29 cell growth by t10c12 may be induced via its modulation of ErbB3 signaling leading to inhibition of Akt activation.

Two Types of Diurnal Variations in Heavy Rainfall during july over Korea

This study examined the characteristics of the diurnal variations of heavy rainfall(≥110 mm in 12 hours)in Korea and the related atmospheric circulation for july from 1980−*** the analysis period,two dominant pattens of diurnal variation of the heavy rainfall emerged:all-day heavy rainfall(AD)and morning only heavy rainfall(MO)*** the AD-type,the heavy rainfall is caused by abundant moisture content in conjunction with active convection in the morning(0000−1200,LST;LST=UTC+9)and the afternoon hours(1200−2400 LST).These systems are related to the enhanced moisture inflow and upward motion induced by the strengthening of the western North Pacific subtropical high and upper-tropospheric *** the MO-type,heavy rainfall occurs mostly in the morning hours;the associated atmospheric patterns are similar to the *** find that the atmospheric pattern related to severe heavy rainfalls in 2020 corresponds to a typical AD-type and resembles the 1991 heavy-rainfall system in its overall synoptic/mesoscale *** present results imply that extremely heavy rainfall episodes in Korea during the 2020 summer may occur again in the future associated with the recurring atmospheric phenomenon related to the heavy rainfall.

Fabrication of TiN/cBN and TiC/diamond coated particles by titanium deposition process

Cubic boron nitride particles coated by titanium nitride (TiN/cBN) as well as diamond particles coated by titanium carbide (TiC/diamond) were prepared by Ti molten salt deposition followed by heat-treatment process. cBN or diamond particles were mixed separately with Ti powders and molten salts (KCl, NaCl and K2TiF6). The mixture was heated at 900 °C under argon atmosphere. The produced particles were heat-treated under hydrogen at 1000 °C. The morphologies and chemical compositions of the produced particles were investigated by scanning electron microscopy (SEM), X-ray diffraction (XRD) and focused ion beam (FIB). The results show that the cBN and the diamond particles are coated by nano-sized Ti layers. By heat-treatment of the Ti/cBN and TiC/diamond coated particles under hydrogen atmosphere, the deposited Ti layers were interacted by the in-situ transformation reaction with the surfaces of cBN and diamond particles and converted to titanium compounds (TiN and TiC), respectively.

Expert Opinions on the Concept of Blood Stasis in China: An Interview Study

Objective: To study various experts’ opinions on the definition and diagnosis of blood stasis in China. Methods: We e-mailed the selected experts to explain the purpose of the study and to invite them to participate and asked them to name a time for the investigator to call them. Eight experts and five organizations were interviewed in the research community investigating blood stasis in China. Results: Six main categories emerged from the interviews:(1) blood stasis concepts;(2) blood stasis-related biomarkers;(3) methods of diagnosing blood stasis;(4) drugs for promoting blood circulation and dissipating stasis;(5) blood stasis-related diseases; and(6) blood stasis-related societies. The consensus among the interviewed experts was that the definition of blood stasis is rather complicated and that there is no gold standard marker for detecting blood stasis. Conclusion: This paper acquired experts’ opinions on the definition and diagnosis of blood stasis in order to establish a modern concept of blood stasis. This paper also developed a diagnostic tool and diagnostic indices for blood stasis and identified biological indices and pathologic mechanisms related to blood stasis, which might be of great reference value in future blood stasis standardization research.

Development of technique to detect and classify small-scale magnetic flux cancellation and rapid blue-shifted excursions

We present a set of tools for detecting small-scale solar magnetic cancellations and the disk counterpart of type II spicules（the so-called Rapid Blueshifted Excursions（RBEs））, using line-of-sight photospheric magnetograms and chromospheric spectroscopic observations, respectively. For tracking magnetic cancellation,we improve the Southwest Automatic Magnetic Identification Suite（SWAMIS） so that it is able to detect certain obscure cancellations that can be easily missed. For detecting RBEs, we use a normalized reference profile to reduce false-positive detections caused by the non-uniform background and seeing condition. Similar to the magnetic feature tracking in SWAMIS, we apply a dual-threshold method to enhance the accuracy of RBE detection. These tools are employed to analyze our coordinated observations using the Interferometric BIdimensional Spectrometer at the Dunn Solar Telescope of the National Solar Observatory and Hinode. We present the statistical properties of magnetic cancellations and RBEs, and explore their correlation using this data set.

Self-expandable metal stents in patients with postoperative delayed gastric emptying after distal gastrectomy

AIM To investigate the efficacy and safety of endoscopic stent insertion in patients with delayed gastric emptying after *** In this study, we prospectively collected data from patients who underwent stent placement for delayed gastric emptying(DGE) after distal gastrectomy between june 2010 and April 2017, at a tertiary referral academic center. Clinical improvement, complications, and consequences after stent insertion were *** Technical success was achieved in all patients(100%). Early symptom improvement was observed in 15 of 20 patients(75%) and clinical success was achieved in all patients. Mean follow-up period was 1178.3 ± 844.1 d and median stent maintenance period was 51 d(range 6-2114 d). During the follow-up period, inserted stents were passed spontaneously per rectum without any complications in 14 of 20 patients(70%). Symptom improvement was maintained after stent placement without the requirement of any additional intervention in 19 of 20 patients(95%).CONCLUSION Endoscopic stent placement provides prompt relief of obstructive symptoms. Thus, it can be considered an effective and safe salvage technique for post-operative DGE.

Although bismuth vanadate(BiVO4)has been promising as photoanode material for photoelectrochemical water splitting,its charge recombination issue by short charge diffusion length has led to various studies about heterostructure *** a hole blocking layer of BiVO4,titanium dioxide(TiO_(2)) has been considered unsuitable because of its relatively positive valence band edge and low electrical ***,a crystal facet engineering of TiO_(2) nanostructures is proposed to control band structures for the hole blocking layer of BiVO4 *** design two types of TiO_(2) nanostructures,which are nanorods(NRs)and nanoflowers(NFs)with different(001)and(110)crystal facets,respectively,and fabricate BiVO4/TiO_(2) heterostructure *** BiVO4/TiO_(2) NFs showed 4.8 times higher photocurrent density than the BiVO4/TiO_(2) *** decay time analysis and time-resolved photoluminescence reveal the enhancement is attributed to the reduced charge recombination,which is originated from the formation of type II band alignment between BiVO4 nanodots and TiO_(2) *** work provides not only new insights into the interplay between crystal facets and band structures but also important steps for the design of highly efficient photoelectrodes.
