Role of rice cytosolic hexokinase OsHXK7 in sugar signaling and metabolism

We characterized the function of the rice cytosolic hexokinase OsHXK7 （Qryza sativa HexokindseZ）, which is highly upregulated when seeds germinate under O2- deficient conditions. According to transient expression assays that used the promoter：luciferase fusion construct, OsHXK7 enhanced the glucose （Glc）-dependent repression of a rice n-amylase gene （RAmy3D） in the mesophyll protoplasts of maize, but its catalytically inactive mutant alleles did not. Consistently, the expression of OsHXK7, but not its catalytically inactive alleles, complemented the Arabidopsis glucose insensitive2-1 （gin2-1） mutant, thereby resulting in the wild type characteristics of Glc-dependent repression, seedling development, and plant growth. Interestingly, OsHXK7-mediated Glc-dependent repression was abolished in the O2-deficient mesophyll protoplasts of maize. This result provides compelling evidence that OsHXK7 functions in sugar signaling via a glycolysis-dependent manner under normal conditions, but its signaling role is suppressed when O2 is deficient. The germination of two null OsHXK7 mutants, oshxk7-1 and oshxk7-2, was affected by O2 deficiency, but overexpression enhanced germination in rice. This result suggests the distinct role that OsHXK7 plays in sugar metabolism and efficient germination by enforcing glycolysis-mediated fermentation in O2-deficient rice.

Multimodal treatments of right gastroepiploic arterial leiomyosarcoma with hepatic metastasis:A case report and review of the literature

Leiomyosarcoma of an artery is very rare, and cases with hepatic metastasis are even rarer. We describe a case of a 70-year-old man who after follow up due to rectal cancer, presented with an intra-abdominal hypervascular mass and a hepatic mass. After surgical resection, it was diagnosed as a leiomyosarcoma of the right gastroepiploic artery with hepatic metastasis. Multiple metastases had recurred at the liver. He has survived more than 53 mo through multimodal treatments(three surgical resections, radiofrequency ablation, transarterial chemoembolization, chemotherapies, and targeted therapy). Multimodal treatments, including active surgical resection, may behelpful in the treatment of aggressive diseases such as arterial leiomyosarcoma with metastasis.

AIM: To confirm the changes in proteins related with hypoxia-induced retinal cell death and to assess the effects of resveratrol(Res).METHODS: The therapeutic effect of Res was verified using an ischemic/reperfusion(I/R) model in vivo and a hypoxia modelin retinal ganglion cells(RGCs) in *** of RGCs were confirmed by TUNEL *** expression was confirmed by Western blotting and *** addition, flow cytometric analysis was used to confirm the response in the cell unit to obtain more accurate ***: ErbB2 expression and apoptosis in the ganglion cell layer(GCL) increased after I/R *** of Res rescued I/R-induced ganglion cell death, downregulated apoptosis and ErbB2 protein expression in the *** subsequent in vitro models, Res affects apoptosis by regulating the phosphorylation and expression of mouse double minute 2 homolog(MDM2), along with those of *** results suggest that Res reverses GCL-specific apoptosis via downregulation of ErbB2 in ischemic ***: In light of Res favorable properties, it should be evaluated in the treatment of RGC death and related retinal disease characterized by ErbB2 and MDM2 ***, Res is appropriate therapeutic agent for treating ischemic injury-related eye diseases by targeting the expression of ErbB2 and MDM2.

In this work,we have prepared the hierarchically nanostructured core–shell NiCo layered double hydroxide(NiCo-LDH)nanosheets-and ZnFe_(2)O_(4) nanocubes-decorated polyacrylonitrile(PAN)/pitch-based carbon nanofibers(PPCNs)webs(NiCo-LDH@PPCNs as cathode and ZnFe_(2)O_(4)@PPCNs as anode materials)with the bonded network structure by a facile and scalable hydrothemal ***,the low-cost pitch with lower softening point(~90℃)as co-precursor was utilized to produce the PAN/pitch-based carbon nanofibers(PPCNs)with enhanced electrical *** obtained PPCNs with pitch content of 30%(PP30CNs)electrode material delivered higher specific capacitance(~67 F g^(-1))than that(~48 F g^(-1))of the PAN-based carbon nanofibers(PCNs)at 1 A g^(-1),due to the increased electrical conductivity and lower interfacial charge transfer resistance(RCT)of~0.16 ***,the NiCo-LDH-decorated PP30CNs(NiCoLDH@PP30CNs)as cathode material showed superior specific capacitance of 1162 F g^(-1) at 1.0 A g^(-1) and ultra-high retention rate of 91.56%at 10 A g^(-1).The ZnFe_(2)O_(4)@PP30CNs as anode material also showed higher specific capacitance of 282 F g^(-1) at 1 A g^(-1) and good rate capability with capacitance retention of 56.73%at 10 A g^(-1).The as-fabricated asymmetric NiCo-LDH@PP30CNs//ZnFe_(2)O_(4)@PP30CNs hybrid supercapacitor device delivered a specific capacitance of~98 F g^(-1) at 1 A g^(-1) and excellent capacitance retention of~88%after 5000 charge–discharge cycles.

Comparison of anterior segment optical coherence tomography findings in acanthamoeba keratitis and herpetic epithelial keratitis

This study is to investigate the characteristic features of Acanthamoeba keratitis（AK） that differentiating it from herpetic epithelial keratitis（HEK） using anterior segment optical coherence tomography（AS-OCT）. Medical records of three eyes of each AK and herpetic keratitis who had AS-OCT examination were reviewed in this study. Slitlamp biomicroscopy and AS-OCT was performed on the initial visit and on every follow-up visits in all patients. In all three AK cases, reflective bands in the corneal stroma that correspond to the area of radial keratoneuritis were observed. The depth of the reflective bands varied in each case. After AK treatment, slit-lamp biomicroscopy confirmed that radial keratoneuritis had resolved and AS-OCT confirmed that reflective bands in the corneal stroma had also disappeared in all patients. Unlike the AS-OCT results found in AK, highly reflective HEK lesions were observed only in the subepithelial area, not in the stroma. AS-OCT seems to be helpful analyzing the specific depth of the lesion which enables to distinguish AK from HEK.

CT scan is regarded as the imaging modality of choice in patients with pancreaticobiliary ductal abnormalities. However, the axial orientation of the CT images provides only limited anatomical view of pancreaticobiliary ductal abnormalities. The technological advances of multi-de-tector row CT and three-dimensional image processing in workstations allows rapid image acquisition and a short postprocessing time. In particular, multiplanar reforma-tions (MPR) and minimum intensity projections (MinIP) offer rapid and accurate images of the anatomy and ab-normalities of the pancreaticobiliary tree. Moreover, MPR and MinIP help determine the relationship between the pancreaticobiliary ductal anatomy and the surrounding structures. This pictorial review illustrates the wide spec-trum of images obtained by the MPR and MinIP of the anomalies and disorders of the pancreaticobiliary tree.

Soil texture affects the conversion factor of electrical conductivity from 1:5 soil-water to saturated paste extracts

Electrical conductivity(EC)of soil-water extracts is commonly used to assess soil ***,its conversion to the EC of saturated soil paste extracts(ECe),the standard measure of soil salinity,is currently required for practical *** many regression models can be used to obtain ECe from the EC of soil-water extracts,the application of a site-specific model to different sites is not straightforward due to confounding soil factors such as soil *** study was conducted to develop a universal regression model to estimate a conversion factor(CF)for predicting EC_(e) from EC of soil-water extracts at a 1:5 ratio(EC_(1:5)),by employing a site-specific soil texture(i.e.,sand content).A regression model,CF=8.9105e^(0.0106sand)/1.2984(r^(2)=0.97,Pil surveys(n=173)and laboratory experiments using artificial saline soils with different textures(n=6,sand content=10%-65%)and salinity levels(n=7,salinity=1-24 dS m^(-1)).Model performance was validated using an independent dataset and demonstrated that EC_(e) prediction using the developed model is more suitable for highly saline soils than for low saline *** feasibility of the regression model should be tested at other *** soil factors affecting EC conversion factor also need to be explored to revise and improve the model through further studies.

Several image-based biomedical diagnoses require high-resolution imaging capabilities at large spatial ***,conventional microscopes exhibit an inherent trade-off between depth-of-field(DoF)and spatial resolution,and thus require objects to be refocused at each lateral location,which is time ***,we present a computational imaging platform,termed E2E-BPF microscope,which enables large-area,high-resolution imaging of large-scale objects without serial *** method involves a physics-incorporated,deep-learned design of binary phase filter(BPF)and jointly optimized deconvolution neural network,which altogether produces high-resolution,high-contrast images over extended depth *** demonstrate the method through numerical simulations and experiments with fluorescently labeled beads,cells and tissue section,and present high-resolution imaging capability over a 15.5-fold larger DoF than the conventional *** method provides highly effective and scalable strategy for DoF-extended optical imaging system,and is expected to find numerous applications in rapid image-based diagnosis,optical vision,and metrology.

Pancreatic adenocarcinosarcoma of monoclonal origin:A case report

Adenocarcinosarcoma, a neoplasm containing both carcinomatous and sarcomatous components, is a rare form of a cancer and the pathophysiology is currently poorly understood. Moreover, definitive treatment guidelines for this disease have not yet been established. Pancreatic adenocarcinosarcoma is even more rare and the prognosis is fatal. Here, we report a case of a 77-year-old male with pancreatic adenocarcinosarcoma and metastasis to the liver. The patient presented at our hospital with uncontrolled glucose levels and diabetes mellitus. The patient’s laboratory findings were unremarkable with the exception of elevated carbohydrate antigen 19-9 levels. Biopsies of the tumors in the pancreas and the liver revealed two types of tumors: pancreatic adenocarcinoma and a poorly differentiated sarcoma. To determine if KRAS mutations were present, we performed a peptide nucleic acid (PNA) clamp PCR-based assay. DNA sequencing by PNA clamp PCR identified a point mutation in codon 12 of exon 2 within KRAS from both tumor types. Because the KRAS mutation is observed in both tumor components, our findings support a monoclonal tumor origin followed by subsequent divergent differentiation into the sarcomatous and carcinomatous tumor populations. After we considered the patient’s status and the late stage of tumor detection, gemcitabine chemotherapy was administered.

BACKGROUND Combined tumors comprising large-cell neuroendocrine carcinoma and hepatocellular carcinoma have been rarely reported in the *** SUMMARY We report a case of a 73-year-old woman with chronic hepatitis B suspected to have a malignant hepatic mass(segment 3;size,4.5 cm)and lymph node metastasis based on computed tomography and magnetic resonance *** being Child-Pugh class A,esophageal varices were *** underwent left lateral sectionectomy and lymph node *** examination revealed a collision tumor consisting of large-cell neuroendocrine(90%)and hepatocellular(10%)*** combined carcinoma had metastasized to one of the three lymph nodes *** patient recovered without any postoperative complications and was discharged in good condition on postoperative day *** chemotherapy was not *** recurrence occurred during a follow-up period of 24 *** To improve the therapeutic management of combined tumors in the liver,it is necessary to discuss each clinical experience and consider an appropriate method for the preoperative diagnosis and treatment.
