Unraveling the drug distribution in brain enabled by MALDI MS imaging with laser-assisted chemical transfer

Accurate localization of central nervous system(CNS)drug distribution in the brain is quite challenging to matrix-assisted laser desorption/ionization(MALDI)mass spectrometry imaging(MSI),owing to the ionization competition/suppression of highly abundant endogenous biomolecules and MALDI ***,we developed a highly efficient sample preparation technique,laser-assisted chemical transfer(LACT),to enhance the detection sensitivity of CNS drugs in brain tissues.A focused diode laser source transilluminated the tissue slide coated with α-cyano-4-hydroxycinnamic acid,an optimal matrix to highly absorb the laser radiation at 405 nm,and a very thin-layer chemical film mainly containing drug molecule was transferred to the acceptor glass ***,MALDI MSI was performed on the chemical film without additional sample *** major advantage of LACT is to minimize ionization competition/suppression from the tissue itself by removing abundant endogenous lipid and protein *** superior performance of LACT led to the successful visualization of regional distribution patterns of 16 CNS drugs in the mouse ***,the dynamic spatial changes of risperidone and its metabolite were visualized over a 24-h ***,the brain-to-plasma(B/P)ratio could be obtained according to MALDI MSI results,providing an alternative means to assess brain penetration in drug discovery.

With good mobility and flexibility,mobile manipulators have shown broad applications in construction *** position(BP)planning,which refers to the robot autonomously determining its working station in the environment,is an important technique for mobile manipulators when performing the construction assembly task,especially in a large-scale construction ***,the BP planning process is tedious and time-consuming for a human worker to carry ***,to improve the efficiency of construction assembly tasks,a novel BP planning method is proposed in this paper,which can lead to appropriate BPs and minimize the number of BPs at the same ***,the feasible BP regions are generated based on the grid division and the variable workspace of the mobile ***,the positioning uncertainties of the mobile manipulator are considered in calculating the preferred BP areas using ***,a set coverage optimization model is established to obtain the minimum number of BPs using an optimization algorithm according to the greedy *** simulated experiment based on a 9-degree of free(DoF)mobile manipulator has been *** results illustrated that the time for BP planning was significantly reduced and the number of BPs was reduced by 63.41%compared to existing manual planning,which demonstrated the effectiveness of the proposed method.

In order to study deposited sediment settlement and consolidation mechanisms, sediment settlement experiments were conducted using a settlement column. Based on the experimental results, sediment settlement stage definition, excessive pore pressure （EPP） dissipation, and consolidation constitutive equations are discussed. Three stages, including the free settlement, hindered settlement, and self-weight consolidation settlement stages, are defined. The results of this study show that sediment settlement is mainly affected by the initial sediment concentration and initial settlement height, and the interface settlement rate is linearly attenuated with time on bilogarithmic scales during the hindered settlement and self-weight consolidation settlement stages. Moreover, the deposited sediment layer in the self-weight consolidation settlement stage experiences large strains, and the settlement amount in this stage is about 32% to 59% of the initial height of deposited sediment. EPP is nonlinearly distributed in the settlement direction, and consolidation settlement is faster than EPP dissipation in the self-weight consolidation settlement stage. Consolidation constitutive equations for the hydraulic conductivity and effective stress, applicable to large-strain consolidation calculation, were also determined and fitted in the power function form.

Through changing the axial load on backfilling material compaction test to reflect different overlying strata pressure on backfilling material, the stress-strain relations in the compaction process of backfilling material under the geological condition can be obtained. Based on the characteristic of overlying strata movement in backfill mining, a model of roof thin plate is established. By introducing the stress-strain relation in compaction process into the model and using RIZT method to analyze the bending deformation of roof, the bending deflection and stress distribution can be obtained. The results show that the maximum roof subsidence and maximum tensile stress occurring at the center are 255 mm and5 MPa, respectively. Tensile fracture of roof under the geological condition of Dongping Mine did not occur. The dynamic measurement results of roof in Dongping Mine verify the theoretical result from the aforementioned model, thereby suggesting the roof mechanical model is reliable. The roof thin plate model based on the compaction characteristic of backfilling material in this study is of importance to research on backfill mining theories and application of backfilling material characteristics.

Studies in the northern South China Sea(SCS)basement remain important for understanding the evolution of the Southeast Asian continental *** to a thick cover of sediments and scarce borehole penetration,little is known about the age and tectonic affinity of this *** this study,an integrated study of zircon U-Pb geochronology,Hf isotopes,and whole-rock major and trace elements on seven basement granitoids from seven boreholes of Qiongdongnan Basin has been carried *** zircon U-Pb results for these granitoids present middle-late Permian((270.0±1.2)Ma;(253±3.4)Ma),middle to late Triassic((246.2±3.4)Ma;(239.3±0.96)Ma;(237.9±0.99)Ma;(228.9±1.0)Ma)and Late Cretaceous ages((120.6±0.6)Ma).New data from this study,in combination with the previous dataset,indicates that granitoid ages in northern SCS basement vary from 270 Ma to 70.5 Ma,with three age groups of 270–196 Ma,162–142 Ma,and 137–71 Ma,*** for the late Paleozoic-Mesozoic rocks in the basement of the northern SCS,a few old zircon grains with the age of(2708.1±17)Ma to(2166.6±19)Ma provide clues to the existence of the pre-Proterozoic *** geochemical signatures indicate that the middle Permian-early Cretaceous granitoids from the Qiongdongnan Basin are I-type granites formed in a volcanic arc environment,which were probably related to the subduction of the Paleo-Pacific Plate.

This article is based on the omnidirectional mobile manipulator with mecanum wheels built at Shanghai University. The article aims to find and analyze the parameters of kinematic equation of the omnidirectional system which affects its motion accuracy. The method of solving the parameter errors involves three phases. The first step is equation operation to achieve the equation of relative errors. The second step is to obtain the displacement errors of the system via experiment and combine the error results with kinematic equation deduction to solve the geometric parameter errors in two methods. The third step is to verify its validity via comparing experiments. We can then revise its kinematics equation afterwards.

Overpressure is a key factor for oil and gas charging in tight reservoirs,but it is still a challenge to evaluate the overpressure evolution and its control on oil and gas *** Xujiahe Formation in the northeastern Sichuan Basin as an example,this paper presented a method for evaluating overpressure and its effect on natural gas charging in tight sandstone in compressional *** abnormally high pressure and its causes were analyzed by measured data and logging *** calculation and PVT simulation were used to investigate the amounts of overpressure resulted from hydrocarbon generation and tecto nic compression,*** the source rock-reservoir pressu re differences were calculated and the characteristics of natural gas charging during the natural gas charging periods were *** was revealed that hydrocarbon generation and tectonic compression were the main causes of the *** overpressure of both source rocks and reservoir exhibited a gradually increasing trend from Middle Jurassic to Early Cretaceous(J2-K1),then decreased since Later Cretaceous(K2),and some of that preserved to *** contributions of the hydrocarbon generation and tectonic compression to overpressure were different in different *** residual pressure difference between the source rocks and the reservoir is the major driving force for tight sandstone gas *** main hydrocarbon generating area of the source rocks and the area of high driving force were major natural gas enrichment areas,and the driving force determined the natural gas charging space in the pore throat system of the *** research helps evaluate the overpressure and pressure difference between source rocks and reservoir in compressed basin,as well as investigate the effective pore throat space of tight gas charging by the driver of overpressure.
