Matched pair analysis of the effect of longer hypothermic machine perfusion time on kidney transplant outcomes

BACkGROUND Hypothermic machine perfusion(HMP)has demonstrated benefits in terms of early kidney transplant function compared to static cold *** longer preservation times have shown detrimental effects,a previous paired study indicated that longer pump times(the second kidney in a pair)might lead to improved *** To revisit the prior paired study's somewhat unexpected results by reviewing our program's *** A total of 61 pairs of transplant recipients who received kidneys from the same donor(2012-2021)were *** were divided into two groups depending on whether they were transplanted first(k1)or second(k2).Therefore,the patients in each pair had identical donor characteristics,except for time on the *** analyses included kaplan-Meyer analysis and paired tests,including McNemar's test,student's paired t-test,or Wilcoxon's test,as *** The two groups of recipients had similar demographics(age,body mass index,diabetes,time on dialysis,sensit-ization and retransplants).Cold ischemic times for k1 and k2 were 8.9(95%CI:7.9,9.8)and 14.7 hours(13.7,15.8)(Pk2 had a higher rate of freedom from biopsy-proven acute rejection at 1 year(P=0.015).Delayed graft function was less common in k2,12/61(20%)than in k1,20/61(33%)(P=0.046).Finally,k2 showed a higher graft survival than k1(P=0.023).CONCLUSION Our results agree with a previous study that suggested possible advantages to longer pump *** studies should encourage further research into HMP's potential anti-inflammatory effect.

It is generally accepted that jasmonate-ZIM domain(jaZ)repressors act to mediate jasmonate(ja)signaling via CORONATINE-INSENSITIVE1(COI1)-mediated ***,we report a cryptic signaling cascade where a jaZ repressor,FvjaZ12,mediates multiple signaling inputs via phosphorylation-modulated subcellular translocation rather than the COI1-mediated degradation mechanism in strawberry(Fragaria vesca).FvjaZ12 acts to regulate flavor metabolism and defense response,and was found to be the target of Fv MPk6,a mitogen-activated protein kinase that is capable of responding to multiple signal ***6 phosphorylates FvjaZ12 at the amino acid residues S179 and T183 adjacent to the PY residues,thereby attenuating its nuclear accumulation and relieving its repression for FvMYC2,which acts to control the expression of lipoxygenase 3(FvLOX3),an important gene involved in ja biosynthesis and a diverse array of cellular *** data reveal a previously unreported mechanism for ja signaling and decipher a signaling cascade that links multiple signaling inputs with fruit trait development.

OBJECTIVE:To investigate the efficacy of Fig fruit powder and olive on hepatic,renal and splenic injury induced by 2-nitropropane(2-NP)in mice,especially if they were used in ***:A total of 40 adult BALB/c male mice weighting 25-30 g/*** were categorized into five groups(8 each).Group 1 as negative *** 2 as positive control group intraperitoneally injected with 2-NP(100 mg/kg b.w.)3 times/weekly for eight *** 3 injected with 2-NP and were orally supplemented with Fig(300 mg/kg).Group 4 injected with 2-NP and were orally supplemented with olive(100 mg/kg).Group 5 injected with 2-NP and were orally supplemented with mixture of Fig and olive(3∶1 respectively).RESULTS:Histopathological observation of liver in mice treated with 2-NP showed cellular degeneration,pyknosis,and congestion of the portal *** kidney there were disorganization of the cortical tissues,cellular necrosis and plenty of inflammatory lymphocytic *** elevations in liver function parameters(alanine aminotransferase and aspartate aminotransferase),m RNA expression levels of tumor necrosis factor-α,nicotinamide adenine dinucleotide phosphate oxidase and cyclooxygenase were detected as anti-inflammatory markers and 5-lipoxygenase,interleukin-1βand interleukin-6 as inflammatory biomarkers for liver and spleen,also significant elevations was detected in lipid peroxidation *** levels of antioxidants,glutathione,glutathione peroxidase,catalase and superoxide dismutase were significantly ***:our findings indicated that Fig fruit powder and olive protected against hepatic,renal and splenic injury induced with 2-NP in mice,especially if they were used in combination.

Genome-wide markers for seed yield and disease resistance in perennial ryegrass

The market success of perennial ryegrass(Lolium perenne L.) cultivars depends on sufficient seed production, as they are propagated by seed. However, breeding for high quality forage production reduces seed yield, and breaking the negative correlation would help to overcome the problem. The foliar disease crown rust is another factor affecting reproductive capacity and thereby seed yield. We evaluated seed yield-related traits and resistance to crown rust in a collection of commercial cultivars and ecotypes of perennial ryegrass and identified genome-wide markers associated with the traits. The study revealed high variation between the ecotype and cultivar groups as well as between years. A genome-wide association study identified 17 DNA single-nucleotide polymorphisms(SNPs) of which eight were associated with crown rust and nine with flag-leaf length. The SNP markers were located within or near predicted genes functioning in defense against pathogens. The identified genes are strong candidates for a further in-depth functional study to continue unravel determination of leaf architecture and crown rust resistance in perennial ryegrass.

Background Pediatric cancer survivors are at increased risk of muscle weakness and low areal bone mineral density(aBMD).However,the prevalence of muscle strength deficits is not well documented,and the associations of muscle strength with aBMD are unknown in this ***,this study aimed to investigate the prevalence of upper-and lower-body muscle strength deficits and to examine the associations of upper-and lower-body muscle strength with age-,sex,and race-specific aBMD Z-scores at the total body,total hip,femoral neck.a.d lumbar *** This cross-sectional study included 116 pediatric cancer survivors(12.1±3.3 years old,mean±SD;42.2%female).Upper-and lower-body muscle strength were assessed by handgrip and standing long jump test,***‑energy X‑ray absorptiometry was used to measure aBMD(g/cm2).Associations between muscle strength and aBMD were evaluated in multivariable linear regression *** regression was used to evaluate the contribution of muscle strength(1-decile lower)to the odds of having low aBMD(Z-score≤1.0).All analyses were adjusted for time from treatment completion,radiotherapy exposure,and body mass *** More than one-half of survivors were within the 2 lowest deciles for upper-(56.9%)and lower-body muscle strength(60.0%)in comparison to age-and sex-specific reference *** strength deficits were associated with lower aBMD Z-scores at all sites(B=0.133–0.258,p=0.001–0.032).Each 1-decile lower in upper-body muscle strength was associated with 30%–95%higher odds of having low aBMD Z-scores at all *** 1-decile lower in lower-body muscle strength was associated with 35%–70%higher odds of having low aBMD Z-scores at total body,total hip,and femoral *** Muscle strength deficits are prevalent in young pediatric cancer survivors,and such deficits are associated with lower aBMD Z-scores at all *** results suggest that interventions designed to improve muscle strength in t

High-grade dysplasia (HGD) and intramucosal carcinoma (IMC) in the setting of Barrett’s esophagus have traditionally been treated with esophagectomy. However, with the advent of endoscopic mucosal resection and endoscopic ablative therapies, endoscopic therapy at centers with expertise is now an established treatment of Barrett’s-esophagus-related neoplasia, including HGD and IMC. Esophagectomy is today reserved for more selected cases with submucosal invasion, evidence for lymph node metastasis, or unsuccessful endoscopic therapy.

以采集的121份野生枸杞(Lycium barbarum L.)种质为材料,用9对SSR引物进行扩增,采用GeneALEX、Power Marker软件进行多态性分析,利用NTSYS、STRUC-TURE软件进行遗传多样性分析、聚类分析和主成分分析。结果表明,9对SSR引物共检测到108个等位基因,等位基因数(Na)、有效等位基因数(Ne)、观测杂合度(Ho)、期望杂合度(He)、多态信息含量(PIC)、Shannon’s信息指数分别为12个、4.620个、0.678、0.651、0.629、1.568;聚类结果显示121份野生枸杞种质可划分为5个亚群,群体遗传结构表明当k=5时,121个种质划分为5个亚群,来自宁夏的NXGY-02、NXGY-032个种质与来自内蒙古的6个种质之间的遗传距离最远,亲缘关系较远,来自青海省、新疆、甘肃省及内蒙古的其余野生枸杞种质之间存在广泛的基因交流现象;宁夏、内蒙古的野生枸杞种质具有独立起源进化的可能,而新疆、甘肃省、青海省的种质与宁夏种质亲缘关系较近,很可能属于同一起源。

The resveratrol-enriched transgenic rice line Ik.a.526(IS526),first developed by the Rural Development Administration of korea using genetic engineering techniques,shows beneficial health effects in mitigating metabolic syndrome and ***,the effects of IS526 on the differentiation of chondrocytes and the underlying mechanism have not been investigated in *** this study,the effects and cellular regulatory mechanisms of IS526 on rabbit articular chondrocytes were *** IS526 callus extract treatment,the expression levels of differentiation-related proteins were detected via western blotting,Alcian blue staining and immune-luorescence ***526 decreased the type Ⅱ collagen and proteoglycan levels in dose-and time-dependent *** further analyzed the effects of IS526 on skeleton genesis in zebrafish larvae using Alcian blue staining,which showed a reduction in cartilage formation along with increased production of matrix metalloproteinase(MMP)-***526 also increased the phosphorylation of ERk1/2 and p38 kinase but inhibited the phosphorylation of *** inhibition of MMP-13 blocked the IS526-induced decrease in type Ⅱ collagen *** of p38 kinase or PI-3k.a.t with SB203580 and LY294002 enhanced the suppression of type Ⅱ collagen,but the blockage of ERk-1/2 by PD98059 rescued IS526-induced *** results suggested that IS526 regulates type Ⅱ collagen and MMP-13 expression via the ERk1/2 and PI-3k.a.t pathways in rabbit articular chondrocytes.

An accelerated k-means clustering algorithm using selection and erasure rules

The k-means method is a well-known clustering algorithm with an extensive range of applications,such as biological classification,disease analysis,data mining,and image ***,the plain k-means method is not fast when the number of clusters or the number of data points becomes large.A modified k-means algorithm was presented by Fahim et al.(2006).The modified algorithm produced clusters whose mean square error was very similar to that of the plain k-means,but the execution time was *** this study,we try to further increase its *** are two rules in our method:a selection rule,used to acquire a good candidate as the initial center to be checked,and an erasure rule,used to delete one or many unqualified centers each time a specified condition is *** clustering results are identical to those of Fahim et al.(2006).However,our method further cuts computation time when the number of clusters *** mathematical reasoning used in our design is included.
