Dihydromyricetin Alleviates H9C2 Cell Apoptosis and Autophagy by Regulating CircHIPK3 Expression and PI3K/AKT/mTOR Pathway

Objective: To investigate the effect and potential mechanism of dihydromyricetin(Dmy) on H9C2 cell proliferation, apoptosis, and autophagy. Methods: H9C2 cells were randomly divided into 7 groups, namely control, model, EV(empty p CDH-CMV-MCS-EF1-Cop GFP-T2A-Puro vector), IV(circHIPK3 interference), Dmy(50 μmol/L), Dmy+IV, and Dmy+EV groups. Cell proliferation and apoptosis were detected by cell counting kit-8 assay and flow cytometry, respectivley. Western blot was used to evaluate the levels of light chain 3 Ⅱ/Ⅰ(LC3Ⅱ/Ⅰ), phospho-phosphoinositide 3-kinase(p-PI3K), protein kinase B(p-AKT), and phospho-mammalian target of rapamycin(p-mTOR). The level of circHIPK3 was determined using reverse transcriptase polymerase chain reaction. Electron microscopy was used to observe autophagosomes in H9C2 cells. Results: Compared to H9C2 cells, the expression of circHIPK in H9C2 hypoxia model cells increased significantly(P<0.05). Compared to the control group, the cell apoptosis and autophagosomes increased, cell proliferation rate decreased significantly, and the expression of LC3Ⅱ/Ⅰ significantly increased(all P<0.05). Compared to the model group, the rate of apoptosis and autophagosomes in IV, Dmy, and Dmy+IV group decreased, the cell proliferation rate increased, and the expression of LC3Ⅱ/Ⅰ decreased significantly(all P<0.05). Compared to the control group, the expressions of p-PI3K, p-AKT, and p-mTOR in the model group significantly reduced(P<0.05), whereas after treatment with Dmy and sh-circHIPK3, the above situation was reversed(P<0.05). Conclusion: Dmy plays a protective role in H9C2 cells by inhibiting circHIPK expression and cell apoptosis and autophagy, and the mechanism may be related to PI3K/AKT/mTOR pathway.

A 3D nano-displacement measurement method,where the difference in phase between the beams in a four-beam laser interference is changed,is *** results demonstrate that the variation of phase difference causes the deviation of the interference pattern in the laser interference *** on this theory,we design and build a four-beam laser interference *** corner cube prism in the optical path is shifted,and the phase of the beam is changed by applying different voltages to a piezoelectric *** phase difference is obtained by analyzing the lattice pattern with subpixel precision,and then the displacement is determined by correlation *** experimental measurement results are consistent with the theoretical analysis,thereby verifying the feasibility of this measurement method.

When debugging encounters artificial intelligence: state of the art and open challenges

Both software debugging and artificial intelligence techniques are hot topics in the current field of software engineering. Debugging techniques, which comprise fault localization and program repair, are an important part of the software development lifecycle for ensuring the quality of software systems. As the scale and complexity of software systems grow, developers intend to improve the effectiveness and efficiency of software debugging via artificial intelligence(artificial intelligence for software debugging, AI4SD). On the other hand, many artificial intelligence models are being integrated into safety-critical areas such as autonomous driving, image recognition, and audio processing, where software debugging is highly necessary and urgent(software debugging for artificial intelligence, SD4AI). An AI-enhanced debugging technique could assist in debugging AI systems more effectively, and a more robust and reliable AI approach could further guarantee and support debugging techniques. Therefore, it is important to take AI4SD and SD4AI into consideration comprehensively. In this paper, we want to show readers the path, the trend, and the potential that these two directions interact with each other. We select and review a total of 165 papers in AI4SD and SD4AI for answering three research questions, and further analyze opportunities and challenges as well as suggest future directions of this cross-cutting area.

Chain Dynamics Heterogeneity in Plasticized Poly(vinyl butyral)(PVB) as Elucidated by Solid-State NMR

The chain dynamics heterogeneity of the poly(vinyl butyral)(PVB) plasticized by triethylene glycol bis(2-ethylhexa noate)(TEG-EH) was investigated by various solid-state NMR *** plasticized PVB shows two domains in distinct molecular dynamics differences,namely,rigid and soft domains,where the latter is the plasticizer-rich *** time domain low field NMR was first used to investigate the dynamics heterogeneity of the plasticized PVB,and the results show the decreasing activated energy of components in the soft domain of plasticized PVB(E_a=20.2 kJ/mol) as compared with that of the pristine one(E_a=24.3 kJ/mol).Detailed dynamics heterogeneity was obtained by high-field NMR with site-specific *** quadrupole-echo ~2H-NMR was adopted to elucidate the dynamics heterogeneity of the vinyl alcohol(VA) units,where only the hydroxyl group of VA is *** ~1H-^(13)C WISE NMR spectra show that there is not much difference in the mobility of the VB unit in PVB with and without plasticizer,whereas the glass transition temperature differed by approximately 53℃.This is further supported by Torchia's T_1 relaxation *** origin of such an unusual phenomenon is attributed to the critical role of the remaining VA(~22%) in the soft domain,where the VA units locally aggregate through hydrogen ***,the existence of a mobility gradient in the VB unit has been ***,the mobility difference for VB with different stereo-geometry(meso or racemic conformation) is observed for the first *** indicates the importance of modulating the ratio of meso over racemic VB for controlling the macroscopic perfo rmance of PVB.

Effect of the interval between two shocks on ejecta formation from the grooved aluminum surface

This work focuses on the effect of the interval between two shocks on the ejecta formation from the grooved aluminum(Al_(1100))surface by using smoothed particle hydrodynamics numerical *** unsupported shocks are obtained by the plate-impact between sample and two flyers at interval,with a peak pressure of approximately 30 GPa for each *** the shock interval varies from 2.11 to 7.67 times the groove depth,the bubble velocity reduces to a constant,and the micro jetting factor R_(J) from spike to bubble exhibits a non-monotonic change that decreases initially and then *** a shock interval of 3.6 times the groove depth,micro jetting factor R_(J) from spike to bubble reaches its minimum value of approximately ***,the micro jetting factor R_(F) from spike to free surface decreases linearly at first,and stabilizes around 0.25 once the shock interval surpasses 4.18 times the groove *** the shock interval is less than 4.18 times the groove depth,the unloading wave generated by the breakout of the first shock wave is superimpose with the unloading part of the second shock wave to form a large tensile area.

Mechanical performance of 22SiMn2TiB steel welded with lowtransformation- temperature filler wire and stainless steel filler wire

TX-80 low-transformation-temperature(LTT)welding wire was used to replace the traditional ER 307Si welding wire to realize the connection of 22SiMn2TiB armor steel in manual overlay *** previously existing issues,such as welding cracks,large welding deformation,and severe welding residual stress,were solved to ensure good strength and ductility *** particular,with the same welding conditions,TX-80 LTT wire eliminates welding *** reduces the welding deformation no matter the base pretreatment of pre-setting angle or no pre-setting *** comparison,it was found that the microstructure at the TX-80 weld is mainly composed of martensite and a small amount of retained *** contrast,the microstructure of the ER 307Si weld consists of a large amount of austenite and a small amount of skeleton-like *** variation trend of residual stress and microhardness from the weld to the base were investigated and compared with the mechanical properties of base *** TX-80 and the ER 307Si tensile samples elongation is 6.76%and 6.01%,while the ultimate tensile strengths are 877 and 667 MPa,*** average impact toughness at room temperature of the ER 307Si weld is 143.9 J/cm^(2),much higher than that of the TX-80 weld,which is only 36.7 J/cm^(2).The relationship between impact and tensile properties with microstructure species and distribution was *** addition,the fracture surface of the tensile and the impact samples was observed and *** dimples,fewer pores,larger radiation zone,and shear lips of TX-80 samples indicate better tensile ductility and worse impact toughness than those of ER 307Si weld.

Realizing high initial Coulombic efficiency in manganese-based layered oxide cathodes for sodium-ion batteries via P2/O'3 biphasic structure optimization

Mn-based layered oxides are among the most promising cathode materials for sodium-ion batteries owing to the advantages of abundance,environmenta friendliness,low cost and high specific capacity.P2 and O'3 are two representative structures of Mn-based layered ***,the P2 structure containing insufficien Na generally exhibits low initial charge capacity,while O'3structure with sufficient Na delivers high initial charge capacity but poor cycle *** study prepared a multitude of Na_(x)MnO_(2)(x=0.7,0.8,0.9)cathode materials with varying P2/O'3 ratios and further investigated their electrochemical *** optimized Na_(0.8)MnO_(2) comprising 69.9 wt%O'3 and 30.1 wt%P2 phase,exhibited relatively balanced specific capacity,Coulombic efficiency and cycle ***,it achieved a high specific capacity of 128.9 mAh·g^(-1) with an initia Coulombic efficiency of 98.2%in half-cell configuration The Na_(0.8)MnO_(2)//hard carbon full cell also achieved a high specific capacity of 126.7 mAh·g^(-1) with an initia Coulombic efficiency of 98.9%.Moreover,the capacity fading mechanism was revealed by combining in-situ and ex-situ X-ray *** findings of this study provide theoretical guidance for further modification design of Mnbased layered cathodes.

Chronic administration of triclosan leads to liver fibrosis through hepcidin-ferroportin axis-mediated iron overload

Triclosan(TCS)has been manufactured as an antibacterial compound for half a ***,it is widely used in various personal care products;however,its potential adverse effects raise a lot of ***,we create a long-term oral administration mouse model and identify the corresponding hepatotoxicity of *** discover that daily intragastric administration of 10 mg/kg TCS to mice for 12 weeks results in severe hepatic *** study displays that hepatic iron increased 18%,23%and 29%upon oral TCS treatment for4,8 and 12 weeks,*** by hepatic iron variation,splenic and duodenal iron are increased,which indicates systemic iron *** only excessive iron accumulated in the liver,abnormal hepatic malondialdehyde,prostaglandin synthase 2 and glutathione peroxidase 4 are pointed to *** study uncovers that hepcidin expression increases 7%,10%,4%in serum and 2.4-,4.8-,and 2.3-fold on transcriptional levels upon TCS exposure for 4,8 and 12 weeks,*** together,the mice in the TCS-treated group show disordered systemic iron homeostasis via the upregulated hepatic hepcidin-ferroportin ***,both hepatic iron overload(systemic level)and hepatocyte ferroptosis(cellular level)are accused of TCS-induced liver ***■,an iron scavenger,significantly ameliorates TCS-induced liver *** summary,this study confirms the impact of TCS on liver fibrosis;a critical signal pathway is also *** significance of the current study is to prompt us to reevaluate the“pros and cons”of TCS applications.
