Rheumatoid arthritis(RA)is a chronic autoimmune disorder marked by persistent synovial inflammation and joint degradation,posing challenges in the development of effective ***,an alkaloid found in lotus leaf,has shown promising anti-inflammatory and anti-tumor effects,yet its efficacy in RA treatment remains *** study investigated the antiproliferative effects of nuciferine on the MH7A cell line,a human RA-derived fibroblast-like synoviocyte,revealing its ability to inhibit cell proliferation,promote apoptosis,induce apoptosis,and cause G1/S phase ***,nuciferine significantly reduced the migration and invasion capabilities of MH7A *** therapeutic potential of nuciferine was further evaluated in a collagen-induced arthritis(CIA)rat model,where it markedly alleviated joint swelling,synovial hyperplasia,cartilage injury,and inflammatory *** also improved collagen-induced bone erosion,decreased pro-inflammatory cytokines and serum immunoglobulins(IgG,IgG1,IgG2a),and restored the balance between T helper(Th)17 and regulatory T cells in the spleen of CIA *** results indicate that nuciferine may offer therapeutic advantages for RA by decreasing the proliferation and invasiveness of FLS cells and correcting the Th17/Treg cell imbalance in CIA rats.

Two-dimensional hafnium-based semiconductors and their heterostructures with native oxides have been shown unique physical properties and potential electronic and optoelectronic ***,the scalable synthesis methods for ultrathin layered hafnium-based semiconductor laterally epitaxy growth and its heterostructure are still restricted,also for the understanding of its formation ***,we report the stable sublimation of alkali halide vapor assisted synthesis strategy for high-quality 2D HfSe_(2) nanosheets via chemical vapor ***-crystalline ultrathin 2D HfSe_(2) nanosheets were systematically grown by tuning the growth parameters,reaching the lateral size of 6‒40μm and the thickness down to 4.5 *** scalable amorphous HfO_(2)and HfSe_(2)heterostructures were achieved by the controllable oxidation,which benefited from the approximate zero Gibbs free energy of unstable 2D HfSe_(2) *** crystal structure,elemental,and time dependent Raman characterization were carried out to understand surface precipitated Se atoms and the formation of amorphous Hf−O bonds,confirming the slow surface oxidation and lattice incorporation of oxygen *** relatively smooth surface roughness and electrical potential change of HfO_(2)−HfSe_(2) heterostructures indicate the excellent interface quality,which helps obtain the high performance memristor with high on/off ratio of 105 and long retention period over 9000 *** work introduces a new vapor catalysts strategy for the synthesis of lateral 2D HfSe_(2) nanosheets,also providing the scalable oxidation of the Hf-based heterostructures for 2D electronic devices.

Thyroid-stimulating hormone(TSH)is important for the thyroid gland,development,growth,and *** in TSH production or the thyrotrope cells within the pituitary gland cause congenital hypothyroidism(CH),resulting in growth retardation and neurocognitive *** human TSH is known to display rhythmicity,the molecular mechanisms underlying the circadian regulation of TSH and the effects of TSH-thyroid hormone(TH)signaling on the circadian clock remain *** we show that TSH,thyroxine(T4),triiodothyronine(T3),and tshba display rhythmicity in both larval and adult zebrafish and tshba is regulated directly by the circadian clock via both E′-box and *** tshba^(−/−)mutants manifest congenital hypothyroidism,with the characteristics of low levels of T_(4)and T_(3)and growth *** or overexpression of tshba alters the rhythmicity of locomotor activities and expression of core circadian clock genes and hypothalamic-pituitary-thyroid(HPT)axis-related ***,TSH-TH signaling regulates clock2/npas2 via the thyroid response element(TRE)in its promoter,and transcriptome analysis reveals extensive functions of Tshba in ***,our results demonstrate that zebrafish tshba is a direct target of the circadian clock and in turn plays critical roles in circadian regulation along with other functions.

While China has become the largest online market in the world with approximately 1 billion internet users,Baidu runs the world's largest Chinese search engine serving more than hundreds of millions of daily active users and responding to billions of queries per *** handle the diverse query requests from users at the web-scale,Baidu has made tremendous efforts in understanding users'queries,retrieving relevant content from a pool of trillions of webpages,and ranking the most relevant webpages on the top of the *** the components used in Baidu search,learning to rank(LTR)plays a critical role and we need to timely label an extremely large number of queries together with relevant webpages to train and update the online LTR *** reduce the costs and time con-sumption of query/webpage labelling,we study the problem of active learning to rank(active LTR)that selects unlabeled queries for an-notation and training in this ***,we first investigate the criterion-Ranking entropy(RE)characterizing the entropy of relevant webpages under a query produced by a sequence of online LTR models updated by different checkpoints,using a query-by-com-mittee(QBC)***,we explore a new criterion namely prediction variances(PV)that measures the variance of prediction res-ults for all relevant webpages under a *** empirical studies find that RE may favor low-frequency queries from the pool for la-belling while PV prioritizes high-frequency queries ***,we combine these two complementary criteria as the sample selection strategies for active *** experiments with comparisons to baseline algorithms show that the proposed approach could train LTR models to achieve higher discounted cumulative gain(i.e.,the relative improvement DCG4=1.38%)with the same budgeted labellingefforts.

High-temperature stress threatens the growth and yield of crops. Basic helix-loop-helix(bHLH) transcription factors(TFs) have been shown to play important roles in regulating high-temperature resistance in plants. However, the bHLH TFs responsible for high-temperature tolerance in cucumbers have not been identified. We used transcriptome profiling to screen the high temperature-responsive candidate bHLH TFs in cucumber. Here, we found that the expression of 75 CsbHLH genes was altered under high-temperature stress. The expression of the CsSPT gene was induced by high temperatures in TT(Thermotolerant) cucumber plants. However, the Csspt mutant plants obtained by the CRISPR-Cas9 system showed severe thermosensitive symptoms, including wilted leaves with brown margins and reduced root density and cell *** Csspt mutant plants also exhibited elevated H_(2)O_(2) levels and down-regulated photosystem-related genes under normal ***, there were high relative electrolytic leakage(REC), malondialdehyde(MDA), glutathione(GSH), and superoxide radical(O_(2)^(·-)) levels in the Csspt mutant plants, with decreased Proline content after the high-temperature treatment. Transcriptome analysis showed that the photosystem and chloroplast activities in Csspt mutant plants were extremely disrupted by the high-temperature stress compared with wildtype(WT) plants. Moreover, the plant hormone signal transduction, as well as MAPK and calcium signaling pathways were activated in Csspt mutant plants under high-temperature stress. The HSF and HSP family genes shared the same upregulated expression patterns in Csspt and WT plants under high-temperature conditions. However, most bHLH, NAC, and bZIP family genes were significantly down-regulated by heat in Csspt mutant plants. Advancements in power electronics necessitate dielectric polymer films capable of operating at high temperatures and possessing high energy *** significant strides have been achieved by integrating inorganic fillers into high-temperature polymer matrices,the inherently low dielectric constants of these matrices have tempered the magnitude of *** this work,we report an innovative nanocomposite based on sulfonylated polyimide(SPI),distinguished by the incorporation of sulfonyl groups within the SPI backbone and the inclusion of wide bandgap hafnium dioxide(HfO_(2))*** nanocomposite has demonstrated notable enhancements in thermal stability,dielectric properties,and capacitive performance at elevated *** simulations at both molecular and mesoscopic levels have elucidated the mechanisms behind these improvements,which could be attributed to confined segmental motion,an optimized electronic band structure,and a diminished incidence of dielectric breakdown ascribed to the presence of sulfonyl ***,the SPI-HfO_(2)nanocomposite demonstrates a high charge-discharge efficiency of 95.7%at an elevated temperature of 150℃and an applied electric field of 200 MV/***,it achieves a maximum discharged energy density of 2.71 J/cm^(3),signalling its substantial potential for energy storage applications under extreme conditions.

Sulfide-based all-solid-state lithium metal batteries(ASSLMBs)have received extensive attention due to their high energy density and high safety,while the poor interface stability between sulfide electrolyte and lithium metal anode limits their ***,a hybrid SEI(LICl/Li F/Li Zn)was constructed at the interface between Li_(5.5)PS_(4.5)Cl_(1.5)sulfide electrolyte and lithium *** Li Cl and Li F interface phases with high interface energy effectively induce the uniform deposition of Li^(+)and reduce the overpotential of Li^(+)deposition,while the Li Zn alloy interface phase accelerates the diffusion of lithium *** synergistic effect of the above functional interface phases inhibits the growth of lithium dendrites and stabilizes the interface between the sulfide electrolyte and lithium *** hybrid SEI strategy exhibits excellent electrochemical performance on symmetric batteries and all-solid-state *** symmetrical cell exhibits stable cycling performance over long duration over 500 h at 1.0 mA cm^(-2).Moreover,the LiNbO_(3)@NCM712/Li_(5.5)PS_(4.5)Cl_(1.5)/Li-10%Zn F_(2)battery exhibits excellent cycle stability at a high rate of 0.5 C,with a capacity retention rate of 76.4%after 350 cycles.
