Synthesis and evaluation of novel analogues of mangiferin as potent antipyretic

Objective:To screen different analogues of mangiferin pharmacologically for antipyretic activity. Methods:The naturally occurring xanthone glycoside mangiferin was isolated by column chromatography from the elhanolic extract of stem bark of Mangifera *** was further converted to 5-(N-phenylamino methyleno) mangiferin.5-(N-p-chlorophenylamino methyleno) mangiferin,5-(N-2-methyl phenylamino methyleno) mangiferin.5-(N-p-methoxy phenylamino methyleno) mangiferin.5-(N,N-diphenylamino methyleno) mangiferin,5-(N-α-napthylamino methyleno) mangiferin and 5-(N-4-methyl phenylamino methyleno) mangiferin *** synthesized compounds were further screened for antipyretic activity along with mangiferin at a dose level of 100 and 200 tng/*** and its analogues were characterized by melting point and R_f value determination and through spectral technique like UV,IR,and NMR spectral ***:The antipyretic activity of mangiferin as well as all analogues was found to be more significant in at higher dose ie.200 mg/kg which was depicted thmugh a decrease in rectal temperature up to 3 ***:The antipyretic activity of mangiferin and its analogues may be attributed to inhibition in synthesis of TNF-αand anti-oxidant activity associated with amelioration of inflammatory actions of cytokines.

Objective: To investigate the therapeutic efficacy of andrographolide, a plant derived compound, against chikungunya virus(CHIKV) infection. Methods: Using flow cytometry and immunoblotting assay, in vitro viral protein expression was studied in THP-1 cells line. In Balb/c mouse neonates, viral RNA copy number was determined by real time PCR. Results:The results showed reduced CHIKV protein expression on andrographolide treatment in CHIKV-infected human peripheral blood mononuclear cells, Vero cells and THP-I cell *** vivo, andrographolide treatment to CHIKV-infected neonates reduced viral RNA copy number. Further. andrographolide also increased cytotoxic T lymphocytes both in vitro and in vivo. Andrographolide also activated host innate immune pathways, viz., protein kinase *** eukaryotic initiation factor 2 α, retinoic acid inducible gene-Ⅰ and interferon regulatory factor 3/7, thereby increasing IFN-a secretion. CHIKV-induced nuclear factor κlight chain enhancer of activated B cells and tumor necrosis factor-a was also reduced on andrographolide treatment. Conclusion: Andrographolide inhibits CHIKV by suppressing viral protein expression and up-regulating host innate immunity and hence could be an effective therapeutic agent against CHIKV infection.

Mesenchymal stem cells(MsCs)play diverse roles ranging from regeneration and wound healing to immune *** investigations have indicated the crucial role of these multipotent stem cells in regulating various aspects of the immune *** express unique signaling molecules and secrete various soluble factors that play critical roles in modulating and shaping immune responses,and in some other cases,MSCs can also exert direct antimicrobial effects,thereby helping with the eradication of invading ***,it has been demonstrated that MSCs are recruited at the periphery of the granuloma containing Mycobacterium tuberculosis and exert"Janus"-like functions by harboring pathogens and mediating host protective immune *** leads to the establishment of a dynamic balance between the host and the *** function through various immunomodulatory factors such as nitric oxide(NO),IDO,and immunosuppressive ***,our group has shown that *** uses MSCs as a niche to evade host protective immune surveillance mechanisms and establish *** also express a large number of ABC efflux pumps;therefore,dormant *** residing in MSCs are exposed to a suboptimal dose of ***,it is highly likely that drug resistance is coupled with dormancy and originates within *** this review,we discussed various immunomodulatory properties of MsCs,their interactions with important immune cells,and soluble *** also discussed the possible roles of MsCs in the outcome of multiple infections and in shaping the immune system,which may provide insight into therapeutic approaches using these cells in different infection models.

The inter-neuronal communication occurring in extensively branched neuronal cells is achieved primarily through the microtubule(MT)-mediated axonal transport *** mechanistically regulated system delivers cargos(proteins,mRNAs and organelles such as mitochondria)back and forth from the soma to the *** proteins like kinesins and dynein mechanistically regulate polarized anterograde(from the soma to the synapse)and retrograde(from the synapse to the soma)commute of the cargos,*** axonal transport of such cargos is achieved by altering the microtubule stability via post-translational modifications(PTMs)ofα-andβ-tubulin heterodimers,core components constructing the *** within the lumen of MTs,K40 acetylation ofα-tubulin viaα-tubulin acetyl transferase and its subsequent deacetylation by HDAC6 and SIRT2 are widely scrutinized PTMs that make the MTs highly flexible,which in turn promotes their *** movement of various motor proteins,including kinesin-1(responsible for axonal mitochondrial commute),is enhanced by this PTM,and dyshomeostasis of neuronal MT acetylation has been observed in a variety of neurodegenerative conditions,including Alzheimer’s disease and Parkinson’s disease(PD).PD is the second most common neurodegenerative condition and is closely associated with impaired MT dynamics and deregulated tubulin acetylation *** the relationship between status of MT acetylation and progression of PD pathogenesis has become a chicken-and-egg question,our review aims to provide insights into the MT-mediated axonal commute of mitochondria and dyshomeostasis of MT acetylation in *** enzymatic regulators of MT acetylation along with their synthetic modulators have also been briefly *** towards a tubulin-based therapy that enhances MT acetylation could serve as a disease-modifying treatment in neurological conditions that lack it.

目的：描述曾行培养角膜缘上皮移植的患者再行穿透性角膜移植（PKP）的早期结果。方法：对因化学烧伤造成角膜缘干细胞缺乏而行培养的角膜缘上皮移植后又进行穿透性角膜移植术的患者有关人口统计学、原发病因、角膜缘移植类型、眼表稳定性、视力、移植片透明度和并发症方面的资料进行回顾和复习。对受体角膜片组织病理学特征进行研究，重点观察上皮状态。结果：在患有角膜缘干细胞缺乏而进行培养的角膜缘上皮移植术的125例患者中，15例接受了穿透性角膜移植术（PKP）。角膜缘上皮移植术后（自体移植组n=11，同种异体移植组n=4）平均间隔时间为7个月（范围2—12个月）。同种异体移植组的4例患者均接受了免疫抑制疗法。15只眼中14只眼（93％）获得了含有稳定的角膜上皮的健全的角膜植片。15只眼中14只眼术前最佳矫正视力低于20／200。PKP后平均随访8．3±5．0个月，8只眼最佳矫正视力高于20／60，5只眼最佳矫正视力20／200至20／60。2只眼低于20／200。15只眼中3只眼发生异体角膜植片的排斥反应但被成功治疗。1只眼发生植片排斥的同时发生了青光眼。同种异体角膜缘上皮移植无一例显示排斥表现。结论：培养的角膜缘上皮移植后，在形成稳定的眼表后行PKP的早期结果是良好的。为了避免排斥反应，培养的同种异体角膜缘上皮移植者行充分的免疫抑制治疗是必需的。角膜同种异体移植能够单独排斥同种异体角膜缘移植物。

In this article,we use the robust optimization approach(also called the worst-case approach)for findingε-efficient solutions of the robust multiobjective optimization problem defined as a robust(worst-case)counterpart for the considered nonsmooth multiobjective programming problem with the uncertainty in both the objective and constraint ***,we establish both necessary and sufficient optimality conditions for a feasible solution to be anε-efficient solution(an approximate efficient solution)of the considered robust multiobjective optimization *** also use a scalarizing method in proving these optimality conditions.

This article deals with some new chain imputation methods by using two auxiliary variables under missing completely at random(MCAR)*** proposed generalized classes of chain imputation methods are tested from the viewpoint of optimality in terms of *** proposed imputation methods can be considered as an efficient extension to the work of singh and Horn(Metrika 51:267-276,2000),singh and Deo(Stat Pap 44:555-579,2003),singh(Stat A J Theor Appl Stat 43(5):499-511,2009),Kadilar and Cingi(Commun Stat Theory Methods 37:2226-2236,2008)and Diana and Perri(Commun Stat Theory Methods 39:3245-3251,2010).The performance of the proposed chain imputation methods is investigated relative to the conventional chain-type imputation *** theoretical results are derived and comparative study is conducted and the results are found to be quite encouraging providing the improvement over the discussed work.

The present study deals with the co-ordination of cytokine （IL-4 and IFN-7） expression and kinetics of peste des petits ruminants （PPR） virus antigen and antibody in PPRV infected and vaccinated goats. The infected animals exhibited mixed cytokine （both TH1 and TH2） responses in the initial phase of the disease. The infected and dead goats had increased IFN-T response before their death; while IL-4 remained at the base level. The cytokine expression in recovered animals was almost similar to that of vaccinated ones, where a unique biphasic response of IL-4 expression was observed with an up-regulation of IFN-T on 7th days post vaccination （dpv）. Analysis of PPR virus antigen and antibody kinetics in different components of blood from infected and vaccinated animais revealed that the PPR virus antigen load was highest in plasma followed by serum and blood of the infected animals, whereas vaccinated animals showed only marginal positivity on 9th dpv. The antibody titer was high in serum followed by plasma and blood in both vaccinated and infected animals. Therefore, it is inferred that the presence of antigen and antibody were significant with the expression of cytokine, and that a decreased response of IL-4 was noticed during intermediate phase of the disease i.e., 7 to 12m days post infection （dpi）. This indicates the ability to mount a functional TH2 response after 14th dpi could be a critical determinant in deciding the survival of the PPR infected animal.
