Ultra-conservative noncoding RnA uc.243 confers chemo-resistance by facilitating the efflux of the chemotherapeutic drug in ovarian cancer

Background:Despite improvements in objective response rates to cisplatin-based combination chemotherapy,the majority of advanced ovarian cancer remains suboptimal,resulting in poor *** has been found that non-coding RnAs(ncRnAs)not only participate in the transmission of signals between various cells but also participate in tumor immunity and anti-tumor immune responses,thereby regulating tumor occurrence and ***,the function and detailed mechanism of ultraconserved RnA(ucRnA)in ovarian cancer chemoresistance is still ***:Western blotting assay,Quantitative real-time PCR analysis(qPCR),and Kapl.n-Meier Plotter analysis were performed to analyze the expression and prognosis of uc.243 in ovarian *** assay and Annexin V assay were performed to analyze the function of uc.243 in cisplatin resistance in ovarian cancer *** pull-down and qPCR experiments were performed to explore the molecular mechanism of uc.243 enhancing cisplatin resistance in ovarian cancer ***:Herein,we found that uc.243 was remarkably upregulated and correlated with patient survival in chemoresistance ovarian cancer patients compared with chemo-sensitive ovarian *** experiment displayed that uc.243 induced cisplatin resistance on ovarian cancer cells by facilitating the efflux of cisplatin(CDDP);but inhibiting the expression of uc.243 significantly reverses this ***,uc.243 can inhibit the binding of RnA binding protein DGCR8 microprocessor complex subunit to pri-miR-155,thereby inhibiting the cleavage of pri-miR-155 and decrease in mature miR-155,subsequently upregulates the expression of ATP binding cassette subfamily B member(ABCB1,ABCC2).Conclusion:Our research findings indicate that uc.243 can induce chemotherapy resistance in ovarian cancer,suggesting that it may become a new prognostic biomarker for malignant ovarian cancer.

Influence of cerium and yttrium addition on strength and electrical conductivity of pure aluminum alloys

To develop pure aluminum alloys with high conductivity and strength, Al-0.2Ce and Al-0.2Ce-0.1Y alloys were prepared by roll.ng and anneal.ng processes in this study. The effects of trace rare earth elements on the strength and electrical conductivity of the pure aluminum alloys were investigated. It is revealed that the addition of Ce and Y to pure aluminum can effectively enhance the strength and electrical conductivity of the alloys. In Al-0.2Ce, the addition of Ce can effectively refine the grain size of a-Al, with an average grain size of 90.68 μm in the as-cast state. The grain size of the alloy is further reduced to 87.55 μm by the simultaneous addition of Y. The synergistic addition of Ce and Y changes the properties of the alloy. The addition of Ce and Y also produces the Al_(11)Ce_(3) and Al_(3)Y second phases, which have coherent relationship with a-Al. The two-dimensional mismatch degree was calculated to be only 4.43%and 0.85%, respectively, which can provide a certain amount of nucleation substrate for a-Al in the incubation stage. The interfacial match between the l1_(2)structure of Al_(3)Y and a-Al was calculated using first-principles simulations. The results indicate that Al_(3)Y has a strong bonding effect with a-Al. nanoscale second phases at grain boundaries can be effective in reducing resistivity due to dislocation *** second phases with better matching interfaces to the substrate have no positive effect on the scattering motion of electrons.

Unlocking the Potential of Poly(butyl.ne succinate)through Incorporation of Vitrimeric network Based on Dynamic Imine Bonds

Poly(butyl.ne succinate)(PBS)exhibits many advantages,such as renewability,biodegradability,and impressive thermal and mechanical properties,but is limited by the low melt viscosity and strength resulted from the l.near *** address this,vitrimeric network was introduced to synthesize PBS vitrimers(PBSVs)based on dynamic imine bonds through melt polymerization of hydroxyl-terminated PBS with vanill.n derived imine containing compound and hexamethyl.ne diisocyanate using trimethylolpropane as a crossl.nking *** with different crossl.nking degrees were synthesized through changing the content of the crossl.nking *** effect of crossl.nking degree on the thermal,theological,mechanical properties,and stress relaxation behavior of the PBSVs was studied in *** results demonstrated that the melt viscosity,melt strength,and heat resistance were enhanced substantially without obvious depression in crystallizability,thermal stability,and mechanical properties through increasing crossl.nking *** addition,the PBSVs exhibit thermal reprocessability with mechanical properties recovered by more than 90%even after processing for three ***,PBSV with improved melt properties shows significantly improved foamability compared to commercial *** research contributes to the advancement of polymer technology by successfully developing PBS vitrimers with improved properties,showcasing their potential applications in sustainable and biodegradable materials.

Double-nyl.n purse-string suture in closing postoperative wounds following endoscopic resection of large(≥3 cm)gastric submucosal tumors

BACKGROUnD Endoscopic full-thickness resection(EFTR)of gastric submucosal tumors(SMTs)is safe and effective;however,postoperative wound management is equally *** on suturing following EFTR for large(≥3 cm)SMTs is scarce and *** To evaluate the efficacy and cl.nical value of double-nyl.n purse-string suture in closing postoperative wounds following EFTR of large(≥3 cm)*** We retrospectively analyzed the data of 85 patients with gastric SMTs in the fundus of the stomach or in the lesser curvature of the gastric body whose wounds were treated with double-nyl.n purse-string sutures after successful tumor resection at the Endoscopy Center of Renmin Hospital of Wuhan *** operative,postoperative,and follow-up conditions of the patients were *** All tumors were completely resected using EFTR.36(42.35%)patients had tumors located in the fundus of the stomach,and 49(57.65%)had tumors located in the body of the *** patients underwent suturing with double-nyl.n sutures after EFTR without laparoscopic assistance or further surgical *** fever and stomach pain were reported in 13(15.29%)and 14(16.47%)patients,*** serious adverse events occurred during the intraoperative or postoperative periods.A postoperative review of all patients revealed no residual or recurrent *** Double-nyl.n purse-string sutures can be used to successfully close wounds that cannot be completely closed with a single nyl.n suture,especially for large(≥3 cm)EFTR wounds in SMTs.

DnA-delivered monocl.nal antibodies targeting the p53 R175H mutant epitope inhibit tumor development in mice

Pl.nt-based meat analogues aggravated lipid accumulation by regulating lipid metabolism homeostasis in mice

To determine the effects of pl.nt-based meat analogues on the metabolic health and the possible mechanisms,mice were fed with a real pork diet(AP),a real beef diet(AB),a pl.nt-based pork analogue diet(PP)and pl.nt-based beef analogue diet(PB)for 68 *** with real meat,the pl.nt-based meat analogues increased food and energy intake,body weight,white fat and liver weight and caused adipocyte hypertrophy,hepatic lipid droplet accumulation,and inflammatory responses in *** revealed that pl.ntbased meat analogues altered the composition of serum metabolites,which regulated lipid metabolism *** PB diet upregulated gene expression related to lipid synthesis,lipolysis and adipocyte differentiation while the PP diet upregulated expression of lipolysis-related genes but downregulated expression of adipocyte differentiation-related genes in white adipose ***,both PP and PB diets upregulated lipid influx-and synthesis-related genes but downregulated lipid oxidation-related genes in *** specific metabolite biomarkers may affect fat accumulation mainly by direct lipid metabolism pathways or indirect amino acid metabolism,protein digestion and absorption,bile secretion,aminoacyl-tRnA biosynthesis,neuroactive ligand-receptor interaction and ABC transporters *** findings provide a new insight into understanding the differences in nutritional functions of meat and pl.nt-based meat analogues.
