Objective:To evaluate the effectiveness of the use of a modified Zhukovsky double(vaginal and uterine)balloon to improve the results of treatment in women with obstetric ***:We conducted an observational controlled study including 701 puerperas,which were divided into two groups:The main group consisted of 508 women,who underwent a combined management,that is,traditional(transfusion of fresh frozen plasma,erythrocyte mass,uterotonics,hemostatic agents),surgical hemostasis in cases of caesarean section,and insertion of a double Zhukovsky balloon;while the comparison group included 193 patients,who were managed *** main group and the comparison group were divided into subgroups according to the main etiology of obstetric hemorrhage:ⅠA andⅠB-postpartum;ⅡA andⅡB-placenta accreta;ⅢA andⅢB-postpartum ***:The most frequent causes of massive obstetric hemorrhage were atony of the uterus(39.2%),placenta previa(29.1%),and placental abruption(11.5%).Of the 508 balloon insertions,345(70.7%)were inserted for hemorrhage during cesarean section and 148(29.3%)for obstetric hemorrhage after spontaneous *** the obstetric hemorrhage at caesarean section,lower segment bleeding prevailed(78.1%),caused mainly by placenta previa,placenta accreta,placental abruption,polyhydramnios,and uterine scar *** management,including surgical hemostasis and insertion of vaginal and uterine balloon of Zhukovsky,reduced blood loss by 1.5 times and the number of hysterectomies by 6.72 times compared to ***:The use of a modified Zhukovsky double balloon in the management of obstetric hemorrhage may reduce the number of hysterectomies and the amount of blood loss.

Atomic force microscopy (AFM) was used for the morphological characterization and precise height meas-urements of two-dimensional molecular layers of carbocyanine dye 3,3’-di(r-sulfopropyl)-4,4’,5,5’-dibenzo-9-ethylthiacarbocyanine betaine pyridinium salt. The AFM measurements reveal three morphological types of molecular aggregates: leaves, stripes and spots. The leaves are stripes have same monolayer height ~1.4 nm and different crystal shapes: the leaves are monoloyers with the lens shape and the stripes are bilay-ers with the shape of extended rectangles. The monolayer height ~1.4 nm was interpreted as indicating the symmetrical packing arrangement of dye molecules. In the symmetrical monolayer, the sulfopropyl groups of all-trans monomer units are located on both monolayer sides whereas the adjacent stacked dye molecules have a lateral slippage providing the J-aggregate optical properties. The lower height of spots ~1 nm was explained by the model of an asymmetric monolayer with sulfopropyl groups of all-trans monomers occupy-ing the same position with respect to the monolayer plane. The packing arrangement of all-trans monomers in the asymmetric monolayer corresponds to H-aggregate. The alternative models of the packing arrange-ment in monolayers with mono-cis1 monomer configuration are discussed.

The basic methods of concentration and purification of liquid radioactive waste on the nuclear power plant are distillation and ionic exchange. During vaporization of oil waste products and the fulfilled washing solutions the part of oil passes into a condensate. Clearing of such condensate on ion-exchanged filters results to oiling of ion-exchanged materials and to decrease number of filter cycles. Due to the often regeneration of ion-exchanged filters additional volumes of waste products as the fulfilled reclaiming solutions, washing and loosening waters are formed. The attention of scientists was involved with methods of clearing of water environments from the organic substances, based on deep oxidizing transformations of hydrocarbons into carbonic gas and water. From processes of oxidation of hydrocarbons up to CO2 and H2O, sold at moderate conditions, our attention has involved photochemical oxidation with the help of Uv-radiation.

Photodynamic therapy(PDT)is one of the most appealing photonic modalities for cancer treatment based on anticancer activity of light-induced photosensitizer-mediated reactive oxygen species(ROS),but a limited depth of light penetration into tissues does not make possible the treatment of deep-seated neoplasms and thus complicates its widespread clinical ***,we introduce the concept of genetically encoded bioluminescence resonance energy transfer(BRET)-activated PDT,which combines an internal light source and a photosensitizer(PS)in a single-genetic construct,which can be delivered to tumors seated at virtually unlimited depth and then triggered by the injection of a substrate to initiate their *** illustrate the concept,we engineered genetic NanoLuc-miniSOG BRET pair,combining NanoLuc luciferase flashlight and phototoxic flavoprotein miniSOG,which generates ROS under luciferase-substrate *** prove the concept feasibility in mice bearing NanoLuc-miniSOG expressing tumor,followed by its elimination under the luciferase-substrate ***,we demonstrate a targeted delivery of NanoLuc-miniSOG gene,via tumor-specific lentiviral particles,into a tumor,followed by its successful elimination,with tumor-growth inhibition(TGI)coefficient exceeding 67%,which confirms a great therapeutic potential of the proposed *** conclusion,this study provides proof-of-concept for deep-tissue“photodynamic”therapy without external light source that can be considered as an alternative for traditional PDT.

The empirical (biochemical, the PCR method) and computational (quantum-chemical, PM3) methods have shown the ability of polysaccharides to catalyze the polymerization of amino acids and nucleotides within the physiologically acceptable temperature range. The possibility of nucleotide aminoacylation in the presence of polysaccharides has been established. The suggestion has been made that abiogenic aminoacylation of nucleotides by polysaccharides served as the prototype of the original aminoacylpre-tRNA-synthetase activity and subsequently determined the formation of the modern mechanism of genetic information transfer via three biopolymer types—nucleic acids, proteins and polysaccharides.

Quantum-chemical calculations and in vitro experiments have demonstrated that polysaccharides are able to adsorb mononucleotides due to the formation of hydrogen bonds. The subsequent dehydration of polysaccharide-nucleotide complexes in the physiologically acceptable temperature range results in the creation of “correct” DNA polymers that are “recognized” by the specific DNA-polymerases. DNA fragments abiogenically formed on polysaccharides vary in size and are characterized by a relatively “simple”, most probably, tandem structure. This research developed our previous concept of template-based polysaccharide synthesis with the participation of DNA tandem repeats (glycotranscription concept) making it possible to assume the existence of reverse glycotranscription in biological systems. Future research in the direction that we outlined experimentally may lead to a revolutionary approach in modern molecular biology—glycogenomics.

Increasing evidence points to a role for killer immunoglobulin-like receptors(KIRs)in the development of autoimmune *** particular,a positive association of KIR3DS1(activating receptor)and a negative association of KIR3DL1(inhibitory receptor)alleles with ankylosing spondylitis(AS)have been reported by several ***,none of the studies analyzed these associations in the context of functionality of polymorphic *** better understand how the KIR3DL1/3DS1 genes determine susceptibility to AS,we analyzed the frequencies of alleles and genotypes encoding functional(KIR3DL1*F)and non-functional(KIR3DL1*004)*** genotyped 83 AS patients and 107 human leukocyte antigen(HLA)-B27-positive healthy controls from the Russian Caucasian population using a two-stage sequence-specific primer PCR,which distinguishes KIR3DS1,KIR3DL1*F and KIR3DL1*004 *** the patients carrying two functional KIR3DL1 alleles,those alleles were additionally genotyped to identify KIR3DL1*005 and KIR3DL1*007 alleles,which are functional but are expressed at low ***3DL1 was negatively associated with AS at the expense of KIR3DL1*F but not of KIR3DL1**** finding indicates that the inhibitory KIR3DL1 receptor protects against the development of AS and is not simply a passive counterpart of the segregating KIR3DS1 allele encoding the activating ***,analysis of genotype frequencies indicates that the presence of KIR3DS1 is a more important factor for AS susceptibility than the absence of KIR3DL1**** activation of either natural killer(NK)or T cells via the KIR3DS1 receptor can be one of the critical events in AS development,while the presence of the functional KIR3DL1 receptor has a protective ***,even individuals with a genotype that carried two inhibitory KIR3DL1 alleles expressed at high levels could develop AS.

Polymorphisms in the SNCA Gene: Association with the Risk of Development of the Sporadic Form of Parkinson’s Disease and the Level of SNCA Gene Expression in Peripheral Blood of Patients from Russia

Parkinson’s disease (PD) is one of the most common human neurodegenerative disorders caused by the loss of dopaminergic neurons in the brain. The α-synuclein (SNCA) gene is one of the most studied genes involved in the pathogenesis of PD. In our study, we conducted a genetic analysis of promoter and intron single-nucleotide polymorphisms (SNPs) in the SNCA gene. We also analyzed the association of genotypes of these SNPs with expression levels of SNCA mRNA. One of four SNPs in the SNCA gene, and the rs2736990 polymorphism, associates with the risk of the sporadic form of PD in Russian population. The risk of PD was increased almost twofold in carriers of allele C (odds ratios = 1.9, 95% confidence interval: 1.2-2.91, p = 0.003). However, no association was found between any of the genotypes of SNPs tested (rs2583988, rs2619363, rs2619364 and rs2736990) and alterations in SNCA levels. Our findings support the hypothesis that the rs2736990 polymorphism is associated with PD. SNPs rs2583988, rs2619363 and rs2619364 in the promoter region of the SNCA gene themselves do not significantly influence the expression of SNCA. Most likely, SNCA gene expression is a very complex process that is affected by different genetic and epigenetic factors.
